A Wd40-Repeat Containing Protein, Similar To A Fungal Co-Repressor, Is Required For Transcriptional Gene Silencing In Chlamydomonas, Chaomei Zhang, Dancia Wu-Scharf, Byeong-Ryool Jeong, Heriberto D. Cerutti

In higher plants, mammals, and filamentous fungi, transcriptional gene silencing is frequently associated with DNA methylation. However, recent evidence suggests that certain transgenes can be inactivated by a methylation independent mechanism. In the unicellular green alga Chlamydomonas reinhardtii, single-copy transgenes are transcriptionally silenced without discernible cytosine methylation of the introduced DNA. We have isolated a Chlamydomonas gene, Mut11, which is required for the transcriptional repression of single-copy transgenes. Mut11 appears to have a global role in gene regulation since it also affects transposon mobilization, cellular growth, and sensitivity to DNA damaging agents. The cultural landscape is shaped by human imprints on the natural environment. Many decisions are made that have rippling effects over time. One type of development that changes the landscape is transportation innovation. As improved transportation links are introduced, people change their movement to include the quicker, more efficient route. As movement changes due to increased accessibility, roads that were once traveled by all now have fewer travelers. The rippling effects of the decision to add new infrastructure start at the opening of the innovation. New businesses will spring up at crossroads and common stopping areas. Neisseria gonorrhoeae and Neisseria meningitidis express an similar to43-kDa alpha-2,3-sialyltransferase (Lst) that sialylates the surface lipooligosaccharide (LOS) by using exogenous (in all N. gonorrhoeae strains and some N. meningitidis serogroups) or endogenous (in other N. meningitidis serogroups) sources of 5'-cytidinemonophospho-N-acetylneuraminic acid (CMP-NANA). Sialylation of LOS can protect N. gonorrhoeae and N. meningitidis from complement-mediated serum killing and from phagocytic killing by neutrophils. The precise subcellular location of Lst has not been determined. We confirm and extend previous studies by demonstrating that Lst is located in the outer membrane and is surface exposed in both N. gonorrhoeae and N. meningitidis. Western …

Genetic Evidence That Interhelical Packing Interactions In The Gp41 Core Are Critical For Transition Of The Human Immunodeficiency Virus Type 1 Envelope Glycoprotein To The Fusion-Active State, Kathryn E. Follis, Scott J. Larson, Min Lu, Jack H. Nunberg

The envelope glycoprotein complex (gp120-gp41) of human immunodeficiency virus type 1 (HIV-1) promotes the fusion of viral and cellular membranes through formation of the fusion-active six-helix bundle in the gp41 ectodomain. This gp41 core structure consists of three C-terminal helices packed in an antiparallel manner into hydrophobic grooves on the surface of the N-terminal trimeric coiled coil. Alanine mutations that destabilize the N- and C-terminal interhelical packing interactions also reduce viral infectivity. Here we show that viruses bearing these mutations exhibit a marked potentiation of inhibition by peptides that make up the gp41 core. By contrast, these viruses are unchanged …

There is increasing interest in the diving behavior of marine mammals. However, identifying foraging among recorded dives often requires several assumptions. The simultaneous acquisition of images of the prey encountered, together with records of diving behavior will allow researchers to more fully investigate the nature of subsurface behavior. We tested a novel digital camera linked to a time-depth recorder on Antarctic fur seals (Arctocephalus gazella). During the austral summer 2000-2001, this system was deployed on six lactating female fur seals at Bird Island, South Georgia, each for a single foraging trip. Black-tailed prairie dogs (Cynomys ludovicianus; hereafter, prairie dogs) are native to the short-grass prairie region of North America from Mexico to Canada (figure 1).According to government documents (64 Federal Register 57 at 14426–14427), before the 19th century expansion of the United States, prairie dogs inhabited millions of acres of the Great Plains and lived in huge colonies west of the Missouri River. Settlement of the Great Plains and the transformation of vast areas from native grassland to tilled farmland forever changed the prairie ecosystem and prairie dog habitat. This study was proposed to examine if there existed significant differences in voters' party identification and voting behavior due to gender, race, age, income, education and personal values.
The sample was chosen from eligible voters in Florida. Of the 1500 questionnaires managed, 161 were identified as useable. The sample was representative of the results of 2000 presidential election in Florida with respect to voters' gender, race, income, and voting choice by party identification. Statistical techniques used were ANOVAs, Chi-square statistics, factor analysis, and logistic regression.

