Thermodynamic Characterization Of Dna Binding By Type I Dna Polymerases From Thermus Aquaticus And Escherichia Coli, Kausiki Datta

DNA binding of the DNA polymerase I from Thermus aquaticus (Taq and Klentaq fragment) and E. coli (Klenow fragment) have been studied as a function of [salt] and temperature in order to understand their DNA binding thermodynamics. Binding of the two different species of polymerases occurs with sub-micromolar affinities in very different salt concentration ranges. Thus, at similar [KCl] the binding of Klenow is ~ 3kcal/mol (150X) tighter than the binding of Taq/Klentaq. Linkage analysis of the [KCl] dependence of DNA binding at 25°C reveals a net release of 2-3 ions for Taq/Klentaq and 4-5 ions for Klenow. DNA binding …

Ant populations and their distribution are affected by many circumstances. Abiotic factors such as weather, water availability, and soil characteristics may determine if a habitat is suitable for ants. Other factors such as level of disturbance, available food resources, reproductive biology, and natural enemies also contribute to defining ant distribution. Three ant species in East Baton Rouge parish, Louisiana were studied to determine what factors affected their distribution by measuring abiotic and biotic factors at sampling sites throughout the parish. Health benefits from consuming rice bran oil (RBO) have been extensively studied in humans. RBO has been shown to decrease cholesterol absorption by inhibiting the oxidation of the LDL-cholesterol. Gamma-Oryzanol is one of the RBO components that possess antioxidative properties; it is a polyphenolic compound resembling tocopherol (vitamin E), which is another component of RBO. There are evidences that oryzanol has potential applications for enhancing shelf-life of foods containing fats processed at a high temperature, decreasing the total cholesterol, LDL-cholesterol and triglyceride levels in plasma lipids, and healing ulcers. This study generated data on tolerances, adaptabilities and production characteristics of four tilapia varieties. Goals of this study were to: (1) compare growth of four tilapia varieties in two types of recirculating systems, (2) evaluate juvenile growth of seven tilapia varieties in fresh and brackishwater mesocosms, and (3) estimate cold tolerance of juveniles of four tilapia varieties in fresh and brackishwater. Eight weaned Holstein steers approximately 4 mo of age (mean BW = 119.47 ± 2.65 kg) were used in a replicated 4 x 4 Latin square designed experiment to study the effects of protein source and level on performance and anabolic hormonal status of weaned dairy calves. Treatments consisted of two diets containing either 16 or 20 % crude protein (CP) with or without fish meal (FM). Standard identification criteria for creating a decedent’s biological profile typically include ancestry, sex, age, and stature, but not body weight. Body weight information may not only assist in creating a more complete biological profile but may also provide insight into other forensic considerations, such as taphonomy and body transport and disposal. The cranial anatomy of plesiosaurs in the family Polycotylidae (Reptilia: Sauropterygia) has received renewed attention recently because various skull characters are thought to indicate plesiosauroid, rather than pliosauroid, affinities for this family. New data on the cranial anatomy of polycotylid plesiosaurs is presented, and is shown to compare closely to the structure of cryptocleidoid plesiosaurs. The morphology of known polycotylid taxa is reported and discussed, and a preliminary phylogenetic analysis is used to establish ingroup relationships of the Cryptocleidoidea. The goal of this study was to develop several techniques that utilize the use of geothermal water that can contribute to increased profitability for commercial catfish producers. The primary objectives of this study were to: 1) determine the heating requirement using degree-days of channel catfish spawning 2) improve the efficiency of artificial spawning and, 3) evaluate the effectiveness of stocking outdoor ponds and pools with channel catfish fry. The heating requirement for channel catfish spawning was determined to be between 99 and 129 degree days at the peak of spawning. The purpose of this study was to investigate the response of participants in the first three Stages of Change (SOC) to a preparation stage-tailored intervention to increase fruit and vegetable (F&V) consumption. An on-line survey was delivered to 400 university employees, and included questionnaires about their demographic information, SOC, self efficacy (SE), and decisional balance (DB). Longleaf pine savannas are perhaps the most threatened ecosystems in North America. Despite a well documented and highly diverse flora, little has been published on insects in general and, in particular, on bees that provide the valuable service of plant pollination. Thus, the aims of this study were to: 1) survey bees found in two savanna types in southeastern Louisiana; and 