We compared the mechanistic and kinetic properties of murine leukemia virus (MuLV) and human immunodeficiency virus type 1 (HIV-1) reverse transcriptases (RTs) during RNA-dependent DNA polymerization and mutation synthesis using pre-steady-state kinetic analysis. First, MuLV RT showed 6.5-121.6-fold lower binding affinity (Kd) to deoxynucleotide triphosphate (dNTP) substrates than HIV-1 RT, although the two RTs have similar incorporation rates (kpol). Second, compared with HIV-1 RT, MuLV RT showed dramatic reduction during multiple dNTP incorporations at low dNTP concentrations. Care Group advising, as it was institutionally developed, is an academic advising method intended to improve the first year experience of undergraduate college students. The purpose of this study was to explore student perceptions of their participation in Care Group advising, as well as faculty and administrative perceptions. In order to evaluate the causes and conditions that lead to cyanobacteria blooms in northern temperate lakes, studies of nutrients such as total phosphorus, total nitrogen, phosphate, and nitrate are necessary. Sensitive, environmentally friendly and cost-effective techniques were developed to detect ppb-level nitrates and phosphates in the lake water. Phosphate was analyzed by luminol chemiluminescence, while the quantification of nitrate was performed by capillary electrophoresis (CE) using indirect UV detection, with a background electrolyte consisting of pyromellitic acid, hexamethonium hydroxide and triethanolamine. To date there are no analytical techniques designed to exclusively measure bioavailable iron in marine environments. The goal of this research is to develop such a technique by isolating the bioavailable iron using the terrestrial siderophore desferrioxamine B (DFB). This project contained many challenging aspects, but the specific goal of this study was to develop a robust analytical technique for quantification of Fe(III)-DFB complexes at nanomolar concentrations. Past work showed that oxalate (Ox) promotes photodissociation of Fe(III)-DFB to Fe(Il), and we are specifically interested in the mechanism of this process. Photochemical Generation Of 2-Methoxy-2methylpropylidene And Its Rearrangement Pathways, Stanislav Presolski

2-methoxy-2-methylpropylidene was photochemically generated from a non-nitrogenous precursor, 1a,9b-Dihydro-IH-cyclopropa[1]phenanthrene-l-etbane 2-methoxy-2-methyl, available in two steps from phenanthrene. Two competing rearrangement pathways were observed for the p-methoxy carbene, 1,2-methyl shift and C-H insertion, which had varying weights depending on the solvent used. Nevertheless, the C-H insertion was always predominant, contrary to the current understanding of B-substituted carbenes, which apparently needs revision. An attempt was also made to measure by LFP the rate constant of rearrangement and lifetime of the stabilized l-chloro-2-methoxy-2-metbylpropylidene.

