The Structural And Functional Analysis Of A Voltage-Dependent Potassium Channel, Vanessa Ruta Jan 2005

The Structural And Functional Analysis Of A Voltage-Dependent Potassium Channel, Vanessa Ruta

Student Theses and Dissertations

Voltage-dependent ion channels are finely tuned to open and allow ion conduction in response to changes in the membrane voltage, a function that lies at the heart of nerve impulse generation and propagation. These channels contain transmembrane voltage-sensing domains that contain highly conserved cationic amino acids known as "gating charges." The gating charge residues move under the influence of the membrane's electric field, a voltage dependent conformational change that induces pore opening. In this thesis, I explore the structural basis of gating charge movement through crystallographic, biochemical and functional studies of KvAP, a voltage-dependent K+(Kv) channel from a …

Exploring The Early Events In The Hiv-1 Life Cycle: From Post-Entry Restriction To Nuclear Import, Tshaka J. Cunningham Jan 2005

Exploring The Early Events In The Hiv-1 Life Cycle: From Post-Entry Restriction To Nuclear Import, Tshaka J. Cunningham

Student Theses and Dissertations

Early events in the viral replication cycle of the human immunodeficiency virus (HIV) determine the virus' ability to sustain a persistent infection in susceptible host species. However, many of the molecular machinations through which the virus progresses during its earliest moments in the cell's interior remain enigmatic. Similar to other virus families, HIV virions are packed with a specific assortment of viral and cellular proteins that allow them to carry out their unique program for replication inside of target cells. One such protein that is present in incoming virions, HIV-1 integrase, is an enzyme that facilitates the fusion of the …

The Role Of The Glyoxylate Cycle In The Pathogenesis Of Mycobacterium Tuberculosis, Ernesto Javier Muñoz Elías Jan 2005

The Role Of The Glyoxylate Cycle In The Pathogenesis Of Mycobacterium Tuberculosis, Ernesto Javier Muñoz Elías

Student Theses and Dissertations

According to the World Health Organization a third of the world's population is infected with Mycobacterium tuberculosis. The unparalleled success of M. tuberculosis as a pathogen reflects the bacterium's extraordinary ability to persist in its host in spite of eliciting a robust immune response. Currently available treatment is inadequate and drug resistance is rapidly spreading. New antibiotics are desperately needed. The substrates and metabolic pathways utilized by pathogens during infection are largely unknown and represent an under-exploited area of investigation. Uniquely, evolution of the genus Mycobacterium has involved extensive duplication of fatty acid metabolism genes, including two homologs encoding prokaryotic- …

Myosin-Like Proteins In S. Cerevisiae: Multifunctional, Structural Components Of The Nuclear Envelope, Mario Niepel Jan 2005

Myosin-Like Proteins In S. Cerevisiae: Multifunctional, Structural Components Of The Nuclear Envelope, Mario Niepel

Student Theses and Dissertations

The nuclear envelope (NE) separates the genetic material from the rest of the cell, delimits and defines the nucleus, organizes the intranuclear architecture and serves as a regulator for multiple nuclear processes. In all eukaryotes, filamentous coiled-coil proteins are associated with the intranuclear surface of the NE and are integral to proper nuclear function. One such protein, called Tpr in vertebrates, attaches to the NPC and appears to form the nuclear basket structure, is conserved throughout all eukaryotes. The two yeast homologs of Tpr are termed Mlp1p and Mlp2p. The Mlp proteins also attach to the nuclear face of the …

Changes In Antioxidative Enzymes Of Young And Mature Leaves Of Tomato Seedlings Under Drought Stress, Serpi̇l Ünyayar, Fazi̇let Özlem Çeki̇ç