The fungus Phytophthora infestans is a major ubiquitous pest in solanaceous crops and causes great damages in the agricultural systems in Nepal. This is especially true in the cash crop orientated areas of the Mid-Hills and the Terai region, where potato and tomato cropping is a prominent income source of the rural population. This fact in combination with cheap and readily available pesticides is responsible for the frequent and perceived over-use of chemical plant protection agents. The modular ecosystem modeling approach was used to create a flexible landscape model structure that is easy to modify and extend for particular case studies and applications. The Library of Hydro Ecological Modules (LHEM; http://iee.umces.edu/LHEM) includes modules that describe hydrologic processes, nutrient cycling, vegetation growth, decomposition, etc., both locally and spatially. LHEM is implemented within the framework of the Spatial Modeling Environment (SME; http://iee.umces.edu/SME3) that integrates modules and places local simulation models into a spatial context. The symptoms of coastal and estuarine eutrophication are the result of a number of interacting processes operating at multiple spatial and temporal scales. Thus, models developed to appropriately represent each of these processes are not easily combined into a single predictive model. We suggest that Bayesian networks provide a possible solution to this problem. The graphical structure explicitly represents cause-and-effect assumptions between system variables that may be obscured under other approaches. These assumptions allow the complex causal chain linking management actions to ecological consequences to be factored into an articulated sequence of conditional relationships. The control and management of a Wastewater Treatment Plant (WWTP) is a complex task that requires the supervision of human experts. To ease this task, Multiagent Systems provide a framework for a more efficiently control in environmental processes. A prototype of multiagent system has been designed for WWTP management. A model to characterize and predict continuous time series from machine learning techniques isproposed. This model includes the following three steps: dynamic discretization of continuous values,construction of probabilistic finite automata and prediction of new series with randomness. The first problemin most models from machine learning is that they are developed for discrete values; however, mostphenomena in nature are continuous. To convert these continuous values into discrete values a dynamicdiscretization method has been used. It is increasingly the case that models are being developed as “evolving” products rather than one-off application tools, such that auditable modelling versus ad hoc treatment of models becomes a pivotal issue. Auditable modelling is particularly vital to “parsimonious modelling” aimed at meeting specific modelling requirements. This paper outlines various contributory factors and aims to seed proactively a research topic by inextricably linking value/risk management to parsimonious modelling. In the framework of "active planetary cover" concept the influence of weathering on the atmosphere carbon and, as a consequence, on the climate is considered. The relatively slow process of chemical weathering, which is accompanied by an uptake of carbon from the atmosphere and its transportation to the intercontinental and epicontinental seas, is significantly amplified by terrestrial ecosystems. The mechanism of amplification is explained by an increase in CO2 concentration in the "soil" atmosphere caused by root respiration and the metabolism of micro-organisms, the so-called "respiration of soil". In this paper, prediction capability of a hybrid Artificial Neural Networks (ANN) was investigatedto solve the groundwater inverse problem. Initially, a Multi Layer Perceptron (MLP) network was developedand it was found that network produced better results when the target range of the parameters is smaller.Therefore, a Self-Organising Network (SON) was used to identify the objective subrange of the parameterand then the MLP model was employed to obtain final estimates. Salinity intrusion is a significant issue in the San Francisco Bay Delta because this estuarycontributes substantially to fresh water supply for much of northern California. Traditionally, salinity alongthe estuary has been measured using monitoring instruments installed at various locations within theSacramento-San Joaquin River Delta system, and these measurements are used to predict future conditionsand evaluate the water management alternatives. The California Department of Water Resources DeltaSimulation Model (DWRDSM) is used for this type of analysis. DWRDSM is an unsteady one-dimensionalhydrodynamic and salt transport model.