The purpose of this project was to develop an upper level undergraduate biochemistry laboratory (CHEM 453) based upon a novel research project, designed to replace an existing instructional laboratory that employs ‘cookbook’ exercises. Methods for the expression, purification, spectral analysis of structural integrity, and electron transfer capability of rabbit cyt b5 were adapted from published work done by Dr. Lucy Waskell of the University of Michigan and Dr. Mansuy of the University of Paris. Microscale heat transfer and microfluidics have become increasingly important to overcome some very complex engineering challenges. The use of very small passages to gain heat transfer enhancement is a well documented method for achieving high heat flux dissipation. However, some interesting experimental results have caused researchers to question if the conventional theories for fluid flow and heat transfer are valid in the microscale passages. By viewing behavior regularities at the individual and collective level as functionally isomorphic, a referent-shift compositional model for the Big 5 personality dimensions is developed. On the basis of this compositional model, a common measure of Big 5 personality at the individual level is applied to the collective as a whole. Within this framework, it is also hypothesized that leadership (i.e., transformational, transactional, and passive) would predict collective personality and that collective personality would be significantly related to collective performance. Calendar-based methods are not usually considered effective or useful methods of family planning among health professionals. However, new “high-” and “low”-tech calendar methods have been developed, which are easy to teach, to use, and may be useful in helping couples avoid pregnancy. The low-tech models are based on a fixed-day calendar system. The high-tech models are based on monitoring urinary metabolites of female reproductive hormones. Both systems have high levels of satisfaction. This article describes these new models of family planning and the research on their effectiveness. Currently, forensic scientists are only able to determine time since death (or postmortem interval) up to the first 60 hours. This is based largely on insect activity. Herein, it is proposed to use the degradation of the intervertebral disc (IVD) after death to determine a relationship between the mechanical properties of cadaveric tissue and time since death in order to extend the 60-hour window. To that end, 1 fresh human spine and 6 pig spines were each separated into sections (6 human and 48 pig), with each section having one intact disc. Soil respiration is commonly estimated as the flux of CO2 emitted from the soil surface (Fc). It represents the sum of CO2 produced by root respiration and by heterotrophic decomposition of root exudates, soil organic matter, and plant litter. The influence of CO2 fixation by autotrophic soil microorganisms, as well as nonbiological reactions such as the chemical oxidation of organic molecules and the precipitation or dissolution of soil carbonates, is small in most situations (Suarez, 1999). Dispersal of European corn borer, Ostrinia nubilalis Hubner was examined by release and recapture of the dye marked adults and by capture of the feral adults in and around the large 50 ha center pivot irrigated fields of Bacillus thuringiensis (Bt) maize. Pheromone and black light traps were used to catch the adults. In 1999, 15 094 marked males and 7993 marked females were released, and in 2001, 13 942 marked males and 9977 marked females were released. Group I introns are catalytic RNAs with the ability to splice out of RNA transcripts, often without the aid of proteins. These self-splicing introns have been reengineered to create ribozymes with the ability to catalyze reactions. One such ribozyme, derived from a Pneumocystis carinii group I intron, has been engineered to sequence specifically remove a targeted segment from within an RNA substrate, which is called the trans excision-splicing reaction.The two catalytic steps of the trans excision-splicing reaction occur at positions on the substrate known as the 5' and 3' splice sites. Bean pod mottle virus (BPMV), a member of the genus Comovirus in the family Comoviridae, is widespread in the major soybean-growing areas in the United States. The complete nucleotide sequences of the genomic RNAs of the naturally occurring partial diploid strain IL-Cb1 were determined. Intermolecular RNA1 recombinants were isolated from strain IL-Cb1 and characterized at the molecular level. Structurally similar recombinant RNA1 was also generated after four passages in soybean derived from plants previously inoculated with a mixture of infectious RNA1 transcripts from two distinct strains. The National Program structure of USDA-ARS provides an opportunity to coordinate research on problems of national and global significance. A team of USDA-ARS scientists is conducting nationally coordinated research to develop predictions of manure N availability to protect water quality and improve farm solvency. Experimental design and research protocols were developed and used in common across all participating locations. Buffalograss [Buchloe dactyloides (Nutt.) Engelm.] is a perennial, warm-season turfgrass that is native to the shortgrass prairie of North America. It offers potential benefits as a turfgrass because of its drought resistance, low incidence of pest problems, and relatively low nutrient requirements. This study was initiated to evaluate the level of organelle DNA diversity among buffalograss genotypes and to determine the mode of inheritance of the chloroplast organelle by cpDNA and mtDNA PCR-RFLPs. The 56 genotypes studied included diploids, triploid, tetraploids, pentaploids, and hexaploids. 'Beefmaker' intermediate wheatgrass [Elytrigia intermedia (Host) Nevski subsp. intermedia = Thinopyrum intermedium subsp. intermedium (Host) Barkw. & D.R. Dewey] (Reg. no. CV-26, PI 634505) is a broadly adapted cultivar that produces forage with high in vitro dry matter digestibility (IVDMD) and high protein concentration in the tall, mid-grass, and short-grass ecoregions of the central Great Plains, USA. It was released in January 2003 by the USDA-ARS and the Agricultural Research Division, Institute of Agricultural and Natural Resources, University of Nebraska. Beefmaker was tested as NE TI1 C1.

The soilborne pathogens Rhizoctonia solani, Pythium ultimum, and Meloidogyne incognita can cause severe economic losses to field- and greenhouse-grown cucumber. A collection of bacterial isolates and isolates GL3 and GL21 of Trichoderma virens were screened for suppression of diseases caused by these pathogens. T. virens isolates GL3 and GL21 provided the most effective suppression of damping-off caused by R. solani in greenhouse bioassays. Burkholderia ambifaria BC-F, B. cepacia BC-1, and Serratia marcescens N1-14 also provided significant suppression of R. solani relative to the pathogen check in some experiments. All introduced natural enemies present a degree of risk to nontarget species. Since most biological control programs use relatively host-specific natural enemies, the risk to nontarget species is generally very low, particularly from biological control of weeds, which uses extensively tested and validated host-specificity testing procedures to predict risk. However, many of the published comments about risks of biological control are superficial or misleading, often inappropriately lumping risk from all taxa of agents as “the risk of biological control,” and ignore the potential benefits, rather than dealing with species-by-species risk and benefits.
