Insulators are DNA elements that establish independent transcriptional domains within eukaryotic genomes. The Drosophila scs and scs′ insulators localize near the borders of a structural domain in the polytene chromosomes, known as a puff, produced by transcription of the 87A heat shock protein (hsp) genes. It has been suggested that scs and scs′ are boundary elements that delimit this decondensed chromatin domain, reflecting the mechanism by which these sequences act to constrain regulatory interactions. Seawater Acclimation Causes Independent Alterations In Na +/Tk+- And H+-Atpase Activity In Isolated Mitochondria-Rich Cell Subtypes Of The Rainbow Trout Gill, Guy S. Hawkings, Fernando Galvez, Greg G. Goss

Mitochondria-rich cells (MR cells) of the gills of rainbow trout undergo changes in relative distribution and biochemical function during acclimation to partial-strength (10‰) and full-strength (30‰) seawater. In isolated total gill cells, Na+/K+-ATPase activity increased fivefold and H+-ATPase activity decreased fourfold when trout were acclimated to either 10‰ or 30‰ seawater. When total MR gill cells were separated based on differential binding to peanut lectin agglutinin (PNA), the PNA subtypes underwent a change in relative distribution in seawater-acclimated fish. In freshwater, the ratio of PNA-: PNA+ was 65:35 while in seawater the distribution changed to 20:80 PNA-:PNA+. Additionally, differential changes …

A computational, rigid body model of a 50th percentile male head and neck utilizing 15 Hill Muscle pairs is used to study the sensitivity of Hill Muscle Model parameters. A 15g linear acceleration is applied within the transverse plane at the lowest vertebral level of the neck (T1). The resultant linear acceleration of the head is analyzed. In response is minimally affected. The peak accelerations did change, and in the case of varying muscle activation, the peak acceleration changed significantly, 36%. Each of the other parameter variations affected the peak acceleration of the head by less than 5%. Overall, …

Use Of Ultrasound Imaging To Determine Sex Of Shovelnose Sturgeon, Robert E. Colombo, Paul S. Wills, James E. Garvey

During November 2002, 51 shovelnose sturgeon Scaphirhynchus platorynchus were sexed by ultrasound imaging using a portable ultrasonograph. We identified males with 96% accuracy (N = 25) and females with 80% accuracy (N = 25); one hermaphroditic individual was misidentified as a male. Overall, ultrasound imaging was 86% accurate. Sex in postspawned females was difficult to determine, 60% being misidentified as males (N = 5). Ultrasonography is an effective noninvasive method for sex determination that can be applied to other species of Acipenseriformes. Modern portable equipment expands its utility to field studies.

To generate reliable age information for fishes, we must assess the robustness of techniques to environmental variation. We quantified daily ring deposition within the sagittal otoliths of known-age larval and juvenile redspotted sunfish Lepomis miniatus reared in ponds. We also determined how daily light : dark cycles mimicking natural conditions, reduced daylight, or constant daylight affected ring deposition in fish housed in aquaria. Additionally, we quantified how constant temperature (26°C), low daily variability in temperature (26 ± 1°C), and high daily variability (26 ± 2°C) affected production of daily rings. Providing children's disclosures to their mental health therapists greater protection in juvenile dependency cases recognizes the importance of privacy in therapeutic communications. Numerous children are required by juvenile courts to divulge the most intimate details of their lives to mental health experts only to have those disclosures revealed in court proceedings. Reversing the presumption that children's mental health records may be discussed openly in dependency litigation and requiring courts to perform in camera reviews affords children the dignity and respect adults take for granted. Ensuring greater confidentiality for children's mental health records is one step the legal system should take …

Anxiety sensitivity is a known precursor to panic attacks and panic disorder, and involves the misinterpretation of anxiety-related sensations. Aerobic exercise has been shown to reduce generalized anxiety, and may also reduce anxiety sensitivity through exposure to feared physiological sensations. Accordingly, 54 participants with elevated anxiety sensitivity scores completed six 20-min treadmill exercise sessions at either a high-intensity aerobic (n = 29) or low-intensity (n = 25) level. Self-ratings of anxiety sensitivity, fear of physiological sensations associated with anxiety, and generalized anxiety were obtained at pretreatment, post-treatment, and one-week follow-up. This study determined the lipid content and FA composition of muscle and a mixture of muscle and viscera from Korean catfish as well as lipid oxidation and hydrolysis. Lipid content and FA compositions in Korean catfish, which were purchased every month or two during September 1999–July 2000, were analyzed. Lipid oxidation and hydrolysis were determined as PV, thiobarbituric acid value, and FFA in the muscle and the mixture during storage at 2°C for 12 d and –14°C for 9 wk. Lipid contents of the muscle and the mixture were 3.2 (w/w) and 5.4%, respectively. Oleic acid was the most abundant …

Models represent our primary method for integration of small-scale, process-level phenomena into a comprehensive description of forest-stand or ecosystem function. They also represent a key method for testing hypotheses about the response of forest ecosystems to multiple changing environmental conditions. This paper describes the evaluation of 13 stand-level models varying in their spatial, mechanistic, and temporal complexity for their ability to capture intra- and interannual components of the water and carbon cycle for an upland, oak-dominated forest of eastern Tennessee. The European Green Crab (Carcinus maenas) is a global invader, successfully colonizing many world regions and having significant ecological and economic impacts. The Green Crab colonized western North America in the late 1980s, spreading primarily northward from the initial establishment in San Francisco Bay to several other bays in northern California, Oregon, and Washington. Initial analysis, based largely upon temperature tolerance of postlarval crabs, suggests Green Crabs will continue to spread and become established throughout much of Alaska. Genomics-driven growth in the number of enzymes of unknown function has created a need for better strategies to characterize them. Since enzyme inhibitors have traditionally served this purpose, we present here an efficient systems-based inhibitor design strategy, enabled by bioinformatic and NMR structural developments. First, we parse the oxidoreductase gene family into structural subfamilies termed pharmacofamilies, which share pharmacophore features in their cofactor binding sites. Then we identify a ligand for this site and use NMR-based binding site mapping (NMR SOLVE) to determine where to extend a combinatorial library, such that diversity elements are directed into the adjacent substrate site. …

The highly polymorphic D1S80 locus has no known genetic function. However, this variable number of tandem repeats (VNTR) locus has been highly valuable in forensic identification. In this study we report the allele and genotype frequencies of five African populations (Benin, Cameroon, Egypt, Kenya, and Rwanda), which can be used as databases to help characterize populations and identify individuals. The allele frequencies were used to infer genetic associations through phylogenetic, principal component, and G test statistical analyses. Compliance with Hardy-Weinberg equilibrium expectations was determined as were FST estimates, theta p values, and power of discrimination assessment for each population. …
