Atomic Force Microscopy (AFM) has been used to image the morphology of developing neurons and their processes. Additionally, AFM can physically interact with the cell under investigation in numerous ways. Here we use the AFM to both three-dimensionally image the neuron and to inflict a nano/micro-puncture to its membrane. Thus, the same instrument used as a tool to precisely penetrate/cut the membrane at the nanoscale level is employed to image the morphological responses to damage. These first high resolution AFM images of living chick dorsal root ganglion cells and cells of sympathetic ganglion and their growing processes provide confirmation of …

This Article addresses ethical and malpractice issues arising from the fact that attorneys who prosecute patents almost inevitably add to the inventor's original disclosure to the attorney. In the course of drafting a patent application-a process in which the attorney describes, necessarily in his own words, what the client has invented-the attorney will, at minimum, contribute ideas, thoughts, and means of expression that the client had not used. The application is not a verbatim transcript of an interview with the client; it is the creation of the patent lawyer. ...

Information on the habitat requirements and effects of habitat management on Greater Sage-Grouse was summarized from information in more than 5,500 published and unpublished papers. A range map is provided to indicate the current range of Greater Sage-Grouse (adapted from Schroeder et al. 2004). Although birds may be observed outside the breeding range indicated, the maps are intended to show areas where managers might concentrate their attention. Transcriptional Profiling Of Caulobacter Crescentus During Growth On Complex And Minimal Media, Craig Stephens, Alison K. Hottes, Maliwan Meewan, Desiree Yang, Naomi Arana, Pedro Romero, Harley H. Mcadams.

Microarray analysis was used to examine gene expression in the freshwater oligotrophic bacterium Caulobacter crescentus during growth on three standard laboratory media, including peptone-yeast extract medium (PYE) and minimal salts medium with glucose or xylose as the carbon source. Nearly 400 genes (approximately 10% of the genome) varied significantly in expression between at least two of these media. The differentially expressed genes included many encoding transport systems, most notably diverse TonB-dependent outer membrane channels of unknown substrate specificity. Amino acid degradation pathways constituted the largest class of genes induced in PYE. In contrast, many of the genes upregulated in minimal …

Successful management of aquatic resources requires comprehensive maps that identify the spatial distribution and extent of potentially vulnerable habitats and their resident biota (e.g., essential fish habitat). We illustrate a technique that describes submerged landscapes not only in terms of geophysical properties, but also in terms of the resident biotic communities. Biological community distribution was highly correlated with the geophysical provinces initially identified by Bell et al. (2000). However, fewer groups of stations (created mainly by combining provinces) explained almost as much of the variation in benthic community structure and was a better description of the major biotopes in this …

From Animal Abuse To Interhuman Violence? A Critical Review Of The Progression Thesis, Piers Beirne

This paper reviews evidence of a progression from animal abuse to interhuman violence. It finds that the ''progression thesis" is supported not by a coherent research program but by disparate studies often lacking methodological and conceptual clarity. Set in the context of a debate about the theoretical adequacy of concepts like "animal abuse" and ''animal cruelty," it suggests that the link between animal abuse and interhuman violence should be sought not only in the personal biographies of those individuals who abuse animals but also in those institutionalized social practices where animal abuse is routine, widespread, and socially acceptable.

Accounting For Activity: Cognition And Oral Proficiency Assessment, Miguel Mantero

Framing the present investigation within activity theory, this research analyzes the speaking guidelines of the American Council on the Teaching of Foreign Languages (ACTFL) and places the role of cognition and co-construction of meaning at the forefront of determining oral proficiency in a foreign language. Eight Spanish Oral Proficiency Interviews (OPIs) were analyzed in an effort to understand the overall role of cognition in developing and assessing oral proficiency. The findings are that although the ACTFL Proficiency Guidelines–Speaking provide for an understanding of the various linguistic functions involved in speaking, they do not completely reflect the level of cognition evident …

Microbial ecologists have discovered novel rRNA genes (rDNA) in mesophilic soil habitats worldwide, including sequences that affiliate phylogenetically within the division Crenarchaeota (domain Archaea). To characterize the spatial distribution of crenarchaeal assemblages in mesophilic soil habitats, we profiled amplified crenarchaeal 16S rDNA sequences from diverse soil ecosystems by using PCR-single-stranded-conformation polymorphism (PCR-SSCP) analysis. PCR-SSCP profiles provide a measure of relative microbial diversity in terms of richness (number of different phylotypes as estimated from the number of unique PCR-SSCP peaks) and evenness (abundance of each phylotype as estimated from the relative area under a peak). We have examined the chemical denaturations of the Klentaq and Klenow large-fragment domains of the Type 1 DNA polymerases from Thermus aquaticus (Klentaq) and Escherichia coli (Klenow) under identical solution conditions in order to directly compare the stabilization energetics of the two proteins. The high temperature stability of Taq DNA polymerase is common knowledge, and is the basis of its use in the polymerase chain reaction. This study, however, is aimed at understanding the thermodynamic basis for this high-temperature stability. Chemical denaturations with guanidine hydrochloride report a folding free energy (ΔG) for Klentaq that is over 20 kcal/mol more favorable …

A new difference probe for nuclear magnetic resonance (NMR) spectroscopy is presented. The difference probe uses two saddle-shaped coils to excite and detect two samples simultaneously. The samples are held in a specially modified 3-mm NMR tube with an Ultem plastic disk to separate the samples. The probe's resonant circuit contains two crossed diodes that passively switch the relative phase of each coil during the NMR experiment. The result is a difference spectrum from the two samples. The degree of cancellation of common signals was determined to be approximately 90%, and the application of the probe to relaxation-edited difference spectroscopy …

In recent years, researchers have increasingly turned their attention to the proliferation of small arms, a transnational trade amounting to over $7 billion in value during 2002. Small arms are difficult to track and are not the stuff of military parades, but they are immensely destructive. The trade in small arms should be understood not as a market but as a network, one that shares some important properties with networked forms of organization studied by sociologists. I make this argument and then employ quantitative methods developed for the study of social networks in an effort to show the basic structure …

Molecular investigations of deep-level relationships within and among the animal phyla have been hampered by a lack of slowly evolving genes that are amenable to study by molecular systematists. To provide new data for use in deep-level metazoan phylogenetic studies, primers were developed to amplify a 1.3-kb region of the alpha subunit of the nuclear-encoded sodium-potassium ATPase gene from 31 bilaterians representing several phyla. Maximum parsimony, maximum likelihood, and Bayesian analyses of these sequences (combined with ATPase sequences for 23 taxa downloaded from GenBank) yield congruent trees that corroborate recent findings based on analyses of other data sets (e.g., the …

The course of study of time-of-day effects on human performance has not been an easy one to chart, with many findings that seem to be in opposition. This review examines the difference between group and individual differences with regard to time-of-day effects; time-of-day effects in individuals; morningness-eveningness as an individual characteristic; morningness-eveningness in adolescents; effect of time of day on cognition and academic performance; time-of-day effects on intelligence, testing, and academic achievement; the effect of matching individuals to their preferred time on academic achievement; and motivation as a primary confounding variable in time-of-day preference/academic performance studies.
