Inactivation by allelic exchange in clinical isolates of the emerging nosocomial pathogen Enterococcus faecium has been hindered by lack of efficient tools, and, in this study, transformation of clinical isolates was found to be particularly problematic. For this reason, a vector for allelic replacement (pTEX5500ts) was constructed that includes (i) the pWV01-based gram-positive repAts replication region, which is known to confer a high degree of temperature intolerance, (ii) Escherichia coli oriR from pUC18, (iii) two extended multiple-cloning sites located upstream and downstream of one of the marker genes for efficient cloning of flanking regions for double-crossover mutagenesis, (iv) transcriptional terminator …

In 1920 there were approximately 6.5 million farms in the United States. 30.1 percent of United States population lived on farms. In 1992 those numbers were down to 2 million farms and less than 2 percent of the population living on farms [Allen and Lueck, 1998, 344].
The farming industry of today bears little resemblance to that of yesterday. Corporate farming has challenged the age-old structure of farming. Proponents of corporate farming say that it is more efficient than family farming and leads to more affordable food supplies. This article considers the story of Terri Schiavo in the context of a broader set of ideological debates in the U.S. Primary among them is the debate about abortion. In the decades since Roe, pro-life advocates have refashioned the rhetoric of the abortion debate. During that period, an underlying concern with the meaning and scope of family relationships was largely displaced in public debate with concern about the ontological status of fetuses and embryos. Pro-life advocates thus talked less often about abortion as a desecration of traditional family life and woman's purpose and more about abortion as the murder of …

This study examined the effect of a visual arts, family-focused school readiness intervention on the parental efficacy and level of family involvement of low-income families. A second, primary focus of the study was to test the potential of academically focused experiences in the visual arts to increase the basic concept development of young children. To increase accessibility, the intervention was conducted in a neighborhood library.
The research design for this descriptive study included qualitative and quantitative components. Data collection consisted of a non-randomized one group pre-test/posttest design from students on a standardized instrument and from primary caregivers using self-report questionnaires. …

Television Viewing Habits, Body Mass Index, Dietary Behaviors And Physical Activity Among University Students, Mary Catherine Ball

This study examined the relationship between TV-viewing, BMI, dietary behaviors and physical activity in a purposive sample of university students. Twenty-three items from the National College Health Risk Behavior Survey were used. For data analysis, students were placed into dichotomous categories for TV-viewing risk; dichotomous categories were also created for each DV. Chi-square tests examined relationships between demographic and risk-behavior correlates and Ss' self-reports of TV-viewing; subsequent analyses were run for Ss who were normal- and overweight/obese. Odds ratios with 95% CI were also computed. TV-viewing was related to decreased use of diet/exercise to control weight and an increased likelihood …

The site-specific management of alfalfa has not been well-evaluated, despite the economic importance of this crop. The objectives of this work were to i) characterize the effects of soil moisture deficits on alfalfa and alfalfa yield components and ii) evaluate the use of canopy reflectance patterns in measuring treatment-induced differences in alfalfa yield. A randomized complete block design with five replicates of subsurface drip irrigation (SDI) and rainfed treatments of alfalfa was established at the University of Kentucky Animal Research Center in 2003. This research study was designed to examine and compare how community engagement knowledge of Program Administrators and Community Health Advisors (CHAs) can be shaped and affected by social capital and community capacity. This was achieved by analyzing the Congregations for Public Health’s Search Your Heart Program. The analysis used a three one-way fixed-effects ANOVA model, one for each of the three research questions. Overall, the results showed that upon examination of each of the three relationship hypotheses there were no primary differences in average knowledge among the groups. African Americans (AA) exhibit greater insulin responses following either an oral or intravenous glucose challenge relative to their European American (EA) counterparts. The etiology of this difference is not fully understood nor has it been confirmed with a more physiologically relevant mixed meal. Insulin secretion is regulated by a variety of factors. Significant among these are the enteric peptides glucagon-like peptide-1 (GLP-1) and glucose-dependent insulinotropic polypeptide (GIP). The role of the incretin system in the ethnic differences in post challenge insulinemia has received little attention. Hence, the purpose of the study was two-fold. Formulated feeds that support weight gain and survival in juvenile sea urchins are necessary for successful sea urchin aquaculture. In Chapter 1, juvenile Lytechinus variegatus were fed ad libitum one of four experimental feeds: 1) mixed-taxa algal biofilm (MTAB), 2) freeze-dried MTAB, 3) a commercial, small mammal feed (Friskies® cat treats), or 4) a commercial, formulated feed for sea urchins. The MTAB and sea urchin feed supported weight gain and survival, suggesting that juveniles as small as 3 to 4 mm can utilize formulated feeds for growth. The efficacy of Limburger cheese as an odour bait for sampling African malaria vectors was evaluated. Studies were done in Suba district, Western Kenya at Lwanda Nyamasare village and in a screen house set up at the Thomas Odhiambo campus of the International Center of Insect Physiology and Ecology (ICIPE). Preliminary investigations compared mosquitoes caught by a Counter Flow Geometry (CFG) trap baited with Limburger cheese, a standard centres for disease control (COC) light trap, an Entry trap and Man Landing Catches (MLC).
