This thesis describes the design, optimisation and shipboard deployment of a flow injection - chemiluminescence (FI-CL) technique for the determination of labile Cu(II) and total copper (by UV irradiation) in seawater. The operational parameters of the FI manifold in a UHP water sample matrix and the 1,10-phenanthroline CL reaction were rigorously optimised. Interferences to the CL reaction were investigated and the good analytical figures of merit obtained presented. Highly branched isoprenoid (HBI) alkenes are ubiquitous lipids which have been identified in numerous geochemical samples, ranging from recent sediments to ancient oils. At the outset of the current investigation, the diatomaceous algae Haslea ostrearia (C25 alkenes) and Rhizosolenia setigera (C25 or C30 alkenes) were the only reported biological sources of these compounds. However, there remained a poor correlation between isomers found in diatoms and those commonly reported in sediments and water column particles. This paper presents a cross-sectional study testing whether dolphins that are born in aquarium pools where they hear trainers’ whistles develop whistles that are less frequency modulated than those of wild dolphins. Ten pairs of captive and wild dolphins were matched for age and sex. Twenty whistles were sampled from each dolphin. Several traditional acoustic features (total duration, duration minus any silent periods, etc.) were measured for each whistle, in addition to newly defined flatness parameters: total flatness ratio (percentage of whistle scored as unmodulated), and contiguous flatness ratio (duration of longest flat segment divided by total duration). The author in this piece reflects on the death penalty in the U.S. in the aftermath of the 9/11 terrorist attacks. The writer goes on to argue that capital punishment is, in and of itself, a form of violence. Also discussed in the article are the gradual removal of executions from public view, issues of deterrence and violent crime, and the author's preference for life-without-possibility-of-parole sentences.

Multibeam volume acoustic backscatterimagery and reverberation measurements are derived from data collected in 200-m-deep waters in the northeastern Gulf of Mexico, with the Toroidal Volume Search Sonar (TVSS), a 68-kHz cylindrical sonar operated by the U.S. Navy’s Coastal System Station. The TVSS’s 360-degree vertical imaging plane allows simultaneous identification of multiple volume scattering sources and their discrimination from backscatter at the sea surface or the seafloor. This imaging capability is used to construct a three-dimensional representation of a pelagic fish school near the bottom. Scattering layers imaged in the mixed layer and upper thermocline are attributed to assemblages of epipelagic …

During March/April 1998 we conducted detailed mapping and sampling of hydrothermal plumes along six segments of Earth's fasting spreading mid-ocean ridge, 27.5°–32.3°S on the East Pacific Rise. We compared the distribution and chemistry of hydrothermal plumes to geological indicators of long-term (spreading rate) and moderate-term (ridge inflation) variations in magmatic budget. In this large-offset, propagating rift setting, these geological indices span virtually the entire range found along fast spreading ridges worldwide. Hydrothermal plumes overlaid ∼60% of the length of superfast (>130 km/Myr) spreading axis surveyed and defined at least 14 separate vent fields. We observed no plumes over the …

This thesis identifies systematically the domains of positive consequences of illness and details the development of a measure of the positive consequences of illness- the Silver Lining Questionnaire (SLQ). Further aims of the thesis include the use of the SLQ as a predictor of health outcome and an investigation of the prevalence and correlates of positivity in illness. The first study questioned 55 people about their positive experiences of illness and uncovered 17 themes. In this study, the mRNA and protein levels of hypoxia inducible factor 1 (HIF-1α), and a number of genes regulated by hypoxia (VEGF, GLUT-1, p53), were determined in four breast carcinoma cell lines, peripheral blood mononuclear cells (PBMCs) of patients with breast cancer and Type 1 diabetes (TIDM), and in human breast and brain tumour tissue. Breast carcinoma cells and PMBCs from both patients and normal controls were exposed to hypoxia (≤1% 0 2) and/or high glucose. There is increasing evidence to suggest that genetic factors are involved in the pathogenesis of microvascular complications in diabetes mellitus. Recent studies have suggested that genetic variations in the aldose reductase (ALR2) gene may contribute to the genetic susceptibility to microvascular complications. Aldose reductase is the first and rate-limiting enzyme of the polyol pathway and is implicated in the pathogenesis of diabetic microvascular disease (nephropathy, retinopathy and neuropathy). This thesis presents an investigation into the responses to extracts of Ascophyllum nodosum (Maxicrop seaweed extracts - MSE) of two different plants species - wheat and strawberry, and their interactions with two soil-borne pathogens, Gaeumannomyces graminis and Phytophthora fragariae respectively, under various environmental conditions. The responses to MSE using hydroponic, glasshouse and field experiments showed that levels of Take-all infection in wheat were reduced by some of the treatments applied. Repeat experiments showed that consistency of results was poor but a positive trend for disease suppression followed MSE treatments. Studies of strawberry infection by Phytophthora fragariae revealed a significantly reduced …

