Eukaryotic selenocysteine (Sec) protein insertion machinery was thought to be restricted to animals, but the occurrence of both Sec-containing proteins and the Sec insertion system was recently found in Chlamydomonas reinhardtii, a member of the plant kingdom. Herein, we used RT-PCR to determine the sequence of C. reinhardtii Sec tRNA ^[Ser]Sec, the first non-animal eukaryotic Sec tRNA ^[Ser]Sec sequence. Like its animal counterpart, it is 90 nucleotides in length, is aminoacylated with serine by seryl-tRNA synthetase, and decodes specifically UGA. Evolutionary analyses of known Sec tRNAs identify the C. reinhardtii form as the most diverged eukaryotic Sec tRNA …

This exploratory, cross-sectional quantitative study was undertaken to examine how County Child Protection Social Workers perceive the concept of child resilience and how social worker self appraisal of use of resilience in interventions was consistent with their rating of resilience of children in their professional career. A self report survey was distributed to County Child Protection Field Social Workers. Univariate analysis was done and descriptive statistics were used to summarize characteristics of the data. Due to low response rate and design of the survey, findings could not be generalized. Future research may use this information to further explore "best practices" …

In vivo-induced antigen technology is a method to identify proteins expressed by pathogenic bacteria during human infection. Sera from 10 patients convalescing from cholera infection in Bangladesh were pooled, adsorbed against in vitro-grown El Tor Vibrio cholerae O1, and used to probe a genomic expression library in Escherichia coli constructed from El Tor V. cholerae O1 strain N16961. We identified 38 positive clones in the screen, encoding pili (PilA and TcpA), cell membrane proteins (PilQ, MshO, MshP, and CapK), methyl-accepting chemotaxis proteins, chemotaxis and motility proteins (CheA and CheR), a quorum-sensing protein (LuxP), and four hypothetical proteins. A synthetic triterpenoid, 2-cyano-3,12-dioxoolean-1,9-dien-28-oic acid (CDDO), has been reported to have anti-inflammatory properties and to decrease the interleukin-1 (IL-1)-induced expression of matrix metalloproteinase-1 (MMP-1) and MMP-13. We have shown previously that IL-1 induces expression of the inhibitor of NF-κB (IκB) family member Bcl-3, and that this contributes to MMP-1 expression. To quantify the effects of CDDO on IL-1-induced MMP-1, MMP-13 and Bcl-3 expression, we stimulated the chondrosarcoma cell line SW-1353 and human primary chondrocytes with IL-1, in the presence or absence of CDDO. We studied perceptions parental differential treatment as reported by parents and children in two different settings. Perceptions of differential affection and control were examined in healthy families and in families that include a child diagnosed with Type 1 diabetes. Parental differential treatment was assessed using questionnaires that measured perceptions of absolute parenting for children and their siblings. Difference scores were subsequently utilized to generate perceived parental differential treatment scores. Participants were 61 parents (half with healthy children, half with one child who has diabetes) and 62 children (half comprising sibling pairs unaffected by any medical problems, half including one child …

In the northern Great Plains of the United States, conflicts between red-winged blackbirds (Agelaius phoeniceus) and sunflower (Helianthus annuus) growers have intensified since the late 1960s due to the expanded commercial production of sunflowers. We studied the potential population effects of the removal of up to 2 million red-winged blackbirds annually under a 5-year program of baiting during spring with DRC-1339 (3-choloro-4-methalalanine)-treated rice. We also examined whether lethal control, in combination with current levels of breeding-habitat management, would be cost-effective in decreasing depredation of sunflower crops during late summer. The purpose of this pilot project was to increase the effectiveness of the three Conservation Commissions in the Exeter River watershed. The Towns of Kensington, Kingston, and Sandown were selected based on their interest in the program and their need for professional planning assistance. Rockingham Planning Commission staff attended monthly meetings of each Conservation Commission to discuss and implement short-term and long-term projects. The three Conservation Commissions selected two natural resource protection projects to work on with RPC assistance. The scope of these projects ranged from very basic, such as establishing a process for record keeping in Kingston, to more …

Recently, the Board of Directors of the RLT voted to expand the organization’s service region to include all the communities in Rockingham County. This action was taken due to an increased demand for the Trust’s services by municipalities and landowners.

The Taxonomic Report of the International Lepidoptera Survey

Hesperia meskei pinocayo Gatrelle and Minno is described from Big Pine Key, Monroe County, Florida, United States. This subspecies is thought to be nearly extinct. Its mainland status is uncertain. It is characterized by bi-colored ventral hindwings with more prominent ventral hindwing spots. Hesperia metea intermedia Gatrelle is described from Pickens County, South Carolina, United States. Its range is from northwestern South Carolina, United States west through Mississippi, United States and probably into Missouri, United States. Males have the least dorsal fulvous of the metea subspecies; ventrally they are well marked. Females are similar to subspecies licinus. Hesperia sassacus …

In vitro formation of Hydrogenobacter thermophilus cytochrome c 552 has previously been demonstrated (Daltrop, O., Allen, J. W. A., Willis, A. C., and Ferguson, S. J. (2002) Proc. Natl. Acad. Sci. U. S. A. 99, 7872-7876). Now we report that the single cysteine variants of H. thermophilus c552, which bind heme via a single thioether bond, also form in vitro. Furthermore, reaction of the apocytochromes containing either AXXCH or CXXAH in the binding motif with 2-vinyldeuteroheme and 4-vinyldeuteroheme resulted predominantly in covalent attachment between Cys-11 and the 2-vinyl moiety and Cys-14 and the 4-vinyl functionality. This study tested the use of physical manipulatives during science instruction – in conjunction with the traditional, textbook-oriented approach – and determines whether manipulatives create more significant learning experiences than text generates alone. The subject sample consisted of 20 third-grade students from a suburban New Jersey elementary school classroom. All subjects functioned at least on a third-grade level in all subjects with the exception of three students, who participated in the Talented and Gifted Program. Subjects from the experimental group and the control group completed a science content pretest of thirteen open-ended cognition questions. Both groups completed assigned activities, and …

The purpose of this study was to evaluate the performance of fourth grade students working in cooperative learning groups. Students worked in three cooperative learning groups in order to determine whether grouping students heterogeneously into cooperative learning groups based on individual students' learning pattern would have a positive correlation with group and individual success. First students were grouped randomly and then homogeneously by learning pattern, and lastly heterogeneously by learning pattern. The purpose of this action research project was to determine if: (a) writing in student journals affected the grades of my students, and (b) if student attitudes were affected by writing in journals and receiving teacher feedback. The research was conducted during the third semester at Triton Regional High School with a total of 49 tenth-grade students enrolled in two Laboratory Biology classes. Students in one class wrote in journals about teacher-posed topics for five minutes on average of once per week. Natural Resources Management and Environmental Sciences

Landscapes held in public trust are difficult to manage; a living National Monument subject to increased public use presents additional challenges. These landscapes are subject to regional and global effects outside the manager's control, such as global climate change and air pollution. They are also subject to innumerable local variables, most of whose effects are difficult to predict. The giant sequoia groves and their surrounding ecosystem are fiercely loved by many; although the goals may be broadly similar, the objectives and policies of these giant sequoia lovers are often sharply at odds.
