Marginal soils, including loose sands, soft clays, and organics are not adequate materials for construction projects. These marginal soils do not possess valuable physical properties for construction applications. The current methods for remediation of these weak soils such as stone columns, vibro-compaction, etc. are typically expensive. Waste materials such as scrap tires, ash, and wastewater sludge, offer a cheaper method for stabilizing marginal soils. As an added benefit, utilizing waste materials in soil stabilization applications keeps these materials from being dumped into landfills, thereby saving already depleting landfill space. Atmospheric deposition of nitrogen species represents an additional nutrient source to natural environments, and can alter the nitrogen cycle by increasing nutrient levels beyond the requirements of organisms. In Tampa Bay, atmospheric deposition of dissolved inorganic nitrogen species (DIN) has been found to be the second largest nitrogen source, but little is known about dissolved organic nitrogen species (DON). The research goal was to improve the dry and wet deposition estimates by inclusion of the DON contribution. The hearing abilities of elasmobranch fishes were examined in response to several types of stimuli using auditory evoked potentials (AEP). Audiograms were acquired for the nurse shark, Ginglymostoma cirratum, the yellow stingray, Urobatis jamaicensis, in a controlled environment using a monopole underwater speaker. A dipole stimulus was used to measure the hearing thresholds of the horn shark, Heterodontus francisi, and the white-spotted bamboo shark, Chiloscyllium plagiosum. The dipole experiments yielded much lower thresholds than any other experiment, suggesting that this type of sound specifically stimulated the macula neglecta by creating a strong velocity flow above the head of the shark. …

Hematological malignancies, such as lymphomas and myelomas, have low cure rates or remain refractory to treatment, although advances have been made in treatment regimens for these patients. Questions still remain as to what is occurring in these cells on a molecular level, specifically at the level of gene transcription. The positive regulatory domain I binding factor 1 (PRDI-BF1) has been shown to directly repress genes required for cell proliferation and maintenance of the B cell phenotype, however very little is known as to its regulation. Analysis of the genomic sequences of Streptococcus mutans UA159 and Group B Streptococcus (GBS) strains Streptococcus agalactiae NEM316 and S. agalactiae 2603V/R indicated the presence of two putative collagenase genes in each organism. smcol1 from S. mutans was previously cloned and analyzed and the results indicated that the enzyme belonged to the U32 family of collagenases/peptidases. This enzyme shares homology with the prtC of Porphyromonas gingivalis, one of the principal examples of the U32 family of peptidases. Research has shown alcohol expectancies to be critically important in understanding maladaptive drinking patterns within alcohol use disorders. Alcohol expectancies, thought to be automatically elicited in the presence of environmental alcohol-related cues, represent both cognitive and affective associations with drinking behavior. However, the automatic and affective properties of alcohol expectancies have not yet been thoroughly measured in the literature. Psychophysiological measures, including skin conductance, heart rate, and the acoustic startle response in particular, offer a uniquely powerful set of indices for the automatic affective processing of alcohol-related cues.

This dissertation investigated the effects of multimedia annotation on L2 vocabulary learning and reading comprehension. The overarching objective of this study was to compare the effects of text-picture annotation and audio-picture annotation on L2 vocabulary immediate recall and reading comprehension. This study also sought to examine the different effects under incidental and intentional learning conditions. The participants were 78 intermediate adult ESL learners from three universities in northwest U.S. The participants read an Internet-based English text. Methicillin-resistant Staphylococcus aureus (MRSA) is among the most important pathogens affecting the human race in our time. In spite of recent medical advances, our therapeutic choices for MRSA infections remain limited due to the propensity of this organism to develop resistance to antimicrobials. Therefore, there is a continuing need to develop newer methods of treating MRSA infections. This dissertation examines the effects of bacteriophages 88 and 92 on ten clinical isolates of MRSA from the central Florida area. . The genus Chrysopsis (Asteraceae) contains eleven species native to Florida, including the newly described species, Chrysopsis delaneyi. Populations of this endemic plant species inhabit the Lake Wales Ridge (LWR) and the Atlantic Ridge (AR) of the Florida peninsula. Differences in morphology have been demonstrated within C. delaneyi, based on their locations. My objective was to determine the relationships between the LWR and the AR populations by analysis of chloroplast sequence and nuclear sequence variation. Approximately 160 samples of C. delaneyi and its sister species C. scabrella have been collected from fifteen sites throughout Florida. Six single base differences were detected, …