Turkish Journal of Biology

The effects of exogenous abscisic acid (ABA) on antioxidant enzymes of young and mature leaves of 4-week old Lycopersicon esculentum cv. Ailsa Craig and ABA-deficient mutant, notabilis, were investigated under drought stress. Although ABA induced increases in ascorbate peroxidase (APX), glutathione reductase (GR) and catalase (CAT) activities in drought-stressed young leaves of notabilis, CAT activities remained unchanged in both ABA-treated young and mature leaves of Ailsa Craig under drought. Superoxide dismutase (SOD) activity did not substantially increase in notabilis or Ailsa Craig in all treatments. APX activity significantly increased in ABA-, drought- and drought+ABA- treated young leaves of notabilis. These …

The Dangers Of Fighting Terrorism With Technocommunitarianism: Constitutional Protections Of Free Expression, Exploration, And Unmonitored Activity In Urban Spaces, Marc Jonathan Blitz Jan 2005

The Dangers Of Fighting Terrorism With Technocommunitarianism: Constitutional Protections Of Free Expression, Exploration, And Unmonitored Activity In Urban Spaces, Marc Jonathan Blitz

Fordham Urban Law Journal

Part I of this article examines how some commentators can plausibly argue that constitutional liberty and privacy protections do not protect the individual liberty and privacy that modern individuals have come to expect in many public spaces, particularly in urban environments. Constitutional liberalism, this section points out, makes this question a difficult one, because it is marked by scrupulous neutrality towards different visions of “the good life.” In other words, the constitutional order does not condemn those who choose a communitarian way of life and favor those who prefer individualism. Rather, it tolerates both of these (and other) preferences about …

A Model Of Abstract Cooperation In Games Of Uncertainty, Hillel Bavli Jan 2005

A Model Of Abstract Cooperation In Games Of Uncertainty, Hillel Bavli

Fordham Urban Law Journal

The idea that evolutionary processes natrually propel a state of affairs toward a higher, perhaps more complex or advanced, state of affairs is one that may extend to any context characterized by a dynamic time frame, including ogliopoly moelds of repeated Prisoner's Dilemma. The author argues that, contrary to the popular assertion that coordinated pricing necessarily requires voluntary coordination, oligopoly markets may evolve to a state of cooperation—one of collective profit maximization—absent a conscious state of coordination among the players, or even knowledge of such cooperation. While Professor Donald Turner proposes a theory of cooperation based on forward-looking consideration of …

From Poverty To Abuse And Back Again: The Failure Of The Legal And Social Services Communities To Protect Foster Children, Sharon Balmer Jan 2005

From Poverty To Abuse And Back Again: The Failure Of The Legal And Social Services Communities To Protect Foster Children, Sharon Balmer

Fordham Urban Law Journal

After decades of legislative reform, stories of foster care abuse still appear on the fron pages of our newspapers, and foster children who are injured while in protective care are turning to the courts to change the system. It is still relatively difficult for a child to prevail in an action against child protective workers and agencies. Opinions addressing children’s issues are few, and courts seem hesitant to expand causes of action. This Comment explores the current state of children’s legal remedies for injuries incurred as the result of a foster care placement. Part I describes the foster care system …

Biometric Technology: The Impact On Privacy, Darcie Sherman Jan 2005

Biometric Technology: The Impact On Privacy, Darcie Sherman

Comparative Research in Law & Political Economy

The introduction of biometric technology has resulted in a significant shift, which defies tradition and some of the very values that we cherish as a society. Additionally, there have been numerous recent developments, which have facilitated a fundamental global re-assessment of the safety and security needs of our communities. Other challenges, like the delivery of government-granted social services, have resulted in governments looking for ways to ensure entitlement prior to the provision of finite resources to individuals. As technology is increasing in sophistication, it is being deployed in novel and creative ways to meet some of these new demands. However, …

How Parents And Children Disappear In Our Courts - And Why It Need Not Ever Happen Again, James A. Cosby Jan 2005

How Parents And Children Disappear In Our Courts - And Why It Need Not Ever Happen Again, James A. Cosby

