Pbp 5: Characterization Of An Escherichia Coli Morphogene., David Emmet Nelson
Theses and Dissertations
The penicillin binding proteins (PBPs) are a group o f enzymes responsible for the latter stages o f peptidoglycan synthesis. Our major research objective in this study was to determine the physiological roles o f the low molecular weight (LMW) PBPs of the Gram negative bacterium Escherichia coli (E. coli). During extensive screening of a set o f mutant strains that lack different combinations of LMW PBP genes we determined that PBP 5 played a role in maintaining E. co li’s normal rod shape. However, loss of PBP 5 did not cause severe morphological alterations unless the cells were also …

In oviparous vertebrates, maternal contributions to yolk material in the form of steroid hormones are thought to be responsible for several growth and behavior effects in the offspring. We initiated a series of experiments to determine the dynamics of yolk hormones in two species with temperature-dependent sex determination (TSD ), the snapping turtle, Chelydra serpentina, and painted turtle,Chrysemys picta, and one species with genetic sex determination (GSD), the Leghorn chicken,Gallus domesticus. Suppressors Of Transcriptional Transgenic Silencing In Chlamydomonas Are Sensitive To Dna-Damaging Agents And Reactivate Transposable Elements, Byeong-Ryool Jeong, Dancia Wu-Scharf, Chaomei Zhang, Heriberto D. Cerutti

In the unicellular green alga Chlamydomonas reinhardtii, the epigenetic silencing of transgenes occurs, as in land plants, at both the transcriptional and posttranscriptional levels. In the case of single-copy transgenes, transcriptional silencing takes place without detectable cytosine methylation of the introduced DNA. We have isolated two mutant strains, Mut-9 and Mut-11, that reactivate expression of a transcriptionally silenced single-copy transgene. These suppressors are deficient in the repression of a DNA transposon and a retrotransposon-like element. In addition, the mutants show enhanced sensitivity to DNA-damaging agents, particularly radiomimetic chemicals inducing DNA double- strand breaks. All of these phenotypes are much …

Synchrotron Radiation-Induced X-Ray Emission To Identify Metal Ions In Preparations Of Purified Protein, Thomas Pfister, Huan Feng, Eckard Wimmer, Keith W. Jones

The suitability of synchrotron radiation-induced X-ray emission (SRIXE) for the detection and identification of metal ions in preparations of purified, soluble proteins was tested. Glutathione S-transferase fused to the proximal zinc finger motif of human transcription factor IIIA or to the cysteine-rich motif of poliovirus protein 2C was expressed in bacteria and purified by affinity chromatography. Aqueous samples containing the purified proteins were analyzed with SRIXE, and trace amounts of zinc and iron were detected. Mutation of the zinc-coordinating residues in the cysteine-rich motif of poliovirus protein 2C resulted in the loss of the zinc-binding ability. Relative quantities of metal …
