Interleukin-12 (IL-12) has been used in numerous immunotherapy protocols against melanoma. However, delivery of IL-12 in the form of recombinant protein can result in severe toxicity, and gene therapy has had limited success against B16.F10 murine melanoma. The purpose of this study was to examine the effectiveness of in vivo electroporation for the delivery of plasmid DNA encoding IL-12 as an antitumor agent against B16.F10 melanoma. We treated mice bearing established B16.F10 melanoma tumors with intratumoral (i.t.) or intramuscular (i.m.) injections of a plasmid encoding IL-12, followed by in vivo electroporation. Improved Energy Model For Membrane Electroporation In Biological Cells Subjected To Electrical Pulses, R. P. Joshi, Q. Hu, K. H. Schoenbach, H. P. Hjalmarson

A self-consistent model analysis of electroporation in biological cells has been carried out based on an improved energy model. The simple energy model used in the literature is somewhat incorrect and unphysical for a variety of reasons. Our model for the pore formation energy E(r) includes a dependence on pore population and density. It also allows for variable surface tension, incorporates the effects of finite conductivity on the electrostatic correction term, and is dynamic in nature. Self-consistent calculations, based on a coupled scheme involving the Smoluchowski equation and the improved energy model, are presented. It is shown that E(r) becomes …

Mechanisms Counteracting Swelling In Brain Cells During Hyponatremia, Herminia Pasantes-Morales, Rodrigo Franco, Benito Ordaz, Lenin D. Ochoa

Water gain in the brain consequent to hyponatremia is counteracted by mechanisms that initially include a compensatory displacement of liquid from the interstitial space to cerebrospinal fluid and systemic circulation and subsequently an active reduction in cell water accomplished by extrusion of intracellular osmolytes to reach osmotic equilibrium. Potassium (K+), chloride (Cl–), amino acids, polyalcohols, and methylamines all contribute to volume regulation, with a major contribution of ions at the early phase and of organic osmolytes at the late phase of the regulatory process. The specificity of the ATP:corrinoid adenosyltransferase
(CobA) enzyme of Salmonella enterica serovar
Typhimurium LT2 for its nucleotide substrate was
tested using ATP analogs and alternative nucleotide donors.
The enzyme showed broad specificity for the nucleotide
base and required the 2’-OH group of the ribosyl
moiety of ATP for activity. 31P NMR spectroscopy
was used to identify inorganic triphosphate (PPPi) as
the byproduct of the reaction catalyzed by the CobA
enzyme. Cleavage of triphosphate into pyrophosphate
and orthophosphate did not occur, indicating that
triphosphate cleavage was not required for release of
the adenosylcorrinoid product. Triphosphate was …

In light of the 6oth anniversary of Pearl Harbor and the 9-11 terrorist attacks, Americans are reflecting on the role of patriotism and the military. For many in the Boise State community, military service was a duty and the horrors of war remained with them for the rest of their lives. In far-flung locations, some with exotic and unpronounceable names, they earned a new appreciation for the rights and freedoms we so often take for anted.
