The death domain (DD) of the protein kinase Pelle adopts a six-helix bundle fold in the crystal structure of the complex with its dimerization partner, Tube-DD. However, in crystals obtained from a solution of 45% 2-methyl-2,4-pentanediol (MPD), the C-terminal half of Pelle-DD folds into a single helix, and the N-terminal half of the molecule is disordered. The helical segment forms an antiparallel dimer with the corresponding helix of a symmetry-related molecule, and together they form extensive lattice interactions similar in number, composition, and buried surface to those in the six-helix bundle of the native fold. In 1993 Congress passed the North American Free Trade Agreement (NAFTA), with great anticipation at home and abroad that the newly formed regional alliance between Mexico, the United States, and Canada would increase productivity, reduce inefficiency, and strengthen the states' economies. However, the agreement was not met with universal enthusiasm. Among many of the rural poor, campesinos, working classes, racial minorities, and indigenous populations of all three states, NAFTA's passage signaled an unprecedented move toward globalization and mounting economic pressures (Mander and Goldsmith, 1996). One of the best ways to begin to understand the cultures of the Latina/o populations of the United States is to consider the term as it has been used and appropriated over time. The term "Latino" (Spanish Latino Americana), according to David Bushnell 0970: 3) was first used by the Colombian publicist Jose Maria Torres Caicedo in 1856 (Miguel Tinker Salas, personal communication, 2001). Knowledge of N and assimilate partitioning in wheat (Triticum aestivum L.) improves management efficacy and crop model development. Our purpose was to describe N and dry matter distribution during vegetative growth of blades, sheaths, and internodes on four culms. Winter wheat grown at the Colorado State University Horticultural Farm was sampled at Haun Stage 5 and jointing. Samples were dried, weighed, and analyzed for N. As the canopy developed and older tissue contributed more of total tissue, N concentration decreased although N mass and dry weight increased. Dry matter and N mass decreased from MC to T1 and T2 …

This review summarizes a collection of lactic acid bacteria that are now undergoing genomic sequencing and analysis. Summaries are presented on twenty different species, with each overview discussing the organisms fundamental and practical significance, environmental habitat, and its role in fermentation, bioprocessing, or probiotics. For those projects where genome sequence data were available by March 2002, summaries include a listing of key statistics and interesting genomic features. These efforts will revolutionize our molecular view of Gram-positive bacteria, as up to 15 genomes from the low GC content lactic acid bacteria are expected to be available in the public domain by …

BACKGROUND: The Blau syndrome (MIM 186580), an autosomal dominant granulomatous disease, was previously mapped to chromosome 16p12-q21. However, inconsistent physical maps of the region and consequently an unknown order of microsatellite markers, hampered us from further refining the genetic locus for the Blau syndrome. To address this problem, we constructed our own high-resolution physical map for the Blau susceptibility region. RESULTS: We generated a high-resolution physical map that provides more than 90% coverage of a refined Blau susceptibility region. The map consists of four contigs of sequence tagged site-based bacterial artificial chromosomes with a total of 124 bacterial artificial chromosomes, …

Remodeling Of Organelle-Bound Actin Is Required For Yeast Vacuole Fusion, Gary Eitzen, Li Wang, Naomi Thorngren, William Wickner

Actin participates in several intracellular trafficking pathways. We now find that actin, bound to the surface of purified yeast vacuoles in the absence of cytosol or cytoskeleton, regulates the last compartment mixing stage of homotypic vacuole fusion. The Cdc42p GTPase is known to be required for vacuole fusion. We now show that proteins of the Cdc42p-regulated actin remodeling cascade (Cdc42p --> Cla4p --> Las17p/Vrp1p --> Arp2/3 complex --> actin) are enriched on isolated vacuoles. Vacuole fusion is dramatically altered by perturbation of the vacuole-bound actin, either by mutation of the ACT1 gene, addition of specific actin ligands such as latrunculin …

The doctrine of informed consent, defined as respect for autonomy, is the tool used to govern the relationship between physicians and patients. Its framework relies on rights and duties that mark these relationships. The main purpose of informed consent is to promote human rights and dignity. Some researchers claim that informed consent has successfully replaced patients' historical predispositions to accept physicians' advice without much explicit resistance.