2) contrast the diversity and species composition of these two savanna types. The intricate circuitry formed by amacrine cells in the inner plexiform layer (IPL) of the retina suggests that these interneurons play a major role in shaping the visual message. The majority of amacrine cells in the vertebrate retina are GABAergic. Thus, elucidating how GABAergic signaling is modulated in the IPL is critical in order to understand how the visual message is processed in the retina. The results presented here suggest that GABAergic signaling between amacrine cells can be modulated by the activation of metabotropic glutamate receptor 5 (mGluR5) or by the production of the second messenger, nitric oxide (NO). The Barataria-Terrebonne estuary has been eroding at a rate of up to 103.6 km2 yr-1 for several decades. If the current rate of loss is not reduced, an additional 2,550 km2 of coastal wetlands will be lost by the year 2050. Currently, stormwater in Terrebonne Parish is pumped into canals, ultimately discharging into the Gulf of Mexico. Random strandboards from mixed southern wood species were manufactured using calcium borate (CB) and zinc borate (ZB) as chemical additives and polymeric methylene diphenylmethane disocyanate (pMDI) as resin. There were four target levels of borate loading and two levels of resin content. Panel properties, including modulus of elasticity (MOE), modulus of rupture (MOR), internal bond (IB) strength, linear expansion (LE), thickness swelling (TS), leachability, and swelling and strength retention properties under cyclic relative humidity, were measured. This content analysis study examined 36 plant-centered children’s science picture books that have a plot and characters published from 1950 to present. Botanical subject matter and learning opportunities offered by these books were analyzed, along with the range and frequency of the National Science Education Standards-consistent and age-appropriate plant science concepts and principles. The science graphics, artistic innovations, and story plot of these books were also examined. The first event in the infection process of Ostertagia ostertagi in cattle is the process of exsheathment. Before trichostrongylid nematodes can transition from a free-living infective stage larva (L₃) on pasture to a parasitic existence within the ruminant host, it must first undergo exsheathment. Exsheathment is the process whereby the L₂ cuticle retained from the previous molt is cast from the L₃. Exsheathment enables the developmental transition from a free-living stage on pasture to a parasitic existence in the bovine host. During the third century C.E., a debate on the relationship between “capacity” (cai 才) and “nature” (xing 性) captured the imagination of the Chinese elite. Historical sources relate that four views were put forward on the subject, which continued to dominate the intellectual scene during the fourth and fifth centuries. Intersexual infants and infants with other genital abnormalities often receive genital surgery for sex assignment or for normalizing purposes. The wisdom and beneficence of these practices have been questioned by intersexual individuals, support groups, some doctors, and the media. Because the practices have been developed without long-term studies to evaluate them, pediatric urologists and parents of such children must face decisions with very little guidance from empirical support. In the face of ignorance about what is really the best medical response to intersexuality or genital abnormalities, some have argued for a moratorium on infant genital surgery until empirical studies are …

Microstructures in two mixed surfactants systems that show novel transformations as a function of composition and temperature have been explored. The Americans with Disabilities Act (ADA) was heralded as an "emancipation proclamation" for people with disabilities, one that would achieve their equality primarily through its reasonable accommodation requirements. Nevertheless, both legal commentators and Supreme Court Justices assert that the ADA's employment mandates distinguish the ADA from earlier antidiscrimination measures, most notably Title VII, because providing accommodations results in something more than equality for the disabled. The Article challenges this prevalent belief by arguing that ADA-mandated accommodations are consistent with other antidiscrimination measures in that each remedies exclusion from employment opportunity by questioning the inherency of established workplace norms, and by …

Although it is not a new environmental problem, overfishing is arguably one of the most serious environmental problems today. Despite decades of government regulation, fisheries are in trouble the world over.

With little doubt, the federal fishery regulations adopted in the last twenty-five years have failed to ensure sustainable utilization of U.S. fisheries. The existing regulatory framework is "a failed experiment that has led to the destruction of United States coastal fisheries."