Cleveland State Law Review

Part One of this Article further examines those moral, factual and legal dynamics in the family that make these cases so difficult. Part Two summarizes the present state of the law, and demonstrates precisely where and why the current legal approach is falling short. I will show how the law specifically fails to adequately define the rights of parents, substantively as well as procedurally and, I will furthermore demonstrate how, in their current forms, the doctrines of parental autonomy and the best interests of the child are far too broad and too rigid for many cases involving the parent-child relationship. …

Rethinking America's Approach To Workplace Safety: A Model For Advancing Safety Issues In The Chemical Industry, Gwen Forte Jan 2005

Rethinking America's Approach To Workplace Safety: A Model For Advancing Safety Issues In The Chemical Industry, Gwen Forte

Cleveland State Law Review

In Part II of this note, I analyze the impact of tort litigation, workers' compensation, collective bargaining, and the Occupational Safety and Health Act on workplace safety. I begin by describing how each of these vehicles operated historically and then I provide a contemporary perspective. In this section, I also consider the advantages and disadvantages of using these approaches to prevent and compensate for injuries. In Part III, I propose an alternative approach to workplace safety: employee board representation. In this section, I analyze and critique various methods of employee board representation and ultimately recommend a form of representation in …

Electrochemical Detection Of Nanoscale Phase Separation In Binary Self-Assembled Monolayers, R. Carlisle Chambers, Christina E. Inman, James E. Hutchison

Faculty Publications - Department of Biological & Molecular Science

Developing methods to probe the nature and structure of nanoscale environments continues to be a challenge in nanoscience. We report a cyclic voltammetry investigation of internal, hydrogen-bond driven phase separation of amide-containing thiols and alkane thiols. Amide-containing thiols with a terminal ferrocene carboxylate functional group were investigated in two binary monolayers, one homogeneously mixed and the other phase separated. The electrochemical response of the ferrocene probe was used to monitor adsorbate coverage, environment and phase separation within each of these monolayers. The results demonstrate that the behavior of ferrocene containing monolayers can be used to probe nanoscale organization.

Preparing The Way For Reform In Higher Education: Drawing Upon The Resources Of The Community-At-Large, Judith A. Ramaley

Public Administration Faculty Publications and Presentations

Higher education is being asked to pay more attention to student learning and to contribute to the enhancement of the social and economic conditions of the community it serves. As a result, educational institutions will no longer be self-contained. Community members and organizations have become not only critical partners in framing the goals and intentions of the educational reform movement, but they also have assets that must be tapped by educational institutions that wish to implement change and respond to social needs.

Faculty Publications - Department of Biological & Molecular Science

The proopiomelanocortin (POMC) gene is expressed in the pituitary and arcuate neurons of the hypothalamus. POMC arcuate neurons play a central role in the control of energy homeostasis, and rare loss of function mutations in POMC cause obesity. Moreover, POMC is the prime candidate gene within a highly significant quantitative trait locus on chromosome 2 associated with obesity traits in several human populations. Here, we identify two phylogenetically conserved neuronal POMC enhancers designated nPE1 (600 bp) and nPE2 (150 bp) located approximately 10 to 12 kb upstream of mammalian POMC transcriptional units. We show that mouse or human genomic regions …

Aerodynamics Of The Hovering Hummingbird, Douglas R. Warrick, Bret W. Tobalske, Donald R. Powers Jan 2005

Aerodynamics Of The Hovering Hummingbird, Douglas R. Warrick, Bret W. Tobalske, Donald R. Powers

Faculty Publications - Department of Biological & Molecular Science

Despite profound musculoskeletal differences, hummingbirds (Trochilidae) are widely thought to employ aerodynamic mechanisms similar to those used by insects. The kinematic symmetry of the hummingbird upstroke and downstroke has led to the assumption that these halves of the wingbeat cycle contribute equally to weight support during hovering, as exhibited by insects of similar size. This assumption has been applied, either explicitly or implicitly, in widely used aerodynamic models, and in a variety of empirical tests. Here we provide measurements of the wake of hovering rufous hummingbirds (Selasphorus rufus) obtained with digital particle image velocimetry that show force asymmetry: hummingbirds produce …