Although the doctrine of informed consent promotes ideals worth pursuing, a successful implementation of these ideals in practice has yet to occur. What has happened in practice is that attorneys, physicians, and hospital administrators …

Biochemical Study Of A Psi-Lhci Complex And Molecular Study Of Fcp Genes In The Diatom Cylindrotheca Fusiformis, Hua Zhang

A photosystem I-light-harvesting complex I (PSI-LHCI) was isolated from oxygen-evolving thylakoids of the diatom Cylindrotheca fusiformis. The circular dichroism (CD) spectrum of the complex resembled the photosystem I (PSI) complex of green plants. A single 77K fluorescence emission was observed at 715 nm. The excitation spectrum confirmed that both chlorophyll c and carotenoids were energetically coupled to chlorophyll a. The complex contained PSI reaction center proteins (PsaA/B, the PSI accessory subunit PsaC, and nine light-harvesting complex (LHC) apoproteins including an 18kD and a 17.5kD protein. Photosystem II core polypeptides were not detected by immuno- or silver staining. Taken together, the …

Offshore oil development in Arctic Alaska is expanding. Since biodegradation is a major removal mechanism of petroleum hydrocarbons from the environment, baseline data on microorganisms present are important. I estimated microbial populations and their degradation activities in Arctic Ocean sediments, and examined how sediment affects phenanthrene bioavalability. Populations of hydrocarbon degraders were significantly higher in sediments from near Prudhoe Bay than near Barrow. However, microbial counts from Prudhoe Bay were similar to those measured in the 1970s suggesting that the difference was not due to oil development activities. Total microbial counts were higher than in more temperate regions. All sediments …

The transition from cross- to self-fertilization is considered a major pattern in the evolution of angiosperms. Yet, evolutionists continue to struggle to explain the evolutionary processes involved in maintaining both self- and cross-fertilization, which often occur within the same species. The diversity of mating systems suggests that selective pressures are also diverse, sometimes promoting selfing and other times promoting outcrossing. Inbreeding depression is commonly invoked as the primary selective force balancing the advantages of selfing or promoting outcrossing. This research began to establish alternate possible means for determining the biological affinity for fragmented skeletal remains between geographically disparate populations, specifically American-Caucasians and Southeast Asians. The goal is to determine the feasibility of developing a method that may be used in the field to differentiate the remains of U.S. servicemen from people of Southeast Asia. This technique is based upon differences in bone histomorphometric variables. The variables used in this research are the osteon area (On. Ar), Haversian canal area (H. The effect of egg weight on survival and development of pink salmon (Oncorhynchus gorbuscha) embryos, alevins, and fry was analyzed; in addition, embryo survival was investigated in relation to additive genetic variation. Embryonic survival to eyeing, development time to hatch, yolk weight, somatic tissue weight, yolk use rate, somatic tissue growth rate, and the survival of first-feeding fry was recorded relative to egg weight. The analyses demonstrated significant egg weight effects on development time to hatch, yolk weight, somatic tissue weight, yolk use rate, and somatic tissue growth rate on alevins. Effects On Fitness Traits Of Intercrossing Three Geographically Separate Populations Of Southeast Alaska Coho Salmon (Oncorhynchus Kisutch), Karla Louise Granath

Adaptive differences among three geographically separate populations of coho salmon (Oncorhynchus kisutch) were investigated by forming first generation intercrosses (hybrid lines) and comparing them to parental types (control lines). Survival, development time, size at ponding, and first year growth were measured as indicators of locally adapted fitness traits. Significant differences (p <0.05) were found between mid-hatching times for control and hybrid groups. Effects of intercrossing on development time are consistent with additive genetic variation, indicating that important genetic divergence exists between the populations. Growth studies were broken down into four different experiments; a 'common garden' experiment, containing all possible controls and reciprocal crosses between pairs of populations, and three 'hybrid' experiments which contained one of the three control lines and all reciprocal crosses associated with the control. Effects of intercrossing on growth were more apparent in the hybrid experiments where there were fewer interactions between different lines.

A Protocol For Assessing The Impacts Of Urbanization On Coho Salmon With Application To Chester Creek, Anchorage, Alaska, Matthew S. Whitman

Coho salmon (Oncorhynchus kisutch) abundance has declined in many urban streams. The causes of these declines can be hard to identify because urban impacts on stream ecology are complex and can vary between watersheds. This makes it difficult to develop appropriate and effective strategies for stream rehabilitation or mitigation aimed at increasing coho productivity. To improve this situation I developed a habitat quality assessment protocol for urban coho salmon to help identify significant habitat degradation as a prelude to restoration planning. Extensive research has been conducted on issues of sexuality for women with Alcohol Use Disorders (AUD). These issues are relevant both to the development of and recovery from AUD. Little of this research has focused on the importance of women's beliefs about sexuality at the time of drinking and during recovery. This study sought to identify these beliefs and to determine their importance in the development of and recovery from AUD. A qualitative research design was used whereby interviews with four women in long-term recovery (3 or more years) were analyzed. It was found that, overall, beliefs about sexuality became …

Body size is an important determinant of life history traits such as survival and fecundity. There is a positive correlation between growth during the first summer and final body size in goose populations. I examined how environmental factors influence growth in Black Brant (Branta bernicla; hereafter brant) goslings. Growth declined seasonally and varied among brood-rearing areas. However, the pattern was not consistent among years. Models containing only environment and maternal effects explained 75% of variation in gosling mass, indicating that little of the observed variation in size is directly of genetic origin. Heritability did not differ from zero for both …
