Pre-Conference Proceedings Of The Focus Symposium On Collaborative Decision-Support Systems, Collaborative Agent Design Research Center
Collaborative Agent Design (CAD) Research Center
The papers included in these pre-conference proceedings reflect the increasing focus on the utilization of computers as collaborating, decision-assistance partners in complex and often time-critical problem situations. There are high expectations that intelligent software agents will solve many of our current information system woes, such as lack of interoperability, multiple failure points, vulnerability to intrusion, making the right information available to the right person at the right time, and proposing solutions under time-critical conditions. Software agents do not have magical human-like capabilities. It is not possible to simply develop a piece of software code that is capable of reasoning about …

The Emerging Knowledge Management Paradigm: Some Organizational And Technical Issues, Jens G. Pohl
Collaborative Agent Design (CAD) Research Center
This paper addresses the expectations, organizational implications, and information processing requirements, of the emerging knowledge management paradigm. A brief discussion of the enablement of the individual through the wide-spread availability of computer and communication facilities, is followed by a description of the structural evolution of organizations, and the architecture of a computer-based knowledge management system. The author discusses two trends that are driven by the treatment of information and knowledge as a commodity: increased concern for the management and exploitation of knowledge within organizations; and, the creation of an organizational environment that facilitates the acquisition, sharing and application of knowledge. …
Surface Salt Bridges Modulate Dna Wrapping By The Type Ii Dna-Binding Protein Tf1, Anne Grove
Surface Salt Bridges Modulate Dna Wrapping By The Type Ii Dna-Binding Protein Tf1, Anne Grove
Faculty Publications
The histone-like protein HU is involved in compaction of the bacterial genome. Up to 37 bp of DNA may be wrapped about some HU homologues in a process that has been proposed to depend on a linked disruption of surface salt bridges that liberates cationic side chains for interaction with the DNA. Despite significant sequence conservation between HU homologues, binding sites from 9 to 37 bp have been reported. TF1, an HU homologue that is encoded by Bacillus subtilis bacteriophage SPO1, has nM affinity for 37 bp preferred sites in DNA with 5-hydroxymethyluracil (hmU) in place of thymine. On the …

Formulation Of Solid Body Wash From Dragon Fruit Peel Waste With Pandan Leaf Extract, Anggun Puspitarini Siswanto, Georgius Aldo
ASEAN Journal of Community Engagement
This paper highlights the formulation of dragon fruit waste material for a solid body wash, which arguably contain better substance for skin health. The result of this experiment opens up a possibility to be applied as part of community project, contributing to sustainable environment and healthier lifestyle. Red dragon fruit (Hylocereus polyrhizus) is a plant originating from a dry tropical climate with a fairly thick peel that accounts for 30%–35% of the fruit’s total weight. The peel is rarely used or reprocessed and thus often becomes waste. The peel’s polyphenol and other important substances component makes it rich …

[ 1] The well-known capability of TOPEX/Poseidon altimetry to map sea levels precisely in the deep oceans motivates its application to the topographically complex Coral Sea and NE Australian continental margin. We assess several global tidal models for correcting TOPEX altimetry in the Coral Sea and find CSR3.0 offers good overall performance, based on comparisons of model-predicted and tide gauge harmonic constituents. Using CSR3.0 tidal corrections, we evaluate residual Sea Surface Height (SSH) Root Mean Square (RMS) variability and residual M2 tidal alias errors. Away from large reefs and islands, CSR3.0 amplitude and phase errors for M2 are typically less …

Functions Of The Activation Loop In Csk Protein-Tyrosine Kinase, Xiaofeng Lin, Sungsoo Lee, Gongqin Sun
Cell and Molecular Biology Faculty Publications
Autophosphorylation in the activation loop is a common mechanism regulating the activities of protein-tyrosine kinases (PTKs). PTKs in the Csk family, Csk and Chk, are rare exceptions for lacking Tyr residues in this loop. We probed the function of this loop in Csk by extensive site-specific mutagenesis and kinetic studies using physiological and artificial substrates. These studies led to several surprising conclusions. First, specific residues in Csk activation loop had little discernable functions in phosphorylation of its physiological substrate Src, as Ala scanning and loop replacement mutations decreased Csk activity toward Src less than 40%. Second, some activation loop mutants, …

Amino Acids Glu323, Tyr324, Glu330, And Val331 Of Factor Va Heavy Chain Are Essential For Expression Of Cofactor Activity, Lisam S. Singh, Michael A. Bukys, Daniel O. Beck, Michael Kalafatis
Chemistry Faculty Publications
We have recently demonstrated that amino acid region 323-331 of factor Va heavy chain (9 amino acids, AP4') contains a binding site for factor Xa (Kalafatis, M., and Beck, D. O. (2002) Biochemistry 41, 12715-12728). To ascertain which amino acids within this region are important for the effector and receptor properties of the cofactor with respect to factor Xa, we have synthesized three overlapping peptides (5 amino acids each) spanning the amino acid region 323-331 and tested them for their effect on prothrombinase complex assembly and function. Peptide containing amino acids 323EYFIA327 alone was found to increase the catalytic efficiency …

The Galapagos Giant Tortoise (Geochelone Elephantopus) And The Spread Of Invasive Plants, Anne D. Schultz
Biology ETDs
Island ecosystems are commonly thought to be more vulnerable to the effects of invasive species than mainland systems. In recent decades, the number of exotic plants known to be present in the Galapagos archipelago has increased exponentially. Several of these plants are aggressive invaders that pose a threat to the biodiversity of the islands. Two of these plant species, Passiflora edulis Sims and Psidium guajava L. were introduced to Santa Cruz Island during the 20th century. Both pose a threat to the floristic diversity of the National Park land on Santa Cruz Island.

