Dictyostelium Myosin-Ie Is A Fast Molecular Motor Involved In Phagocytosis, Ulrike Durrwang, Setsuko Fujita-Becker, Muriel Erent, F. Jon Kull

Class I myosins are single-headed motor proteins, implicated in various motile processes including organelle translocation, ion-channel gating, and cytoskeleton reorganization. Here we describe the cellular localization of myosin-IE and its role in the phagocytic uptake of solid particles and cells. A complete analysis of the kinetic and motor properties of Dictyostelium discoideum myosin-IE was achieved by the use of motor domain constructs with artificial lever arms. Class I myosins belonging to subclass IC like myosin-IE are thought to be tuned for tension maintenance or stress sensing. In contrast to this prediction, our results show myosin-IE to be a fast motor. …

Association for Himalayan Policy Research: Through various scholarly activities, NSC aims to create a global network of scholars, professionals, and policy practitioners interested in the policy and development of Nepal and the Himalayan region. Eventually, NSC plans to create an Association for Himalayan Policy Research. Journal publications and Himalayan-focused conference at Wisconsin University are important activities contributing to this collaboration, and our initial attempt to create this network has been very successful. Many scholars from the US, Europe, Far East, Australia, and Nepal are joining this network.

Although Nepal is an extremely insignificant emitter of global carbon – emitting less than 0.1% of global emissions - it is burdened by a disproportionately high degree of vulnerability to the adverse effects of climate change. Nepal’s exposures include the possibilities of increased GLOF (Glacial Lake Outburst Floods) phenomenon, the melting mountains syndrome, and decrease in agricultural production, increase in vector borne diseases, loss of important bio-diversity assets and change in the hydrological cycles and resulting effects. The 1971 Convention on Wetlands of International Importance, especially for Waterfowl Conservation (hereafter referred to as Ramsar) is historically and currently important from several standpoints. It came into force in 1975 and is the oldest of the international conservation conventions and the only one that protects one general ecosystem type. Through the main text and various policy documents published over the past 30 years, Ramsar provides signatory nations with many general and specific guidelines for conserving and managing wetlands resources and their associated wildlife. Widespread use of environmentally persistent herbicides has led to increasing concern over their impact on aquatic ecosystems and nontarget organisms. The triazine herbicide atrazine (6-chloro-N-ethyl-N ¹1-[1-methylethyl]-1,3,5-triazine-2,4- diamine) remains one of the most heavily used pesticides worldwide, and was the second most commonly used conventional pesticide in the U.S. agricultural market in 2001. Atrazine is an S-triazine herbicide that targets the photosynthetic process to control broadleaf weeds. Due to the moderate solubility (33 mg/L at 22°C) and relative persistence of atrazine in water (Solomon et al. 1996), contamination of surface waters as a result of non-point source …

Adherens Junction-Associated Protein Distribution Differs In Smooth Muscle Tissue And Acutely Isolated Cells, Thomas J. Eddinger, Jessen D. Schiebout, Darl R. Swartz

This study was designed to examine how smooth muscle (SM) cell (SMC) isolation affects the distribution of some adherens junction (AJ) complex-associated proteins. Immunofluorescence procedures for identifying protein distribution were used on gastrointestinal and tracheal SM tissues and freshly isolated SMCs from dogs and rabbits. As confirmed by force measurements, relaxation, Ca²⁺ depletion, and cholinergic activation of SM tissues do not cause significant redistribution of the AJ-associated proteins vinculin, talin, or fibronectin away from the plasma membrane. Unlike SMCs in tissue, freshly isolated SMCs show a variable peripheral/ cytoplasmic vinculin and talin distribution that is not altered by activation. …

Recent discussions of contentious politics have focused on struggles in and over space and place. This article builds upon these concerns by using ethnographic, interview, and documentary data to analyze the spatial politics of street market vendors in Santiago, Chile. Drawing upon Lefebvre’s concepts of perceived, conceived, and lived space as well as ideas drawn from research on space and protest, I show how street market vendors build upon spatial routines, a sense of place, political alliances, and scale jumping in their self-defense strategies at the local, national, and international scales. Background

Little is known about the genome sequences of lepidopteran insects, although this group of insects has been studied extensively in the fields of endocrinology, development, immunity, and pathogen-host interactions. In addition, cell lines derived from Spodoptera frugiperda and other lepidopteran insects are routinely used for baculovirus foreign gene expression. This study reports the results of an expressed sequence tag (EST) sequencing project in cells from the lepidopteran insect S. frugiperda, the fall armyworm.

Results

We have constructed an EST database using two cDNA libraries from the S. frugiperda-derived cell line, SF-21. The database consists of 2,367 ESTs …

The cross section for bond breaking at the site of a dissociative temporary negative ion state through the dissociative electron attachment process can be considerably enhanced by the presence of a second longer-lived temporary negative ion state elsewhere in the molecule, even one quite remote from the first. In a series of chloroalkenes possessing both C–Cl and C=C bonds separated by various distances, we show that the cross sections are determined by the lifetime of the lower anion state created by the mixing of the anion states of these two moieties, with the wave function’s coefficients giving the probability that …

Few users of computer applications seek help from the documentation. This paper reports the results of an empirical study of why this is so and examines how, in real work, users solve their usability problems. Based on in-depth interviews with 25 subjects representing a varied cross-section of users, we find that users do avoid using both paper and online help systems. Few users have paper manuals for the most heavily used applications, but none complained about their lack. Online help is more likely to be consulted than paper manuals, but users are equally likely to report that they solve their …
