We exploited the unique ecological niche of oil fly larval guts to isolate a strain of Staphylococcus haemolyticus which may be the most solvent-tolerant gram-positive bacterium yet described. This organism is able to tolerate 100% toluene, benzene, and p-xylene on plate overlays and saturating levels of these solvents in monophasic liquid cultures. A comparison of membrane fatty acids by gas chromatography after growth in liquid media with and without toluene showed that in cells continuously exposed to solvent the proportion of anteiso fatty acids increased from 25.8 to 33.7% while the proportion of 20:0 straight-chain fatty acids decreased from …

OBJECTIVE: To obtain information on the feasibility of employing recombinant human bone morphogenetic protein-7 (rhBMP-7), mixed with bone marrow as a substitute for autologous bone graft. METHODS: We carried out the study in the University Clinic Center, Sarajevo, Bosnia and Herzegovina, from February 2002 to January 2004. Histone Lys methylation plays an important role in determining chromatin states and is mostly catalyzed by SET domain– containing proteins. The outcome, transcriptional repression or activation, depends on the methylated histone residue, the degree of methylation, and the chromatin context. Dimethylation or trimethylation of histone H3 Lys 4 (H3K4me2 or H3K4me3) has been correlated with transcriptionally competent/active genes. However, H3K4 methylation has also been implicated in gene silencing. This dualistic nature of the H3K4 methyl mark has thus far remained unresolved. An approach for combined imaging of elastic and electromechanical properties of materials, referred to as piezoacoustic scanning probe microscopy (PA-SPM), is presented. Applicability of this technique for elastic and electromechanical imaging with nanoscale resolution in such dissimilar materials as ferroelectrics and biological tissues is demonstrated. The PA-SPM signal formation is analyzed based on the theory of nanoelectromechanics of piezoelectric indentation and signal sensitivity to materials properties and imaging conditions. It is shown that simultaneous measurements of local indentation stiffness and indentation piezocoefficient provide the most complete description of the local electroelastic properties for transversally isotropic materials, thus making piezoacoustic SPM …

A Wheat streak mosaic virus (WSMV) genome lacking HC-Pro was constructed and confirmed by reverse transcription-PCR to systemically infect wheat, oat, and corn. Coupled in vitro transcription/translation reactions indicated that WSMV P1 proteinase cleaved the polyprotein at the P1/P3 junction of the HC-Pro null mutant. The WSMV HC-Pro null mutant was competent for virion formation, but the virus titer was reduced 4.5-fold relative to that of the wild type. Collectively, these results indicate that WSMV HC-Pro is dispensable for replication and movement, two essential processes that are disrupted by point and small-insertion mutations introduced into potyvirus HC-Pro.

This research extends a mathematical framework to select the optimal sensor ensemble and fusion method across multiple decision thresholds subject to warfighter constraints for a combat identification (CID) system. The formulation includes treatment of exemplars from target classes on which the CID system classifiers are not trained (out-of-library classes) and enables the warfighter to optimize a CID system without explicit enumeration of classifier error costs. A time-series classifier design methodology is developed and applied, yielding a multi-variate Gaussian hidden Markov model (HMM). Glutamate151 (E151) has been shown to be catalytically essential for the aminopeptidase from Vibrio proteolyticus (AAP). E151 acts as the general acid/base during the catalytic mechanism of peptide hydrolysis. However, a glutamate residue is not the only residue capable of functioning as a general acid/base during catalysis for dinuclear metallohydrolases. Recent crystallographic characterization of the d-aminopeptidase from Bacillus subtilis (DppA) revealed a histidine residue that resides in an identical position to E151 in AAP. As a result of the morbidity associated with anterior cruciate ligament (ACL) reconstruction with a bone–patellar–tendon– bone graft, many orthopaedic surgeons prefer hamstrings as the graft for ACL reconstruction. However, this selection is not based on solid scientific evidence. In vitro research shows that this graft cannot restore control of tibial rotation. Our recent in vivo research work has also demonstrated the same result. In particular, patients undergoing ACL repair who were reconstructed with a quadrupled hamstring tendon graft showed excessive tibial rotation during a dynamic activity when compared with healthy control subjects. A protocol was developed and evaluated for nonovulating breastfeeding women to determine potential fertility with an electronic hormonal fertility monitor. The amount of required abstinence (i.e., days of potential fertility) through the first menstrual cycle indicated by the fertility monitor was significantly lower (17% of the total days) compared with the amount of abstinence (50% of the total days) indicated by the self-observation of cervical mucus.

Stream biota in urban and suburban settings are thought to be impaired by altered hydrology; however, it is unknown what aspects of the hydrograph alter fish assemblage structure and which fishes are most vulnerable to hydrologic alterations in small streams. We quantified hydrologic variables and fish assemblages in 30 small streams and their subcatchments (area 8-20 km 2) in the Etowah River Catchment (Georgia, USA). We stratified streams and their subcatchments into 3 landcover categories based on imperviousness (20% of subcatchment), and then estimated the degree of hydrologic alteration based on synoptic measurements of baseflow yield. High-Reflectance, Sputter-Deposited Aluminum Alloy Thin Thin Films For Micro-Electro-Mechanical Systems, Lance Barron

Theses

This study comprises an investigation of highly reflective thin film for microelectro- mechanical systems (MEMS) consisting of reflective components. For these applications, the desired film system must have (1) low stress, (2) high specular reflectivity, (3) improved nano-hardness relative to pure aluminum, (4) compatibility with traditional semiconductor fabrication techniques. This study is an in depth investigation of both the specular reflectance and mechanical properties of thin film reflectors formed by low-thermal (<200°C) processing. Six different aluminum films (containing Cu, Ti, Cr) were chosen based on extensive literature research. Each film was characterized by a variety of optical, electrical, and mechanical analytical techniques to obtain data relating microstructure to the film’s reflectivity and mechanical properties. A complete dielectric function for each deposited aluminum alloy has been developed utilizing Drude-Lorentz parameters and effective media approximations (EMA) to account for film surface topography. Results show that copper solute addition generate films that maintain much of the bulk reflectance of pure aluminum while refining surface morphology to create a more specular surface consisting of smaller, more uniform grains. Electron and atomic force microscopy and x-ray diffraction studies reveal that copper inclusion into the aluminum lattice cause both a reduction in the preferred orientation of the film and change in the lattice parameter. Copper concentrations of 1.0% and above lead to spatial variation in copper content within the films, with copper precipitating out of solution. Film hardness, resistivity, and stress increase as a function of copper content, with the absolute magnitude of stress being acceptable for MEMS applications. The addition of titanium and chromium to Al-1.0%Cu films cause further microstructure refinement as well as increased stress, resistivity, and hardness. Al-2.0%Ti-1.0%Cu and Al-2.0%Cr-1.0%Cu exhibit reflectivity spectra that differ significantly in both inter- and intraband absorption from that of pure aluminum. The Drude-Lorentz/EMA dielectric function model has been successfully applied to all alloy films explaining the observed reflectivity and showing an excellent agreement between measured and Drude resistivity. This study will enable to model and predict the optical response of thin films in terms of their intrinsic and extrinsic properties
