Objective: This study investigates whether 99mTc pyrophosphate (PYP) imaging provides a quantitative non-invasive assessment of the extent of electroporation injury, and of the effect of poloxamer in vivo on electroporated skeletal muscle. Methods: High-voltage electrical shock was used to produce electroporation injury in an anesthetized rat's hind limb. In each experiment, the injured limb was treated intravenously by either poloxamer-188, dextran, or saline, and subsequently imaged with 99mTc PYP. The radiotracer's temporal behavior among the experimental groups was compared using curve fitting of time-activity curves from the dynamic image data. Results: The washout kinetics of 99mTc PYP changed in proportion …

This report was commissioned by the USDA Forest Service Lake Tahoe Basin Management Unit to synthesize existing information on the ecology and management of aspen (Populus tremuloides) in the Sierra Nevada of California and surrounding environs. It summarizes available information on aspen throughout North America from published literature, internal government agency reports, and experienced scientists and managers. The historic distribution, abundance, and ecologic role of aspen in the Sierra Nevada are discussed, along with the reproductive physiology of aspen. Issues that affect aspen health and vigor in the Sierra Nevada and elsewhere are covered, along with methodology for …

Utilization Of Sea Scallop (Placopecten Magellanicus Gmelin) Microsatellite Markers For Phylogenetic Applications In Bay Scallops (Argopecten Irradians Lamarck), Lilliana E.J. Kim, James J. Campanella, Paul Ax Bologna

In previous research, microsatellite markers have been used to geographically and phylogenetically characterize and identify populations of sea scallops (Placopecten magellanicus Gmelin). Given that sea scallops are a closely related species to bay scallops (Argopecten irradians Lamarck), we have investigated whether the flanking DNA sequences of sea scallop microsatellite loci have been conserved in the bay scallop. We found that in all seven sets of published sea scallop PCR primers (Pma130, -132, -135, -180, -200, -212, and -275) that were tested with bay scallop DNA, there was sequence conservation and amplification of microsatellite regions. In response to ever-increasing anthropogenic changes to natural ecosystems, regional, national and international organizations have established guidelines for monitoring biological diversity. Most monitoring programs, however, do not take full advantage of the potential afforded by molecular genetic markers, which can provide information relevant to both ecological and evolutionary time frames, while costing less and being more sensitive and reliable than traditional monitoring approaches. As several molecular and computational approaches are relatively new, many technical and theoretical issues remain to be resolved. Here, we illustrate how DNA and population genetic data can provide valuable information, often unattainable via other approaches, for …

JECP stands for a java electron crystallography project that includes a series of practical java stand-alone programs for electron diffraction/microscopy and crystallography applications. The aim of the JECP project is twofold, i) as teaching tools to show students the principles of electron diffraction/microscopy and crystallography, ii) as research tools to analyze experimental results. Although there are commercial and public domain computer programs available that allow a user to simulate electron diffraction patterns or processing HREM images, there are always situations when we need to perform operations that are not a feature of any of the existing programs. The bacterial plant pathogen Pseudomonas syringae requires a type III protein secretion system (TTSS) to cause disease. The P. syringae TTSS is encoded by the hrp-hrc gene cluster. One of the genes within this cluster, hrpJ, encodes a protein with weak similarity to YopN, a type III secreted protein from the animal pathogenic Yersinia species. Here, we show that HrpJ is secreted in culture and translocated into plant cells by the P. syringae pv. tomato DC3000 TTSS. A DC3000 hrpJ mutant, UNL140, was greatly reduced in its ability to cause disease symptoms and multiply in Arabidopsis thaliana …

The DOE Consortium for Research on Enhancing Carbon Sequestration in Terrestrial Ecosystems (CSiTE) conducts multi-scale research to acquire basic knowledge for underpinning the implementation of soil carbon (C) sequestration in an environmentally acceptable and economically feasible manner. Research is based on the premise that identifying and understanding the basic mechanisms controlling sequestration across managed and unmanaged ecosystems are fundamental to developing approaches for enhancing C capture and long-term storage. Objective: To assess the effects of energy dilution with non-fermentable and fermentable fibers on abdominal fat and gut peptide YY (PYY) and glucagon-like peptide (GLP)-1 expressions, three rat studies were conducted to: determine the effects of energy dilution with a non-ferment-able fiber, compare similar fiber levels of fermentable and non-fermentable fibers, and compare similar metabolizable energy dilutions with fermentable and non-fermentable fibers. Research Methods and Procedures: In Study 1, rats were fed one of three diets with different metabolizable energy densities. The African witchweed (Striga spp.) and lepidopteran stemborers are two major biotic constraints to the efficient production of maize in sub-Saharan Africa. Previous studies had shown the value of intercropping maize with Desmodium uncinatum in the control of both pests. The current study was conducted to assess the potential role of other Desmodium spp., adapted to different agro-ecologies, in combined control of both pests in Kenya. Treatments consisted of intercropped plots of a Striga hermonthica- and stemborer-susceptible maize variety and one Desmodium sp. or cowpea, with a maize monocrop plot included as a control. In today's debate on digital rights management systems, there is a considerable divide between the rights holders, their investors and representatives on the one hand and academics, consumer advocates, and civil libertarians on the other. These two groups often talk past each other, concocting their own doomsday scenarios while arguing for laws and policies that vindicate their positions. Unfortunately, neither side has sufficient empirical evidence to either support its position or disprove its rivals'. As the digital economy grows, the debate intensifies, and the divide between the two sides widens. Today, there has emerged an urgent need to find the …