The purpose of this study was to investigate several biological markers that may predict the response of muscle invasive transitional cell carcinoma TCC of the bladder to radical radiotherapy. The specific markers chosen were tumour angiogenesis (CD31 &CD34), tumour cell proliferation (Ki-67) and apoptosis (bcl-2), intratumour macrophage infiltration (CD68) and p53. Archival formalin fixed paraffin embedded pre treatment bladder biopsies from 101 patients with muscle invasive TCC were obtained. All patients subsequently received radical radiotherapy as their only treatment for the disease. 4µm sections were graded according to the WHO grading system and staged by the TNM classification. Female Maxwell's duikers (Cephalophus maxwellii) were studied at the Wildlife Survival Center on St. Catherine's Island, Georgia. The objectives were to quantify the effects of housing on activity budgets and progesterone cycling. Four animals were studied across two housing situations: housed individually versus housed in a grouped setting. Behavioral observations revealed no significant change in access to resources, rumination, or repetitive behaviors, once animals were housed together. Fecal samples were collected in order to determine levels of progesterone through radioimmunoassay (RIA). Fluctuations in progesterone levels revealed no clear estrous cycles. Animals showed less variability in progesterone levels and …

This thesis documents the development and application of the Automated Benthic Image Scaling System (ABISS), a novel structured lighting array for calculating image scale, accounting for perspective, to allow quantitative non-destructive megafaunal sampling using observations from a Remotely Operated Vehicle (ROV). Megafauna are important components of marine soft sediment assemblages that influence the composition of the associated assemblage and the flux of energy across the sediment-water interface, by altering the physical and chemical characteristics of the sediment during bioturbation. One hundred years ago, Giemsa's stain was employed for the first time for malaria diagnosis. Giemsa staining continues to be the method of choice in most malarious countries, although, in the recent past, several alternatives have been developed that exhibit some advantages. Considerable progress has been made with fluorescent dyes, particularly with Acridine Orange (AO). The literature on the discovery, development and validation of the AO method for malaria diagnosis is reviewed here. Compared with conventional Giemsa staining, AO shows a good diagnostic performance, with sensitivities of 81.3%-100% and specificities of 86.4%-100%. The Nature Preserve at the Clark County Wetlands Park is a constructed wetland that has the potential to produce new mosquito habitat. This thesis evaluates the potential for the development of a mosquito problem at the newly constructed wetland system by assessing the population dynamics of mosquitoes within the Nature Preserve. Based on data collected from May 2001 through January 2002, I describe fluctuations in mosquito genus and relative abundance during the first summer through winter period of wetland development.

Fucus serratus was used in a series of laboratory experiments to study the effects of Cu2+ exposure on the early development of zygotes and physiological effects on adults collected from Cu2+ polluted and clean habitats. Comparative studies showed that Cu2+ tolerance in Fucus is an inherited character. Zygote development and growth of embryos and adults during Cu2+ exposure indicated that Fucus serratus from a Cu2+ polluted habitat had a higher tolerance limit than material from two clean habitats. Furthermore, there was no difference in the tolerance level of zygotes, embryos and adults from the same habitat. Chlorophyll fluorescence measurements showed …

Studies were undertaken to examine the extent of molecular variation among isolates of the fish pathogen Renibacterium salmoninarum. As many isolates as possible were gathered from diagnostic laboratories within the UK, and checked for viability and contamination. The isolates were derived mainly from infected rainbow trout and Atlantic salmon that were farmed in England, Scotland and Wales, subject to the requirements of statutory legislation. Incomplete histories were available for the sources of the isolates, which spanned a time scale of 36 years, from 1962-1998. This study used content analysis to examine contemporary young adult fiction to see how the grandparent-grandchild relationship was portrayed. The titles were randomly chosen books taken from the American Library Association Best Books for Young Adults annual lists 1990-2000. Simple perceptions of grandparents as older adults who see their grandchildren on special occasions are no longer realistic. There is statistical evidence that many grandparents have a significant effect on the lives of young adults. Grandparents are becoming more active in the family household.
