Breast-feeding has been linked to slowed postnatal growth. Although the basis for this "weanling’s dilemma" is unclear, environmental contaminants in human milk may be of relevance. We studied a Faroese birth cohort of 182 singleton children, born at term in 1994-95. Concentrations of mercury in cord blood and of polychlorinated biphenyls in maternal milk were measured, and duration of breast-feeding was recorded. At 18 months, children who had been exclusively breast-fed for at least 6 months weighed 0.59 kg less [95% confidence interval (CI) = 0.03, 1.16 kg] and were 1.50 cm [95% CI = 0.52, 2.47 cm] shorter than …

Until the 1990s, Amblyomma americanum was regarded primarily as a nuisance species, but a tick of minor importance as a vector of zoonotic pathogens affecting humans. With the recent discoveries of Ehrlichia chaffeensis, Ehrlichia ewingii, and “Borrelia lonestari,” the public health relevance of lone star ticks is no longer in question. During the next 25 years, the number of cases of human disease caused by A. americanum-associated pathogens will probably increase. Based on current trajectories and historic precedents, the increase will be primarily driven by biological and environmental factors that alter the geographic distribution and intensity of …

A Preliminary Linkage Map Of The Hard Tick, Ixodes Scapularis, A. J. Ullmann, J. Piesman, M. C. Dolan, W. C. Black Iv

Blackwell Publishing Ltd. A linkage map of the Ixodes scapularis genome was constructed, based upon segregation amongst 127 loci. These included 84 random amplified polymorphic DNA (RAPD) markers, 32 Sequence-Tagged RAPD (STAR) markers, 5 cDNAs, and 5 microsatellites in 232 F1 intercross progeny from a single, field-collected P1 female. A preliminary linkage map of 616 cM was generated across 14 linkage groups with one marker every 10.8 cM. Assuming a genome size of ~ 10 9 bp, the relationship of physical to genetic distance was found to be ~ 300 kb/cM in the I. scapularis genome.

Interferon-γ (IFN-γ) contributes to host resistance during acute infection with Trypanosoma cruzi, the causative agent of Chagas’ disease. Inducibly expressed guanosine triphosphatase (IGTP), a 48-kDa guanosine triphosphatase (GTPase), is a member of a family of GTPase proteins inducibly expressed by IFN-γ. The expression pattern of IGTP suggests that it may mediate IFN-γ–induced responses in a variety of cell types. IGTP has been demonstrated to be important for control of Toxoplasma gondii infection but not for resistance against Listeria monocytogenes. Six commercially available bacterial identification products were tested with Vibrio alginolyticus (12 strains), V. cholerae (30 strains), Photobacterium (Vibrio) damselae (10 strains), V. fluvialis (10 strains), V. furnissii (4 strains), V. hollisae (10 strains), V. metschnikovii (9 strains), V. mimicus (10 strains), V. parahaemolyticus (30 strains), and V. vulnificus (10 strains) to determine the accuracy of each system for identification. The products included API 20E, Crystal E/NF, MicroScan Neg ID2 and Rapid Neg ID3, and Vitek GNI+ and ID-GNB. Each product was tested only with those species that were listed in its database. Hepatic fibrois results from excess extracellular matrix produced primarily by hepatic stellate cells (HSC). In response to injury, HSC differentiate to a myofibroblastic phenotype expressing smooth muscle actin and fibrillar collagens. Relaxin is a polypeptide hormone shown to have antifibrotic effects in fibrosis models. In this study, activated HSC from rat liver were treated with relaxin to determine if relaxin can reverse markers of HSC activation. Relaxin treatment resulted in a decrease in the expression of smooth muscle actin, but had no effect on cell proliferation rate. The mannose 6-phosphatelinsulin-like growth factor I1 receptor (M6P/IGF-IIR) is a protein that facilitates the transport of acid hydrolases into the lysosome. We have shown that chronic ethanol consumption lowers the M6P/IGF-IIR content in rat hepatocytes. Here, we determined the steady-state level of mRNA encoding M6P/IGF-IIR, as well as the rate of receptor synthesis, to ascertain whether the ethanol-elicited reduction in receptor protein content is related to changes in either or both of these parameters. Rats were pair-fed the normal carbohydrate (NC) or low carbohydrate high-fat (LC) liquid diets containing either ethanol or isocaloric maltose-dextrin for 7-8 weeks. RNA was isolated …

During receptor-mediated endocytosis (RME), extracellular molecules are internalized after being recognized and bound to specific cell surface receptors. In previous studies of the asialoglycoprotein receptor (ASGPR) in rats, we showed that ethanol impairs RME at multiple ASGPR sites. Ethanol administration has been shown to increase apoptosis, and we demonstrated increased sensitization to apoptotic induction in hepatocytes from ethanol-fed rats. Although a physiological role for the ASGPR has not been identified, investigators have shown its involvement in the uptake/clearance of apoptotic cells in vitvo. This suggests a potential role for the ASGPR in the removal of apoptotic cells, and the recent …

