This study is an informative discussion on the history behind Walt Disney's film, Song of the South, based on the Uncle Remus tales originally written by Joel Chandler Harris, which will be addressed in Chapter One. Chapter Two elaborates on the methods within the vehicles of two distinct media versions in two different time periods as the problematic source of perception. Chapter Three relates why this is due to cultural sensitivities pertaining to the film and the suggested media guidelines by minority organizations that have gained power since the Civil Rights movement. Spx is a global regulator discovered in Bacillus subtilis to suppress basal growth and development processes and activate transcription of genes involved in thiol homeostasis when a cell encounters oxidative stress. Its activity relies on reversible thiol-disulfide bond formation and binding RNA polymerase rather than DNA. The discovery that Staphylococcus aureus global virulence regulator, SarA, is more active upon cysteine reduction suggests that redox response could mediate virulence in this important human pathogen. We describe the cloning of spx from S. aureus strain RN6390, overexpression in Escherichia coli, and purification of native protein. Antibodies against Spx were raised for western …

In this thesis, a new synthetic methodology for the high yield synthesis of spineltype transition metal ferrite nanoparticles has been developed. This approach is based on the complexation of the first-row transition metal cations with diethylene glycol (DEG) followed by the hydrolysis of the resulting chelate iron alkoxide complexes in the presence of an alkaline hydroxide. Due to the passivation of their surfaces with DEG molecules, the as-prepared nanoparticles are stable against agglomeration and can be easily dispersed in polar protic solvents (water, alcohols, etc.). Gossypol and its methylated derivatives are produced in the leaves, seeds, stems, and roots of cotton plants. Although gossypol is toxic to many animals, other uses for gossypol are actively being investigated. To develop an experimental model for gossypol biosynthesis, a project to produce hairy root cultures from cotton was initiated. Hairy root cultures from two cotton species, Gossypium hirsutum (DPL 90) and Gossypium barbadense (Sea Island) were developed in this study. Gossypol was synthesized and retained by hairy root tissue at levels similar to that found in cottonseed. Cultures originating from a single transformation event were more similar in …

In this paper, the particle swarm optimization (PSO), originated as a simulation of a simplified social system with swarm intelligence and having exploring and exploiting characteristics of the particle swarm, is adopted to deal with the global optimization problems. To begin with, four non-linear two-dimensional functions are adopted as benchmark examples for determining the suitable ranges of parameters in the PSO. It is found that uses of the PSO can make the search quickly converge to the global optimum. Then, several benchmark functions below ten-dimensions are adopted especially for comparisons of search efficiency on the PSO and other algorithms. Results …

After years of relative neglect, the past few decades have witnessed a dramatic renewal of interest in the natural law tradition within philosophical circles. This natural law renaissance, however, has yet to bear much fruit within American constitutional discourse, especially among commentators on the left. In light of its low profile within contemporary constitutional debates, an effort to formulate a natural law constitutionalism is almost by definition an event worthy of sustained attention. Invasive species are having severe ecological (Mack et al. 2000) and economic (Pimentel et al. 2005) impacts on ecosystems around the world. Expression of selenocysteine (Sec)-containing proteins requires the presence of a cis-acting mRNA structure, called selenocysteine insertion sequence (SECIS) element. In bacteria, this structure is located in the coding region immediately downstream of the Sec-encoding UGA codon, whereas in eukaryotes a completely different SECIS element has evolved in the 3’-untranslated region. Here, we report that SECIS elements in the coding regions of selenoprotein mRNAs support Sec insertion in higher eukaryotes. Comprehensive computational analysis of all available viral genomes revealed a SECIS element within the ORF of a naturally occurring selenoprotein homolog of glutathione peroxidase 4 in fowlpox virus. Land use change results in significant amounts of soil migrating into aquatic streams during storm events. This suspended sediment, even in the absence of adsorbed contaminants, may be a significant stressor to aquatic organisms. The data on ecological effects of suspended sediment has mainly focused on salmonids or ecosystem level consequences such as habitat alteration. Few studies have quantified physiologic effects of suspended solids on water column organisms. The Lebanese families who arrived in South Carolina found themselves in a different environment than most had anticipated. Those who had spent time elsewhere in the U.S. found predominantly rural and predominantly Protestant South Carolina to be almost as alien as they or their parents had found the United States due partly to the religious differences and partly to the cultural differences between the Northeast, where most of them had lived for at least a few years after arriving in the United States, and the Southeast. Advancements in the field of biomaterials are being made to produce an artificial silk fiber. A gene was constructed which utilized components from both the dragline silk of Nephila clavipes and nematode collagen. For this engineered protein, the yeast strain Pichia pastoris was chosen to be the host organism. Previous research has shown P. pastoris to have comparatively low amounts of specific protein productivity. Therefore, this problem must be compensated for by obtaining extremely high cell densities. Flavobacterium columnare and Edwardsiella ictaluri are the leading causes for losses in the aquaculture industry. The losses from the diseases caused by these pathogens total $2.5 million for catfish and $100 million for total aquaculture. F. columnare's infection rates and stressors associated with outbreaks have been well-characterized, but the specific inflammatory responses of catfish neutrophils to the bacteria have not been studied. The elevated neutral-to-earth voltage (NEV), and the related phenomenon called stray voltage, is analyzed in multigrounded distribution systems. Elevated NEV is typically caused by fundamental frequency currents returning to the source via the neutral conductor and earth. However, harmonic distortion is also found to contribute to elevated NEV. A multiphase harmonic load flow algorithm is developed to examine the effects of various factors on the NEV, including unsymmetrical system configuration, load unbalance and harmonic injection. The purpose of this study was to develop and assess a potential key feature in a minimally-invasive breast reconstruction device and to collect information for an in vitro tissue test system. Tannic acid is a hydrolysable plant tannin, and it has been determined that tannic acid functions as a collagen crosslinking agent through both hydrogen bonding mechanisms and hydrophobic effects. Additionally, current research suggests that tannic acid may have anti-tumor properties.
