Turkish Journal of Veterinary & Animal Sciences
In this study, the thyroid hormone levels in sheep experimentally treated with selenium were investigated. Twelve Akkaraman sheep were used in the investigation. Selenium was given intramuscularly at doses of 0.1 (Group 1) and 0.4 mg/kg (Group 2). The blood samples were taken from the jugular vein at 1, 2, 4, 8, 24, 48, 96 and 144 hours after the administration. Serum triiodotyronine (T3), thyroxin (T4), free triiodotironine (fT3) and free thyroxin (fT4) levels were determined using a hormone autoanalyzer. In the comparison between the groups, it was shown that serum T3 levels reached the maximum levels at the 96^{th} …

Postpartum anoestrus is widely known to cause infertility in buffaloes. This study was conducted to determine the problem of postpartum anoestrus using rectal palpation and milk progesterone level in 8 Murrah buffaloes calved during late spring or early summer in the Kocatepe Agricultural Research Institute, Afyon, Turkey. In addition, the pregnancy rates of experimental animals were investigated using a combined therapy of a progesterone releasing intravaginal device (PRID) and pregnant mare serum gonadotropin (PMSG). The ovaries of each buffalo were examined by rectal palpation once a week and the milk progesterone level was detected twice a week using enzyme immunoassay …

This study was carried out to investigate the changes occurring in the levels of serum thyroid hormone in rams given indomethacin, furosemide and PGF2\alpha. In the study, 20 Akkaraman rams, approximately 2 years old were used. Indomethacin (3.0 mg/kg), furosemide (2.0 mg/kg) and PGF2\alpha (0.1 mg/kg) were intramuscularly given to the animals in one dose and repeated for four consecutive days. Blood samples were collected after drug administrations. Free-total thyriiodothyrionine and thyroxine (T3 and T4) levels were determined using a hormone autoanalyser. Serum free T3 and T4 levels in rams given PGF2\alpha were found to have fallen compared to the …

Detection Of Infectious Bronchitis Virus In Tracheal Organ Cultures Using The Immunoperoxidase Technique, Ayşi̇n Şen, Gürsel Sönmez, Vi̇ldan Caner, M. Özgür Özyi̇ği̇t
Turkish Journal of Veterinary & Animal Sciences
The applicability of the immunoperoxidase (IP) technique in unfixed-fresh tracheal organ cultures (TOCs) was investigated for rapid detection of infectious bronchitis virus (IBV). For this purpose, TOCs prepared from one-day-old SPF chicks were inoculated with IBV M41 strain in serial dilutions. IBV antigen detected by IP technique in TOCs 18 h postinoculation. IP staining was observed before siliostasis in all dilutions. The IP technique was also applied to detect IBV antigen in formalin-fixed, paraffin-embedded sections of TOCs inoculated with IBV M41 strain. Viral antigen was also detected in these sections. Histopathological examinations of IBV M41 infected tracheal rings showed loss …

Due to the recent increasing mortality rates in 1- to 4-month-old Thoroughbred foals with Rhodococcus equi infection in Turkey, serum samples from 454 foals between 30 and 60 days on studfarms of the Marmara Region, where the Thoroughbred foal population is concentrated, were investigated by ELISA for early diagnosis of the infection. The Tween 20-extracted antigen of the R. equi ATCC 6939 strain was used in the ELISA. Two hundred and nine (46%) of the 454 foals were found to be ELISA-positive. Optical density (OD) values of 0.3 (positive limit) or higher were detected in 43% of foals aged 30-40 …

Turkish Journal of Veterinary & Animal Sciences
The aim of this study was to diagnose Theileria annulata infected cattle by microscopy, IFAT and nested PCR and to compare the sensitivity and specificity of these diagnostic methods. A total of 147 blood and sera samples were collected from healthy cattle in four localities of Polatlı district, Ankara, where tropical theileriosis is prevalent. Examination results revealed positivity rates of 31.3%, 44.9% and 61.2% for microscopy, IFAT and nested PCR, respectively. It was shown that nested PCR is more sensitive and specific when compared to microscopical examination and serological findings. It was also shown that nested PCR is more sensitive …

Turkish Journal of Veterinary & Animal Sciences
The liver damage and obstructions of the biliary tract were evaluated ultrasonographically in experimentally bile duct ligated dogs. Seven healthy dogs of different breeds were used. The common bile duct was double ligated at the duodenal region after clinical and ultrasonographic examinations. After that, clinical examinations, biochemical analyses and examinations of the liver, gallbladder and biliary tract were done ultrasonographically over 6 weeks. All dogs were necropsied and macroscopic and histologic alterations were determined at the end of the study. It was determined that the levels of serum ALT, ALP, GGT and T. Bilirubin were (p < 0.001-0.05) statistically significantly increased after the operation in all dogs. As a result of ultrasonographic examinations, it was determined that the bile ducts and gallbladder were distended, the neck of the gallbladder was larger and more tortuous than normal, hypoechoic sludge was prominent in the gallbladder, and perivascular, multifocal hyperechoic areas appeared in the liver parenchyma toward the end of the study. These alterations were confirmed macroscopically and microscopically at the end of the autopsy in all dogs. We consider that as well as blood serum parameters, ultrasonographic examinations are useful in the early diagnosis of biliary diseases and in differentiating them from hepatic diseases.

Cloning And Expression Of Bacteriophage T4 Lysozyme Gene (Gene E) In Escherichia Coli Via Pcr Amplification, Ali̇ İrfan Güzel, Numan Özcan, Hali̇l Kasap
Turkish Journal of Veterinary & Animal Sciences
Lysozyme (EC 3.2.1.17) is an enzyme that catalyzes the hydrolysis of \beta(1-->4) glycosidic bonds between N-acetylglucosamine (NAG) and N-acetylmuramic acid (NAM), which are present in peptidoglycan heteropolymers of the prokaryotic cell wall. The purpose of this study was the cloning and expression of the bacteriophage T4 lysozyme gene (gene e) via PCR amplification in Escherichia coli using the pUC18 and pRS416 vectors. PCR amplification is carried out by using the primers having Bam HI recognition sites at their 5' ends. After digesting the vectors and PCR product with Bam HI endonuclease, two recombinant plasmids were constructed by ligation reaction …

Turkish Journal of Veterinary & Animal Sciences
In this study, the lethal chlorine concentrations (LC) and residual chlorine concentrations in water for a fouling organism, the pearl oyster Pinctada radiata (Leach, 1814) [Avicula], were investigated during May 1999-April 2000. The study was carried out under laboratory conditions at 20ºC. Acute toxicity tests were conducted under a flow through system by using continuous application of chlorine according to bioassay test methods. Test solutions were made by using an appropriate amount of sodium hypochlorite (NaOCl^{-}). According to the results, the 24h-LC_{50} was 1.75 mgl^{-1} and the residual chlorine was 0.47 mgl^{-1} for LC_{50} of Pinctada radiata.
