Reconstitution Of A Minimal Dna Replicase From Pseudomonas Aeruginosa And Stimulation By Non-Cognate Auxiliary Factors, Thale C. Jarvis, Amber A. Beaudry, James M. Bullard, Nebojsa Janjic, Charles S. Mchenry

DNA polymerase III holoenzyme is responsible for chromosomal replication in bacteria. The components and functions of Escherichia coli DNA polymerase III holoenzyme have been studied extensively. Here, we report the reconstitution of replicase activity by essential components of DNA polymerase holoenzyme from the pathogen Pseudomonas aeruginosa. We have expressed and purified the processivity factor (β), single-stranded DNA-binding protein, a complex containing the polymerase (α) and exonuclease (ϵ) subunits, and the essential components of the DnaX complex (τ3δδ′). Efficient primer elongation requires the presence of αϵ, β, and τ3δδ′. Pseudomonas aeruginosa αϵ can substitute completely for …

Understanding The Continual Reassessment Method For Dose Finding Studies: An Overview For Non-Statisticians, Elizabeth Garrett-Mayer

The Continual Reassessment Method (CRM) has gained popularity since its proposal by O’Quigley et al. [1]. Many variations have been published and discussed in the statistical literature, but there has been little attention to making the design considerations accessible to non-statisticians. As a result, some clinicians or reviewers of clinical trials tend to be wary of the CRM due to safety concerns. This paper presents the CRM in a non-technical way, describing the original CRM with some of its modified versions. It also describes the specifications that define a CRM design, along with two simulated examples of CRMs for illustration.

Automated Migration Analysis Based On Cell Texture: Method & Reliability, Jianfeng Qin, Thomas W. Chittenden, Ling Gao, Justin D D. Pearlman

Background: In this paper, we present and validate a way to measure automatically the extent of cell migration based on automated examination of a series of digital photographs. It was designed specifically to identify the impact of Second Hand Smoke (SHS) on endothelial cell migration but has broader applications. The analysis has two stages: (1) preprocessing of image texture, and (2) migration analysis.

Results: The output is a graphic overlay that indicates the front lines of cell migration superimposed on each original image, with automated reporting of the distance traversed vs. time. Expert preference compares to manual placement of leading …

The Deep Structure Of Law And Morality, Robin B. Kar

This Article argues that morality and law share a deep and pervasive structure, an analogue of what Noam Chomsky calls the “deep structure” of language. This structure arises not to resolve linguistic problems of generativity, but rather from the fact that morality and law engage psychological adaptations with the same natural function: to allow us to resolve social contract problems flexibly. Drawing on and extending a number of contemporary insights from evolutionary psychology and evolutionary game theory, this Article argues that we resolve these problems by employing a particular class of psychological attitudes, which are neither simply belief-like states nor …
