Production And Purification Of A Chimeric Monoclonal Antibody Against Botulinum Neurotoxin Serotype A , Mark C. Mowry, Michael Meagher Lead Investigator, Leonard Smith, Anuradha Subramanian

Papers in Biotechnology

Production of recombinant antibodies against botulinum neurotoxin is necessary for the development of a post-exposure treatment. CHO-DG44 cells were transfected with a plasmid encoding the light and heavy chains of a chimeric monoclonal antibody (S25) against botulism neurotoxin serotype A. Stable cell lines were obtained by dilution cloning and clones were shown to produce nearly equivalent levels of light and heavy chain antibody by an enzyme-linked immunosorbent assay (ELISA). In suspension culture, cells produced 35 μg/ml of chimeric antibody after 6 days, corresponding to a specific antibody productivity of 3.1 pg/cell/day. A method for the harvest and recovery of an …

The activation, localization, phenotypic changes, and function of CFSE-labeled naive influenza-specific CD8+ and CD4+ T cells following influenza infection were examined. Response of adoptively transferred CD8+ T cells was seen earliest in draining lymph node. Highly activated cells were found later in the lung, airways, and spleen, were cytolytic, and expressed IFN-γ upon restimulation. Similar amounts of division at early time points, but higher numbers of CD8+ T cells, were detected at 9 and 30 days postinfection after cotransfer of CD4+ and CD8+ T cells followed by infection. The legislative session that concluded in the spring of 2004 brought much-needed hope and some tangible results to higher education in Tennessee. Realizing that work still must be done, we are greatly appreciative of the salary improvements that were approved for our faculty and staff. ETSU also succeeded in acquiring some $11 million in capital outlay funding to renovate the old Sherrod Library, providing critically needed space for our College of Nursing. We have the largest nursing enrollment in Tennessee, and we have plans to increase enrollment in the B.S.N. program by 50 percent, a major step in our continued …

Investigation Of The Antibiotic Susceptibility Patterns Of Pathogens Causing Nosocomial Infections, Akgun Yaman, Yesim Tasova, Filiz Kibar, Ayse S. Inal, Nese Saltoglu, Ozlem Buyukcelik, Behice Kurtaran, Ismail H. Dundar

OBJECTIVE: The aim of this study is to determine the resistance patterns of bacteria causing nosocomial infections. The outcome of this resistance was followed for 3 years. METHODS: This study was carried out during 2000 to 2002 at a university hospital in Turkey. The resistance patterns of 570 bacteria (390 Gram-negative, 180 Gram-positive) against meropenem, imipenem, ceftazidime, cefotaxime, cefepime, piperacillin/tazobactam, ciprofloxacin and tobramycin were investigated using the E-test. Extended-spectrum beta-lactamase (ESBL) production was determined using ceftazidime and ceftazidime/clavulanic acid E-test strips. RESULTS: Meropenem was the most effective antibiotic against Gram-negative organisms (89.0%); this was followed by imipenem (87.2%) and piperacillin/tazobactam …

The cytosine deaminase APOBEC3G, in the absence of the human immunodeficiency virus type 1 (HIV-1) accessory gene HIV-1 viral infectivity factor (vif), inhibits viral replication by introducing G→A hypermutation in the newly synthesized HIV-1 DNA negative strand. We tested the hypothesis that genetic variants of APOBEC3G may modify HIV-1 transmission and disease progression. Single nucleotide polymorphisms were identified in the promoter region (three), introns (two), and exons (two). Genotypes were determined for 3,073 study participants enrolled in six HIV-AIDS prospective cohorts. Gun violence continues unabated. Regulation of these deadly instruments is woefully inadequate, and legislatures are compounding the problem by barring or restricting access to the courts for the death and injuries that guns cause. In short, Congress and state legislators have repeatedly acquiesced to the demands of the gun lobby.

During the past several years, cities have struck back by filing public nuisance claims against those gun sellers whose practices pose a risk to the public's health and safety. In June 2004, the Food and Drug Administration cleared the first application for leeches (Hirudo medicinalis) to be used in modern medicine as medical devices. By definition, a medical device is an article intended to diagnose, cure, treat, prevent, or mitigate a disease or condition, or to affect a function or structure of the body, that does not achieve its primary effect through a chemical action and is not metabolized.