OBJECTIVE: The brain reserve hypothesis was examined in a secondary analysis of cross-sectional data from a community-based sample of 468 older adults residing in Charlotte County, Florida. The objective of the analysis was to determine the association between head circumference and eight cognitive outcomes and to assess any potential effect modification of existing associations by Apolipoprotein E (APOE) genotype. METHODS: Cognitive outcomes include scores from the Modified Mini-Mental State Exam (3MS), the Hopkins Verbal Learning Test-Revised (HVLT-R), Stroop Color-Word Test, Trail-Making Test A and B, and a word-stem completion task measuring implicit memory. This study investigated effects of plant quality and natural enemies on the abundance and survivorship of several leaf miner species on Florida scrub oaks over several ecological scales. Three oak species (Quercus laevis, Q. geminata, and Q. myrtifolia) and four leafminer species (Acrocercops albinatella, Brachys tesselatus, Stilbosis quadripustulatus, and Cameraria sp. nova) were the main focus of five separate studies, addressing effects of bottom-up and top-down factors at regional, local, and individual scales. Bottlenose dolphins (Tursiops truncatus) and other odontocete cetaceans rely on sound for communication, navigation, and foraging. Therefore, hearing is one of their primary sensory modalities. Both natural and anthropogenic noise in the marine environment could mask the ability of free-ranging dolphins to detect sounds, and chronic noise exposure could cause permanent hearing losses. In addition, several mass strandings of odontocete cetaceans, especially beaked whales, have been correlated with military exercises involving mid-frequency sonar, highlighting unknowns regarding hearing sensitivity in these animals.Auditory evoked potential (AEP) methods are attractive over traditional behavioral methods for measuring the hearing of marine mammals because they …

Tobacco smoking has been associated with the development, protraction, and exacerbation of chronically painful conditions. Conversely, there is reason to believe that smokers may be motivated to use tobacco as a means of coping with their pain. To date, no controlled, experimental studies have tested for a causal relationship between pain and smoking motivation. The primary aim of the current study was to test the hypothesis that laboratory-induced cold-pressor pain would enhance smoking motivation, as measured by self-reported urge to smoke and observation of immediate smoking behavior. A comparison of Elysia (=Tridachia) crispata (Mörch, 1863) from the Virgin Islands with elysiid slugs from the mangrove swamps and canals in the Florida Keys that have previously been identified as E. crispata reveals many differences in habitat, gross and microscopic anatomy, food preferences of juveniles, sources of symbiotic chloroplasts and their localization within the digestive tubules, radular morphology, and nucleic acid sequences of two genes. The differences between the two groups of slugs are such that the Florida Keys animals are considered to represent a new species, Elysia clarki. The principal isoform of Src homology (SH) 2-domain containing 5' inositol phosphatase protein 1 (SHIP) is a 145kDa protein primarily expressed by cells of the hematopoietic compartment. The enzymatic activity of SHIP is responsible for hydrolyzing the 5' phosphate of phosphatidylinositol-3,4,5-trisphosphate (PI(3,4,5)P3), and thereby preventing the recruitment of pleckstrin homology domain containing effector proteins. Furthermore, SHIP contains protein-protein interaction domains, such as an SH2 domain, two NPXY and several proline-rich motifs. All of these different domains endow SHIP with the capacity to impact signaling pathways important for proliferation, survival, differentiation and activation. Streptococcus pneumoniae is a major bacterial pathogen which causes pneumoniae, meningitis, otitis media, and bacteremia. Currently there are two vaccines available, Pneumococcal Polysaccharide Vaccine (PPV) for adults and the Pneumococcal Conjugate Vaccine (PCV) for children. The PCV vaccine was developed in 2000 specifically for children and infants due to the ineffectiveness of the PPV vaccine in children. The historic use of asbestos-containing materials during the manufacturing of automobiles has resulted in a perception of an increased risk of asbestos-related pulmonary diseases within mechanics. This study was conducted to assess the potential asbestos exposures mechanics encounter while servicing vehicles assembled with parts containing asbestos, in addition to compare the cumulative lifetime asbestos exposures for different maintenance activities against theorical threshold exposures for asbestosis, lung cancer and mesothelioma. The MicroImager is a fast, high resolution, real time, digital autoradiographic imaging tool with broad applications. This study utilizes the MicroImager to evaluate radiolabeled drug behavior in subcutaneous tissue. Experiments were conducted in conjunction with mathematical models to determine the diffusion coefficient (D) and elimination constant (k) for radiolabeled dexamethasone. Osmotic pumps containing [3H]dexamethasone were implanted into rat subcutaneous tissue over 6h, 24 h, and 60 h. Local tissue was explanted and slides were prepared for imaging. This work explores the relationship between nutrient loading and changes in water quality in a sub-tropical, above-ground, off-stream municipal water supply reservoir, the C.W. Bill Young Regional Reservoir. The three source waters for the reservoir have varied but high levels of nutrients such as phosphorus and nitrogen. In other reservoirs, these nutrients have been linked to deterioration of water quality and increased expense in water treatment. In 1977, the National Institute for Occupational Safety and Health (NIOSH) issued the publication: Criteria for a Recommended Standard....Occupational Exposure to Waste Anesthetic Gases and Vapors. The discovery of nitric oxide (NO) as the endothelial-derived relaxing factor has led to significant research on NO and the proteins involved in its function, generation, location and regulation. Synthesis of NO by blood vessel endothelial cells results from the enzymatic oxidation of arginine by endothelial nitric oxide synthase (eNOS) resulting in the formation of equimolar amounts of NO and citrulline. Citrulline is sequentially recycled to arginine by successive reactions involving the enzymes argininosuccinate synthase (AS) and argininosuccinate lyase (AL), respectively.