We tested the influence of IFNγ on proteasome activity in parental Hep G2 cells that do not metabolize ethanol, as well as in recombinant Hep G2-derived cells that express either or both alcohol dehydrogenase (ADH) and cytochrome P4502E1 (CYP2E1). IFNγ treatment increased proteasome activity in VL-17A (ADH+, CYP2E1+) and E-47 (CYP2E1+) cells, but not in Hep G2, VI-R2 (parental cells with empty vectors) or in VA-13 (ADH+) cells. Mental health services are essential and need to be equally accessible and non-discriminatory to all individuals within our society. Traditionally, the Deaf community has encountered many hindrances when attempting to access mental health seruices. This study examines the systemic barriers that prevent Deaf individuals from obtaining mental health services in the Twin Cities Metro and regional area. Questionnaires were sent out to 100 individuals that serve Deaf adults with mental illness within the Twin Cities Metro Area. Aggregation properties of meso-tetratolylporphyrins bearing cationic substituents of a lipophilic nature such as ammonium, pyridinium, phosphonium, sulfonium, and isothiouronium were studied by UV/vis and resonance light scattering spectroscopy. The exciton point-dipole approximation was used to predict the structural alignment of the porphyrin units within the aggregate. The contribution of various types of aggregates depends on the porphyrin substitution, ionic strength, and temperature of aqueous solution of the porphyrin. In general, the preferred structure of an aggregate formed by temperature-induced aggregation is the J-aggregate exhibiting the characteristically narrow, red-shifted Soret band indicating a parallel "side-by-side" arrangement of the porphyrin units. A peptide-based electron-transfer system has been designed in which the specific positions of redox-active metal complexes appended to either an a-helix, or an a-helical coiled-coil, can be reversed to test the effect of the helix dipole in controlling photoinduced electron-transfer rates. Two 30-residue apopeptides were prepared having the following sequences: (1) Ac-K-(IEALEGK)(ICALEGK)(IEALEHK)-(IEALEGK)-G-amide, and (II) Ac-K-(IEALEGK)(IHALEGK)-(IEALECK)(IEALEGK)-G-amide. Each apopeptide was reacted first with [Ru(bpy)(2)(phen-ClAc)](2+), where bpy = 2,2'-bipyridine and phen-ClAc = 5-chloroacetamido-1,10-phenanthroline, to attach the ruthenium polypyridyl center to the cysteine side-chain of the polypeptide. The isolated products were then reacted with [Ru(NH3)(5)(H2O)](2+) to yield the binuclear electron-transfer metallopeptides ET-I and ET-II. …

Heads of hunter-harvested deer and elk were collected throughout South Dakota (USA) and within established chronic wasting disease (CWD) surveillance areas from 1997–2002 to determine infection with CWD and bovine tuberculosis (TB). We used immunohistochemistry to detect CWD-infected individuals among 1,672 deer and elk sampled via geographically targeted surveillance. A total of 537 elk (Cervus elaphus nelsoni), 813 white-tailed deer (Odocoileus virginianus), and 322 mule deer (O. hemionus) was sampled for CWD. Estimated overall prevalence and associated confidence intervals (95%) in white-tailed deer was 0.001% (0–0.007%). Similarly, estimated overall prevalence in elk and mule …

Mycobacterium infections occur in numerous wildlife species worldwide and this bacterial genus can also cause disease in humans and livestock. Two Mycobacterium species and one subspecies that infect wildlife are important from a wildlife damage management perspective. Mycobacterium avium, avian tuberculosis, is a ubiquitous and cosmopolitan disease of wild and domestic birds. Wild and domestic ruminants are host to M. avium paratuberculosis, also known as Johne’s disease. Finally, Mycobaterium bovis, bovine tuberculosis, is primarily associated with domestic and wild bovines and ungulates, but also infects many other species of mammals. While crossing of specialized sire and dam breeds can be biologically efficient overall, a by-product is produced in the form of (crossbred) males of the dam line that are unwanted for breeding. In the UK sheep indusiru, most commercial females are first crosses between males of 'longwool' crossing sire breeds, predominantly the Biuefaced Leicester, and females of hill breeds. We present several algorithms for identifying thioredoxin (Trx)-fold proteins containing a conserved CxxC motif (two cysteines separated by two residues). The low conservation of primary sequence in this protein superfamily makes conventional methods difficult to use. Therefore, we use structural properties to build our classifiers. These structural properties include secondary structure patterns as well as various properties of the residues in the protein sequences. We use this information to model Trx-fold proteins via hidden Markov models, decision trees, and algorithms in the multipleinstance learning model. We describe a generalization of the multiple-instance learning model in which a bag’s label is not based on a single instance’s proximity to a single target point. Rather, a bag is positive if and only if it contains a collection of instances, each near one of a set of target points. We list potential applications of this model (robot vision, content-based image retrieval, protein sequence identification, and drug discovery) and describe target concepts for these applications that cannot be represented in the conventional multiple-instance learning model. We then adapt a learning-theoretic algorithm for learning in this model and present empirical …
