Systematics Of Mesocestoides (Cestoda: Mesocestoididae): Evaluation Of Molecular And Morphological Variation Among Isolates, Kerry A. Padgett, Steven A. Nadler, Linda Munson, Ben Sacks, Walter M. Boyce
Harold W. Manter Laboratory of Parasitology: Faculty Publications
A hypothesis-based framework was used to test if 3 genetic strains of Mesocestoides (clades A, B, and C) are distinct evolutionary lineages, thereby supporting their delimitation as species. For comparative purposes, 3 established cestode species, Taenia pisiformis, Taenia serialis, and Taenia crassiceps were assessed using the same methods. Sequence data from mitochondrial rDNA (12S) and the second internal transcribed spacer of nuclear rDNA (ITS-2) revealed derived (autapomorphic) characters for lineages representing clade A (n = 6 autapomorphies), clade B (n = 4), and clade C (n = 9) as well as T. pisiformis (n = 15) and T. serialis …

Harold W. Manter Laboratory of Parasitology: Faculty Publications
Individual specimens of Anisakis, Pseudoterranova, and Contracaecum collected from marine mammals inhabiting northern Pacific waters were used for comparative diagnostic and molecular phylogenetic analyses. Forty-eight new sequences were obtained for this study of 14 Anisakis taxa, 8 Pseudoterranova taxa, 4 Contracaecum taxa, and 4 outgroup species. Partial 28S (LSU) and complete internal transcribed spacer (ITS-1, 5.8S, ITS-2) ribosomal DNA was amplified by the polymerase chain reaction and sequenced. Sequences of ITS indicated that Pseudoterranova specimens from Zalophus californianus (California sea lion), Mirounga angustirostris (northern elephant seal), Phoca vitulina (harbor seal), Enhydra lutris (sea otter), and Eumetopias jubatus (Steller’s …

Predicting Plant Extinction Based On Species-Area Curves In Prairie Fragments With High Beta Richness, Brian J. Wilsey, Leanne M. Martin, H. Wayne Polley
United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications
Species-area relationships and island biogeography theory are commonly used to predict how species richness will decline with fragmentation. There are a variety of largely untested assumptions in these approaches, including the assumptions that populations are distributed uniformly before fragmentation, and that local extinctions are due to effects of small population sizes. If populations are not distributed uniformly, then populations can be abundant locally but rare globally. This would cause extinction rates to be smaller than predicted. We tested extinction theory by developing estimates of the number of plant species that should be present in small tallgrass prairie fragments and then testing …

Patterns Of Plant Species Diversity In Remnant And Restored Tallgrass Prairies, H. Wayne Polley, Justin D. Derner, Brian J. Wilsey
United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications
To restore diversity of native vegetation, we must understand factors responsible for diversity in targeted communities. These factors operate at different spatial scales and may affect the number and relative abundances of species differently. We measured diversity of plant species and functional groups of species in replicated plots within paired restored and remnant (relic) tallgrass prairies at three locations in central Texas, U.S.A.
A Modified Molybdenum Blue Method For Orthophosphate Determination Suitable For Investigating Enzymatic Hydrolysis Of Organic Phosphates, Zhongqi He, C. Wayne Honeycutt
A Modified Molybdenum Blue Method For Orthophosphate Determination Suitable For Investigating Enzymatic Hydrolysis Of Organic Phosphates, Zhongqi He, C. Wayne Honeycutt
United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications
In characterizing organic phosphorus (Po) by phosphatase hydrolysis, the quantity of hydrolyzable Po is represented by the difference in orthophosphate [i.e., inorganic P (Pi)] determined after and prior to enzymatic incubation. Therefore, precise determination of Pi is of major importance for accurate application of the enzymatic hydrolysis approach. The strong acid conditions required for conventional molybdenum blue methods interferes with Pi determination due to rapid hydrolysis of labile Po and precipitation of enzymes (proteins). The molybdenum blue method of Dick and Tabatabai in 1977 reduced errors pertaining to nonenzymatic hydrolysis of P …

