Single-nucleotide polymorphisms (SNPs) provide an abundant source of DNA polymorphisms in a number of eukaryotic species. Information on the frequency, nature, and distribution of SNPs in plant genomes is limited. Thus, our objectives were (1) to determine SNP frequency in coding and noncoding soybean (Glycine max L. Merr.) DNA sequence amplified from genomic DNA using PCR primers designed to complete genes, cDNAs, and random genomic sequence; (2) to characterize haplotype variation in these sequences; and (3) to provide initial estimates of linkage disequilibrium (LD) in soybean. Feeding sub-therapeutic levels of antibiotics to livestock has been associated withdevelopment and spread of antibiotic resistant bacteria. The present experiment was conductedto investigate the effect of antibiotic alternatives (caffeic acid, chlorogenic acid, and carbadox)on the microbial ecology of swine feces in vitro.Minimum inhibitory concentrations of caffeic and chlorogenic acids were determined forseveral pathogens using macrobroth and agar dilution techniques. Gram-negative bacteria werenot inhibited. Caffeic acid inhibited four Staphylococcus aureus strains at 200 ppm or less, andtwo Clostridium perfringens strains at 300 ppm. Chlorogenic acid inhibited all S. aureus strainsat 500 ppm, and one C. perfringens strain at 400 ppm.Effects of …

The purpose of this study was to investigate the status of the science program in a selected Union Conference of the Seventh-day Adventist school system. Purpose of the Study. Female-on-female aggression is often inferred, or drawn from studies conducted with children or males. Little or no information is available that reports behaviors perceived as mistreatment or abuse among women. The purposes of this study were to investigate (a) behaviors demonstrated by women that women consider abuse or mistreatment; (b) the extent to which these perceptions of abuse/mistreatment were related to gender profiles; and (c) the extent to which personal experiences as victims or perpetrators of abuse were related to age, race, and education.

Problem. In an environment that challenges the non-profit sector, including church-based organizations, to do more with less, it is important to understand the mechanisms that build and sustain two key resources-individual giving and volunteering. Research indicates that there is a great deal of variation in the extent to which individuals provide help. An integrative theoretical model is utilized to examine and compare the giving and volunteering behaviors, socialization, attitudes, reasons, and motivations of adults with Seventh-day Adventist and other Christian religious identities, with secondary consideration given to the effect of potential ethnic variations.

We measured the dynamic I–V characteristics and vacuum ultraviolet (VUV) emission lines of the second continuum in xenon (170 nm) and argon (130.5 nm) from pulsed microhollow cathode discharges (MHCD). For pulse lengths between 1 and 100 μs the dynamic I–V characteristics are similar in both inert gases. Only the time variation of the VUV emission line at 170 nm for xenon can be related to the dimer excited states. In argon the energy transfer between the Ar*2 dimers and the oxygen impurity atoms is responsible for a qualitatively different time behavior of the resonance line at 130.5 nm. …

Yeast protoplasts from haploid auxotroph strains DC6 (MAT\alpha) and FY73 (MAT\alpha) were exposed to an inhomogeneous alternating field (AC 200 V/cm). Due to the dielectrophoretic aggregation two or more cells with close membrane contact were formed between the two electrodes. The effects of different DC pulses were investigated to determine critical fusogenic pulse strength. In the results, the maximum fusant cells were obtained only when 8 kV field strengths were applied. These findings showed that the optimum fusogenic pulse strength for yeast protoplasts is 8 kV/cm under our experimental conditions. After fusion, the DNA contents of both hybrid and individual …

Şereflikoçhisar Lake is the largest salt lake in central Turkey. This lake is a major source of solar salt for food, hide and other industries locally. Due to the economic importance of salt obtained from this lake, a microbial survey has been conducted. Six salt and three brine samples were obtained from the lake for physico-chemical and microbiological analyses. Physico-chemical analysis showed that the salts and brines contain sufficient ions and hardness to support extremely halophilic bacteria. Waste baker's yeast (Saccharomyces cerevisiae) was used as a biosorbent for Cu^{+2} biosorption. The yeast cells were treated with caustic soda, ethanol and heat to increase their biosorption capacity. Among the treatment methods used, the highest copper uptake (21.1 mg g^{-1}) was obtained with the caustic treatment of baker's yeast. The effect of initial copper concentration and pH on biosorption for caustic treated yeast was studied. The highest Cu^{+2} uptake (120.7 mg g^{-1}) was obtained at pH 4.0, for 198.2 mg I^{-1} initial copper ion concentration. Since the 1960s, biological macromolecules have occupied an increasingly important role in evolutionary and systematic studies. These biological macromolecules, used as sources of molecular characteristics, are proteins and various types of DNA such as a nuclear genome, nuclear ribosomal DNA, mitochondrial genome, and chloroplast genome. Electrophoresis and immunological techniques are used for generating systematic data from proteins. Generally, electrophoretic methods focus on two general forms of protein data, isozymes and allozymes, while the basis of immunological studies is antigen-antibody reactions. Seventy-four different Streptomyces isolates were obtained from the soils of Muğla province. Antimicrobial activity was determined in 45.9% of the isolates. Fifteen isolates showed strong activity against coagulase-negative Staphylococcus (CoNS). These isolates were extensively studied for their in vitro antimicrobial activity against Gram-positive and Gram-negative bacteria and yeasts. The results indicated that five isolates were highly active against CoNS and yeast with an inhibition zone at ? 20 mm. Three of these isolates were identified as Streptomyces antibioticus (MU106, MU107) and S. rimosus (MU114). The UV spectra of the culture extracts for the active isolates showed absorbance peaks ranging between …