This research identified and evaluated possible uses of environmental monitoring data collected and reported by industrial facilities under the Clean Water Act requirements and determined whether the current regulatory system supported any of those uses. Federal policies and state-level policies in the United States, Florida, and California were evaluated in order to determine whether the current regulatory system supported any of the identified uses. West Nile virus (WNV) and St. Louis Encephalitis (SLEV) are arthropod-borne viruses belonging to the genus Flavivirus and are classified as significant human pathogens of global epidemiological importance. Since its introduction into the United States in 1999, WNV has spread throughout most of the country and has caused major epidemics of neuroinvasive disease (Hayes and Gubler, 2005). Aggregating proteoglycans (PG) bearing chondroitin sulfate (CS) side chains are well-known inhibitors of neural plasticity and associate with hyaluronan and tenascin-R to form a complex of extracellular matrix (ECM) in the central nervous system (CNS). Little is known about whether proteolytic cleavage of the core protein affects neural plasticity. Several members of the ADAMTS (a disintegrin and metalloproteinase with thrombospondin motifs) family of metalloproteinases are glutamyl-endopeptidases that cleave aggregating PGs. Our initial studies determined that neural cultures secrete a brevican-containing matrix, and that these neural cultures also produced ADAMTS4, a protease that cleaves brevican. Synoicum adareanum is a colonial tunicate commonly found on the benthos around Palmer Station on Anvers Island, Antarctica. A comprehensive chemical investigation of the lipophilic extract of the frozen tunicates gave a new series of polyketide macrolides, palmerolides A-E and H. The structure elucidation of these compounds was accomplished by extensive NMR and mass spectral studies.The palmerolides are unusual 20-membered macrolides displaying functionality more commonly found in sponges or cyanobacteria. In the U.S., the number of prostate biopsies increases annually. This is partly due to elevated prostate specific antigen (PSA) values identified during PC screening. This study's goal was improving prostate cancer (PC) detection through developing a clinical decision rule (CDR), based on an elevated PSA and laboratory biomarkers. This decision rule could be used after an elevated PSA, providing the patient and clinician information to consider prior to biopsy. The elasmobranch fishes possess a remarkable diversity of feeding mechanisms for a group containing relatively few species (~1200). The three most prevalent of these mechanisms involve prey capture during which the predator overtakes its prey (ram), prey is drawn into the mouth of the predator (suction), and relatively stationary consumption of sessile or substrate affixed prey (biting). Neurodegenerative diseases are associated with loss of neurons ultimately leading to a decline in brain function. Alzheimer's disease (AD) is considered one of the most common neurodegenerative disorders that affects 16 million people worldwide. The cause of the disease remains unknown, although significant evidence proposes the amyloid Beta-peptide (A-Beta) as a potential culprit. The binding of Cu2+ by the soluble fragments of A-Beta have shown to form Type-3 copper centers and catalyze the oxidation of catechol-containing neurotransmitters.