Comparison Of Protein Solubilization Methods Suitable For Proteomic Analysis Of Soybean Seed Proteins, Savithiry Natarajan, Chenping Xu, Thomas J. Caperna, Wesley M. Garrett
United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications
Extraction of soybean seed proteins for two-dimensional polyacrylamide gel electrophoresis (2D–PAGE) and mass spectrometry analysis is challenging and inconsistent. In this study, we compared four different protein extraction/solubilization methods—urea, thiourea/urea, phenol, and a modified trichloroacetic acid (TCA)/acetone—to determine their efficacy in separating soybean seed proteins by 2D–PAGE. In all four methods, seed storage proteins were well separated by 2D–PAGE with minor variations in the intensity of the spots. The thiourea/urea and TCA methods showed higher protein resolution and spot intensity of all proteins compared with the other two methods. In addition, several less abundant and high molecular weight proteins were …

Size-Dependent Feeding And Reproduction By Boll Weevil (Coleoptera: Curculionidae), S. M. Greenberg, D. W. Spurgeon, Thomas W. Sappington, M. Sétamou
United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications
The considerable variation in adult size of the boll weevil, Anthonomus grandis grandis Boheman, has been well documented, but the influences of adult size on reproductive rate are not known. We examined the relationship between the size of boll weevils and their feeding and oviposition. Weevils weighed to the nearest milligram were grouped into five categories based on pupal weight:≤5, 6-10, 11-15, 16-20, and>20 mg. Numbers of lifetime punctures produced in flower buds (squares) of cotton, Gossypium hirsutum L., by both sexes of adults tended to increase with pupal weight. Boll weevil females with pupal weights >10 mg produced …
Bioprospecting, Philip A. Reed
Bioprospecting, Philip A. Reed
STEMPS Faculty Publications
The article discusses that the product applications of bioprospecting are almost limitless. This area of biotechnology has been labeled bioprospecting, and it is a practice that is creating worldwide controversy. Defined simply, bioprospecting is "scientific research that looks for a useful application, process, or product in nature." However, as with most biotechnologies, the definition does not address the complexities of bioprospecting. Archeologists are finding that some biotechnologies, such as the use of herbs for medicine and the use of fermentation and yeast in food products, date back 5,000 to 10,000 years (De Miranda, 2004). The four main categories of biotechnologies …

Practical tools are needed to identify and advance sustainable management practices to optimize economic return, conserve soil, and minimize negative off-site environmental effects. The objective of this article is to review current research in non-saline soils of the central U.S. to consider bulk soil electrical conductivity (ECa) as an assessment tool for: (1) tracking N dynamics, (2) identifying management zones, (3) monitoring soil quality trends, and (4) designing and evaluating field-scale experiments. The interpretation and utility of ECa are highly location and soil specific; soil properties contributing to measured ECa must be clearly understood. In soils …
Soil Heat Flux, Thomas J. Sauer, Robert Horton
Soil Heat Flux, Thomas J. Sauer, Robert Horton
United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications
Climatic conditions on the earth’s surface are in part a function of varying physical position (elevation, latitude, and aspect) and the influence of large-scale meteorological forces such as air and ocean currents. The density and architecture of plant canopies in natural systems are directly influenced by these climatic factors. By contrast, for agricultural systems, it is the crop canopies that often influence local microclimate. In both instances, the soil plays an important role in affecting climate near the surface. Properties of the surface soil layer including color, water content, texture, and density affect the partitioning of incident radiation and how …

United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications
Obesity is recognized as a major public health problem. Obesity is a multifactorial disease and is often associated with a wide range of comorbidities including hypertension, non-insulin dependent (Type II) diabetes mellitus, and cardiovascular disease, all of which contribute to morbidity and mortality. This review deals with stable isotope mass spectrometric methods and the application of stable isotopes to metabolic studies of obesity. Body composition and total energy expenditure (TEE) can be measured by mass spectrometry using stable isotope labeled water, and the metabolism of protein, lipid, and carbohydrate can be measured using appropriate labeled tracer molecules.

