Theses and Dissertations

The Internet and other large computer networks have become an integral part of numerous daily processes. Security at the network layer is necessary to maintain infrastructure survivability in the case of cyber attacks aimed at routing protocols. In order to minimize undesired overhead associated with added security at this level, the notion of selective security is proposed. This thesis identifies elements in network topologies that are most important to the survivability of the network. The results show that the strategic placement of network security at critical elements will improve overall network survivability without the necessity of universal deployment.

A Single Amino Acid Substitution In Soybean Vspα Increases Its Acid Phosphatase Activity Nearly 20-Fold, Oranuch Leelapon, Gautam Sarath, Paul E. Staswick

Soybean [Glycine max (L.) Merr.] contains two proteins called vegetative storage proteins (VSPs) that function as temporary storage reserves, but are also closely related to plant acid phosphatases of the haloacid dehalogenase (HAD) superfamily. This study examined the biochemical basis for the relatively low catalytic activity previously reported for these VSPs. The specific activity of purified recombinant VSPα on GMP was about 40-fold lower than for a related soybean root nodule acid phosphatase (APase), which had a specific activity of 845 U mg^-1 protein. Conversion of Ser¹⁰⁶ to Asp increased VSPα activity about 20-fold. This Asp residue …

The large coiled-coil protein NuMA plays an essential role in organizing microtubule minus ends at spindle poles in vertebrate cells. Here, we use both in vivo and in vitro methods to examine NuMA dynamics at mitotic spindle poles. Using fluorescence recovery after photobleaching, we show that an exogenously expressed green-fluorescent-protein/NuMA fusion undergoes continuous exchange between soluble and spindle-associated pools in living cells. These dynamics require cellular energy and display an average half-time for fluorescence recovery of approximately 3 minutes. A report recently released by an external review panel evaluates a controversial five-year contract between the University of California, Berkeley, and Novartis, the Swiss pharmaceutical and agrochemical company now known as Syngenta. The report, which was written by a team of consultants from Michigan State University and released in early August, addresses a number of issues related to the increasingly complex relationship between public universities and private industry. Given the size of the agreement and that it involved an entire academic department, the Berkeley-Novartis contract quickly became a focus for concerns relating to the basic mission of land-grant universities and …

Structural genomics is providing a means to determine the molecular and cellular function for the vast amount of proteins in the Human proteome that lack any explicit experimental information by characterizing the complete range of protein folds (Montelione, 2001). The Northeast Structural Genomics Consortium (NESG; http://www.nesg.org/) is a pilot project funded by the National Institutes of Health Protein Structure Initiative, focusing on proteins from eukaryotic model organisms including humans. The thermophillic archaea Archaeglobus fulgidis AF2095 protein is an example of a protein of unknown biological function targeted for structural analysis by NESG.
