Genetic Diversity Of Bean Pod Mottle Virus (Bpmv) And Development Of Bpmv As A Vector For Gene Expression In Soybean, Chunquan Zhang

University of Kentucky Doctoral Dissertations

Bean pod mottle virus (BPMV), a member of the genus Comovirus in the family Comoviridae, is widespread in the major soybean-growing areas in the United States. The complete nucleotide sequences of the genomic RNAs of the naturally occurring partial diploid strain IL-Cb1 were determined. Intermolecular RNA1 recombinants were isolated from strain IL-Cb1 and characterized at the molecular level. Structurally similar recombinant RNA1 was also generated after four passages in soybean derived from plants previously inoculated with a mixture of infectious RNA1 transcripts from two distinct strains. BPMV was developed as a plant viral vector that is appropriate for gene expression …

Buffalograss [Buchloe dactyloides (Nutt.) Engelm.] is a perennial, warm-season turfgrass that is native to the shortgrass prairie of North America. It offers potential benefits as a turfgrass because of its drought resistance, low incidence of pest problems, and relatively low nutrient requirements. This study was initiated to evaluate the level of organelle DNA diversity among buffalograss genotypes and to determine the mode of inheritance of the chloroplast organelle by cpDNA and mtDNA PCR-RFLPs. The 56 genotypes studied included diploids, triploid, tetraploids, pentaploids, and hexaploids. These genotypes represented 273 buffalograsses collected during 1994 and 1995 from diverse locations in the …

Risk And Ethics In Biological Control, Ernest S. Delfosse Jan 2005

Risk And Ethics In Biological Control, Ernest S. Delfosse

United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications

All introduced natural enemies present a degree of risk to nontarget species. Since most biological control programs use relatively host-specific natural enemies, the risk to nontarget species is generally very low, particularly from biological control of weeds, which uses extensively tested and validated host-specificity testing procedures to predict risk. However, many of the published comments about risks of biological control are superficial or misleading, often inappropriately lumping risk from all taxa of agents as “the risk of biological control,” and ignore the potential benefits, rather than dealing with species-by-species risk and benefits. Particularly confounding accurate predictions is the common mixing …

Synthesis And Characterization Of Nanocrystalline Hydroxyapatite Powder; And The Effects Of Oxide-Based Sintering Additives On Tricalcium Phosphate, Himesh A. Bhatt

Electronic Theses and Dissertations

Calcium phosphate (CP) materials have been used extensively for bone replacement and augmentation due to their similarity to the mineral component of bone. In addition to being non-toxic, they are biocompatible, not recognized as foreign materials in vivo, and most importantly, exhibit osteoconductive behavior, being able to help in bone formation during healing. CPs form an intimate physicochemical bond with the host tissue, termed osseointegration. However the main limitation of calcium phosphates is their inherent brittle nature and poor mechanical performance under complex stress states. As a result, these materials have been used clinically only in non-load-bearing applications, primarily as …

Role Of Matrix Metalloproteinase-2 In Therosclerosis And Abdominal Aortic Aneurysms In Apolipoprotein E Deficient Mice, Jing Huang

University of Kentucky Doctoral Dissertations

Matrix metalloproteinase-2 (MMP-2, gelatinase A, type IV collagenase) is a member of a family of zinc-dependent metalloendopeptidases that functions in the degradation of elastin, collagens, and other components of extracellular matrix (ECM). Both secretion and activation of MMP-2 are elevated in human atherosclerotic lesions and abdominal aortic aneurysms (AAA). In this dissertation project, we sought to test the hypothesis that MMP-2 plays a critical role in both atherosclerosis and AAA. We also sought to determine the detailed mechanism. We first examined the atherosclerosis and AngII-induced AAAs development in MMP-2-/- x apolipoprotein (apoE)-/- mice in vivo. It was surprising that MMP-2 …

Interactions Between Selenium And Polychlorinated Biphenyls (Pcbs), Divinia Nolasco Stemm Jan 2005

Interactions Between Selenium And Polychlorinated Biphenyls (Pcbs), Divinia Nolasco Stemm

University of Kentucky Doctoral Dissertations

This study investigated the interaction between polychlorinated biphenyls (PCBs) and selenium to explain the mechanism involved that could affect selenium metabolism and its anti-cancer property. PCBs congeners and mixtures were previously found to reduce hepatic Se and Se-dependent glutathione peroxidase activity. I hypothesized that certain PCB congeners affect selenium metabolism in the rat liver resulting in diminished antioxidant capacity of selenoproteins, which could alter the ability of Se to protect against PCBs induced tumor promotion. In the first study, the influence of 3,3,4,4-tetrachlorobiphenyl (PCB 77) on hepatic Se and glutathione peroxidase (GPx1) activity as well as cytochrome P450 1A1 induction …

Genetic Regulation Of Hematopoietic Stem Cell Numbers In Mice, Ying Liang Jan 2005

Genetic Regulation Of Hematopoietic Stem Cell Numbers In Mice, Ying Liang

University of Kentucky Doctoral Dissertations

Hematopoietic stem cells (HSCs) transplantations are widely used for the treatment of hematological and non-hematological disorders in clinic. Successful transplantation requires sufficient number and efficient homing of HSCs. Many studies have focused on developing an effective strategy to expand functional HSC population. Some regulatory molecules have been recently shown great promise for controlling the amplification of HSCs. In these dissertation studies, I first aim to identify gene(s) and their allelic variants contributing to strain-specific difference in HSC numbers between C57BL/6 (B6, low) and DBA/2 (D2, high) mice by using a classic forward genetic approach. Firstly, 3 quantitative trait loci (QTL) …