Nepetalactone, the primary component of catnip oil, was compared with the repellent N,N-diethyl-3-methylbenzamide (deet) for its ability to affect the host-seeking ability of Aedes aegypti (L.). A triple cage olfactometer was used to bioassay each substance and to assess its attraction inhibition (spatial repellent) attributes when combined with the following attractants: carbon dioxide, acetone, a blend of L-lactic acid and acetone, and human odors. Repellent tests were conducted with each substance against female Ae. aegypti, Anopheles albimanus Weidemann, and Anopheles quadrimaculatus Say. Catnip oil and deet were both weakly attractive to Ae. aegypti, catnip oil was the better spatial …
Habitat Corridors Function As Both Drift Fences And Movement Conduits For Dispersing Flies, Joanna H. Fried, Douglas J. Levey, Jerome Hogsette
Habitat Corridors Function As Both Drift Fences And Movement Conduits For Dispersing Flies, Joanna H. Fried, Douglas J. Levey, Jerome Hogsette
United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications
Corridors connect otherwise isolated habitat patches and can direct movement of animals among such patches. In eight experimental landscapes, we tested two hypotheses of how corridors might affect dispersal behavior. The Traditional Corridor hypothesis posits that animals preferentially leave patches via corridors, following them into adjacent patches. The Drift Fence hypothesis posits that animals dispersing through matrix habitat are diverted into patches with corridors because they follow corridors when encountered. House flies (Musca domestica L.), a species that prefers the habitat of our patches and corridors, were released in a central patch (100x100 m) and recaptured in peripheral patches …
The Politics Of Infertility: Recognizing Coverage Exclusions As Discrimination, Elizabeth Pendo
The Politics Of Infertility: Recognizing Coverage Exclusions As Discrimination, Elizabeth Pendo
All Faculty Scholarship
Infertility affects approximately ten percent of the reproductive-age population in the United States, and strikes people of every race, ethnicity and socio-economic level. It is recognized by the medical community as a disease, one with devastating physical, psychological, and financial effects.
In 1998, the Supreme Court held in Bragdon v. Abbott that reproduction is a major life activity within the meaning of the ADA. Many lawyers, activists and scholars thought that coverage for infertility treatment would follow soon after. In fact, in 2003 in the first major case applying Bragdon to health benefits, Saks v. Franklin Covey, the Second Circuit …
Mitochondrial Dna Mutations, Apoptosis, And The Misfolded Protein Response., Justin L. Mott, Dekui Zhang, Hans Peter Zassenhaus
Mitochondrial Dna Mutations, Apoptosis, And The Misfolded Protein Response., Justin L. Mott, Dekui Zhang, Hans Peter Zassenhaus
Journal Articles: Biochemistry & Molecular Biology
Studies of transgenic mice with accelerated accumulation of mtDNA mutations specifically in the heart lead us to propose that apoptotic signaling and cell death is central to the pathogenesis of mtDNA mutations in aging. It is the cellular response to that apoptotic signaling and the organ?s compensatory response to a loss of cells that specify the phenotype of an accumulation of mtDNA mutations. In the heart, cardiomyocytes induce a vigorous anti-apoptotic, pro-survival response to counteract mitochondrial apoptotic signaling. The heart up-regulates contractility of remaining myocytes in order to maintain cardiac output. We hypothesize that mutant mitochondrial proteins originate apoptotic signaling …