It has become increasingly and widely recognised that bacteria do not live as isolated entities but instead exist as communities that exploit elaborate systems of intercellular communication to facilitate their adaptation to changing environmental conditions. A well-characterised example of such intercellular communication is quorum sensing. Quorum sensing depends on the production of diffusible signal molecules termed autoinducers or pheromones, which enable a bacterium to monitor its own cell population density. A variety of physiological processes in a range of bacterial species is regulated by quorum sensing. In this study, the success of in vivo and in vitro micrografts of pistachio (Pistacia vera L. cv. "Siirt") materials are presented. The only variable tested was age (1, 5, 10, and 30-year-old trees). Ten- to 12-day-old axenic seedlings germinated in vitro or seedlings (3 to 5 months-old) grown in pots in vivo were used as rootstocks. Shoot tips collected from the four age classes of mature trees of pistachio were the source of scions. Adventitious shoots were obtained from the diploid Diyarbakır watermelon cultivar "Sürme" by culturing cotyledons on shoot regeneration medium for 3 weeks. The effects of two cytokinins, benzyl amino purine (BA) and kinetine (Kin), on shoot organogenesis were examined. The number of shoots per explant were approximately more than 50% higher when 0.5 mg/l BA was used compared to the most effective kinetin concentration (1 mg/l). Plants were obtained by transferring 1 to 2 cm shoots to MS medium supplemented with 1-4 mg/l naphthalene acetic acid. Fifteen soil samples taken from the Bird Paradise National Park in Bandırma were investigated using the soil dilution plate and soil washing methods. Thirty-three species and four different sterile microfungal taxa were identified. Twenty-eight of these species belonged to the Hyphomycetes and 5 to the Mucorales. The genus with the highest species diversity found in the study area was Penicillium, represented by 16 species. Some chemical properties of the soil samples were also established. Lactic acid bacteria (LAB) are a key element in the dairy industry. The gene products encoded by the plasmids of LAB carry out fermentation. To date, many successful plasmid isolation studies have been conducted with Escherichia coli, Staphilococcus, Streptococcus, Salmonella, Listeria and many strains of LAB. Salivaricin B (SalB) is a protein encoded by Lactobacillus salivarius M7. This protein is insensitive to proteolytic enzymes, such as pepsin, trypsin, and chymotrypsin, but is partially sensitive to proteinase K. SalB is also stable and maintains its activity over a range of pHs, from 3 to 9. SalB was found to be effective against many closely related lactic acid bacteria such as, L. fermentum, L. plantarum, L. acidophilus and distantly related gram-positive bacteria, including Listeria monocytogenes, Streptococcus faecalis, Staphylococcus aureus and Staph. epidermidis. In experiments with Escherichia coli B/r WP2(trpE-), mice marrow cells and human lymphocytes the antimutagenic activity of an extract from Zizyphus mill was performed. The extract was tested separately and in mixture with Triticum aestivum. In experiments with different treatment schemes it has been established that the extract from wheat sprouts added to an antimutagenic preparation from Zizyphus mill intensifies the genoprotective properties of the Zizyphus mill extract.

The present study represents an investigation into the effect of dietary fatty acids on the fatty acid composition of the phospholipid fraction of Gambusia affinis, a small freshwater fish living in a source located on the campus of Dicle University. The fatty acid composition of the phospholipid fraction was analysed by gas chromatography for fish collected from the freshwater source, which were naturally fed, and also fish fed an artificial diet with a w6/w3 ratio of 5.5:1 for 30 days. The present study describes a multispecies toxicity test that uses community level responses of plankton samples from a natural lake using polyurythane foam (PF) as artificial substrate. These substrates were suspended and left for a period of 14 days. Colonized substrates were then placed in a series of microcosms dosed with a gradient of tannery effluent from 1 to 15% concentration.
