Facts: Shortly before the election for Prime Minister in 2001, the Knesset passed a law that gave certain persons an entitlement to grants for the purchase or extension of an apartment in Jerusalem. After the election, some six weeks later, the Knesset suspended the grants, and went so far as to include a provision to the effect that the suspension of the grants was retroactive, from the date on which the grants originally came into effect. Control Of Mitochondrial Motility And Distribution By The Calcium Signal: A Homeostatic Circuit., Muqing Yi, David Weaver, György Hajnóczky

Mitochondria are dynamic organelles in cells. The control of mitochondrial motility by signaling mechanisms and the significance of rapid changes in motility remains elusive. In cardiac myoblasts, mitochondria were observed close to the microtubular array and displayed both short- and long-range movements along microtubules. By clamping cytoplasmic [Ca2+] ([Ca2+]c) at various levels, mitochondrial motility was found to be regulated by Ca2+ in the physiological range. Maximal movement was obtained at resting [Ca2+]c with complete arrest at 1-2 microM. Movement was fully recovered by returning to resting [Ca2+]c, and inhibition could be repeated with no apparent desensitization. The inositol 1,4,5-trisphosphate- or …

Reconstitution Of Copii Vesicle Fusion To Generate A Pre-Golgi Intermediate Compartment, Dalu Xu, Jesse C. Hay

What is the first membrane fusion step in the secretory pathway? In mammals, transport vesicles coated with coat complex (COP) II deliver secretary cargo to vesicular tubular clusters (VTCs) that ferry cargo from endoplasmic reticulum exit sites to the Golgi stack. However, the precise origin of VTCs and the membrane fusion step(s) involved have remained experimentally intractable. Here, we document in vitro direct tethering and SNARE-dependent fusion of endoplasmic reticulum-derived COPII transport vesicles to form larger cargo containers. The assembly did not require detectable Golgi membranes, preexisting VTCs, or CON function. Therefore, COPII vesicles appear to contain all of the …

Consultation services in school settings is gaining increased recognition in research, training and practice, and is expanding to include a variety of related professionals as consultants (e.g., special educators, counseling psychologists, community psychologists, speech pathologists; Gutkin & Curtis, 1999). There also appears to be a trend in increased consultation between medical professionals (e.g., general physicians, pediatricians, psychiatrists) and school personnel. Whereas the term consultation may have broad meanings across educational, medical, and psychological practice, one common feature is that consultation generally consists of two or more people working together to address concerns regarding a third-party client (Sheridan & Kratochwill, 1991). …

Squared Extrapolation Methods (Squarem): A New Class Of Simple And Efficient Numerical Schemes For Accelerating The Convergence Of The Em Algorithm, Ravi Varadhan, Ch. Roland

We derive a new class of iterative schemes for accelerating the convergence of the EM algorithm, by exploiting the connection between fixed point iterations and extrapolation methods. First, we present a general formulation of one-step iterative schemes, which are obtained by cycling with the extrapolation methods. We, then square the one-step schemes to obtain the new class of methods, which we call SQUAREM. Squaring a one-step iterative scheme is simply applying it twice within each cycle of the extrapolation method. Here we focus on the first order or rank-one extrapolation methods for two reasons, (1) simplicity, and (2) computational efficiency. …