Climatic matching and pre-release performance evaluation were useful predictors of parasitoid establishment in a retrospective analysis of a classical biological control program against Bemisia tabaci biotype ‘‘B’’ in the USA. Laboratory evaluation of 19 imported and two indigenous parasitoid species in quarantine on B. tabaci showed that the Old World Eretmocerus spp, had the highest attack rate. The climate matching program CLIMEX was used to analyze the establishment patterns of five Old World Eretmocerus spp. introduced to the Western USA. The top matches ±10% for the climate of the area of introduction and origin of the introduced parasitoids always included …
A Study Of The Abundance, Distribution And Daily Activities Of The Australian Raven (Corvus Coronoides) In Urban Wetland Parks, Desiree L. Moon
A Study Of The Abundance, Distribution And Daily Activities Of The Australian Raven (Corvus Coronoides) In Urban Wetland Parks, Desiree L. Moon
Theses : Honours
Habitat destruction as a result of urbanisation has resulted in the population decline of some bird species. However, the Australian Raven (Corvus coronoides), has adapted well to the urban environment and is thriving. Although naturally occurring seasonal food sources are at times restricted in an urban setting, this adaptive species finds alternative food sources. As carrion-eaters this includes roadkill, as well as refuse discarded on the ground and into bins. Coupled with few natural predators, the species enjoys optimal conditions for breeding and survival. This biogeographical study investigated whether seasonal-influxes of populations of ravens cause problems at three urban wetland …
Is The Ability To Regenerate Damaged Skeletal Muscle Genetically Determined? : Can Allelic Variation In The Gene, Pax7, Explain Observed Differences In Recovery Of Force Following Damage By Eccentric Exercise?, Zoran Groznica
Theses : Honours
High force eccentric muscle actions, during forcibly lengthening contracting muscle, have been found to produce pronounced muscle damage, especially if exercise is unaccustomed. A cascade of events is initiated in order to repair damaged skeletal muscle tissue including degeneration and regeneration processes. For the duration of muscle regeneration, satellite cells are the primary source of new myoblasts in adult muscle. This process requires activation of quiescent mononucleated satellite cells which proliferate, differentiate and fuse together to form fully functional muscle fibres. The satellite cells are specified by the gene, Pax7, during development. Experimental studies of Pax7 null mice that lack …
Towards Persistent Resource Identification With The Uniform Resource Name, Luke Brown
Towards Persistent Resource Identification With The Uniform Resource Name, Luke Brown
Theses : Honours
The exponential growth of the Internet, and the subsequent reliance on the resources it connects, has exposed a clear need for an Internet identifier which remains accessible over time. Such identifiers have been dubbed persistent identifiers owing to the promise of reliability they imply. Persistent naming systems exist at present, however it is the resolution of these systems into what Kunze, (2003) calls "persistent actionable identifiers" which is the focus of this work. Actionable identifiers can be thought of as identifiers which are accessible in a simple fashion such as through a web browser or through a specific application. This …

Sound Scripts
This paper will continue to trace negotiations outlined in Part 1 of the music/noise dichotomy as expressed in modernist and postmodernist works.1 Drawing connections with the trajectory of "glitch" in popular music since the 1970s. The paper will examine a number of key ways in which the music/noise dichotomy has been addressed as a borderline dispute between, for example, the embodied and the disembodied, the scored and the unscored, the accidental and the intentional, sense and nonsense, culture and nature. Two key figures from the highly influential group of sound artists who came together at Melbourne's Clifton Hill Community Centre …

Civil and Environmental Engineering Faculty Publications and Presentations
The continued decline of Columbia River salmon (Oncorhynchus spp.) populations has long focused concerns on habitat changes upriver, particularly the effects of large hydroelectric dams. Increasing evidence that ocean conditions strongly influence salmon production, however, has raised questions about the importance of the estuarine environment to salmon and whether the hydropower system has affected estuarine-rearing habitats. In response to Northwest Power Planning Council recommendations, we initiated a review of what is known about the effects of the hydroelectric system on the hydrology, habitats, and ecology of the Columbia River estuary. Our goal was to develop recommendations for improving estuarine conditions …

Estimation Of Columbia River Virgin Flow: 1879 To 1928, Pradeep K. Naik, David A. Jay
Civil and Environmental Engineering Faculty Publications and Presentations
The Columbia River has historically been a major source of economic activity for the Pacific Northwest, and is one of the more heavily modified rivers in the United States today. Understanding human and climate-induced changes in its hydrologic properties is, therefore, a topic of considerable interest. Long streamflow records are essential to determining how runoff has changed over time. Daily streamflow records of the Columbia River at The Dalles dates back to June 1978. However, the observed daily flow does not alone provide enough information to understand or separate anthropogenic and climate effects. It is necessary also to have an …

African American Breast Cancer Health Disparities, Theresa Lynn Gaul
African American Breast Cancer Health Disparities, Theresa Lynn Gaul
Honors Program Theses
Health disparities are "the differences in the incidence, prevalence, mortality, and burden of cancer and related adverse health conditions that exist among specific population groups in the United States" (National Cancer Institute [NCI], 2002). Racial and ethnic minorities experience serious disparities in health access and outcomes. Disparities in the burden of death and illness experienced by African Americans in particular, as compared with our nation's population as a whole, are particularly striking. There are a variety of factors contributing to these inequalities. The purpose of this report is to examine these factors and the consequential effects on the African American's …