University of Nebraska State Museum: Entomology Papers
We describe a new genus of anomaline scarab, Anomalorhina (Coleoptera: Scarabaeidae: Rutelinae: Anomalini) from Costa Rica and Nicaragua. Anomala turrialbana Ohaus is transferred into this genus, and a new species, Anomalorhina osaensis, is described. Anomalorhina is characterized by the clypeal apex that is abruptly reflexed, the base of the frons that has prominent tubercles in males, the pronotum with a depressed fovea in males, and the pronotum with the apical bead posteriorly produced and V-shaped at the middle in females. We summarize the biological importance of the 16 genera of New World anomaline scarabs, including comments on agricultural pests, …

Spatiotemporal Multicast And Partitionable Group Membership Service, Qingfeng Huang
Spatiotemporal Multicast And Partitionable Group Membership Service, Qingfeng Huang
All Computer Science and Engineering Research
The recent advent of wireless mobile ad hoc networks and sensor networks creates many opportunities and challenges. This thesis explores some of them. In light of new application requirements in such environments, it proposes a new multicast paradigm called spatiotemporal multicast for supporting ad hoc network applications which require both spatial and temporal coordination. With a focus on a special case of spatiotemporal multicast, called mobicast, this work proposes several novel protocols and analyzes their performances. This dissertation also investigates implications of mobility on the classical group membership problem in distributed computing, proposes a new specification for a partitionable group …

Characterization Of Mutations And Loss Of Heterozygosity Of P53 And K-Ras2 In Pancreatic Cancer Cell Lines By Immobilized Polymerase Chain Reaction, James Butz, Eric Wickstrom, Jeremy Edwards
Department of Biochemistry and Molecular Biology Faculty Papers
Background
The identification of known mutations in a cell population is important for clinical applications and basic cancer research. In this work an immobilized form of the polymerase chain reaction, referred to as polony technology, was used to detect mutations as well as gene deletions, resulting in loss of heterozygosity (LOH), in cancer cell lines. Specifically, the mutational hotspots in p53, namely codons 175, 245, 248, 249, 273, and 282, and K-ras2, codons 12, 13 and 61, were genotyped in the pancreatic cell line, Panc-1. In addition LOH analysis was also performed for these same two genes in Panc-1 by …

Functional And Comparative Genomic Analyses Of An Operon Involved In Fructooligosaccharide Utilization By Lactobacillus Acidophilus, Rodolphe Barrangou, Eric Altermann, Robert Hutkins, Raul J. Cano, Todd Klaenhammer
Biological Sciences
Lactobacillus acidophilus is a probiotic organism that displays the ability to use prebiotic compounds such as fructooligosaccharides (FOS), which stimulate the growth of beneficial commensals in the gastrointestinal tract. However, little is known about the mechanisms and genes involved in FOS utilization by Lactobacillus species. Analysis of the L. acidophilus NCFM genome revealed an msm locus composed of a transcriptional regulator of the LacI family, a four-component ATP-binding cassette (ABC) transport system, a fructosidase, and a sucrose phosphorylase. Transcriptional analysis of this operon demonstrated that gene expression was induced by sucrose and FOS but not by glucose or fructose, …

The Structures Of Fluorene– (H2o)1,2 Determined By Rotational Coherence Spectroscopy, David M. Laman, Alan G. Joly, Douglas Ray
All Faculty Scholarship for the College of the Sciences
Rotational coherence spectroscopy ~RCS!, via time-correlated single photon counting, and two-color resonant two-photon ionization ~R2PI! time-of-flight mass spectrometry, have been used to characterize fluorene–(water)1,2[FL– (H2O)1,2] van der Waals clusters generated in supersonic jets. Rotational coherence traces have been obtained at excitation energies corresponding to several resonant features in the S1<-S0 R2PI spectra of FL– (H2O)1,2 . RCS simulations and diagonalization of the moment of inertia tensor have been used to obtain S1 excited state rotational constants and structures of FL– (H2O)1,2 that are consistent with …-s

Short Pulse Laser Propagation Through Tissues, Champak Das, Ashish Trivedi, Kunal Mitra, Tuan Vo-Dinh
Short Pulse Laser Propagation Through Tissues, Champak Das, Ashish Trivedi, Kunal Mitra, Tuan Vo-Dinh
Biomedical Engineering and Sciences Faculty Publications
An experimental and numerical study is performed to analyze short pulse laser propagation through tissue phantoms without and with inhomogeneities/tumors imbedded in it. Short pulse laser probing techniques has distinct advantages over conventional very large pulse width or cw lasers primarily due to the additional information conveyed about the tissue interior by the temporal variation of the observed signal. Both the scattered temporal transmitted and reflected optical signals are measured experimentally using a streak camera for samples irradiated with a short pulse laser source. Parametric study involving different scattering and absorption coefficients of tissue phantoms and inhomogeneities as well as …
