Identification Of A Lactose Utilization And Copper Resistance Plasmid In Lactococcus Lactis Subsp. Lactis Mcl64, Mustafa Akçeli̇k

Turkish Journal of Veterinary & Animal Sciences

Lactose negative and copper sensitive (Lac-/Cus) mutants of Lactococcus lactis subsp. lactis MCL64 were isolated after extended incubation periods at 32ºC and 37ºC. A 30.0 kb plasmid was absent in all Lac-/Cus mutants. These results showed that lactose metabolism and copper resistance were linked to the 30.0 kb plasmid in strain MCL64. Using MCL64 as a donor, the 30.0 kb plasmid transferred into Lac-/Cus recipient strains P81 and MCL64-90 with the frequencies of 4.1x10-5- 8.2x10-3 per donor cell, respectively. All transconjugants were found to be of a Lac+/ Cur phenotype.

The effects of outdoor raising on the survival and growth of Simmental calves in individual hutches were studied at Kazova State Farm, the location having a mixture of Black Sea and semiarid continental climate conditions. A total of 63 female calves born in each of the four seasons between March 1996 and February 1997 were used. The animals were kept in individual hutches for 3 months (preweaning period) and then in groups of 10 until 6 months of age (postweaning period). Fifteen New Zealand rabbits were used in this study. Buffered saline solution was given through the common carotid artery to remove the blood from the cranial veins. The jugular vein was ligated prior to giving blue latex to 10 rabbits and blue Batson's No. 17 to 5 rabbits through the maxillary vein. In this study, our aim was to diagnose lymphoid leukosis and Marek's disease, which are neoplastic diseases of chickens, by ELISA and Indirect Immunofluorescence Test (IIF), respectively. Samples (eggs and tissues) were selectively taken from the flocks suspected of having lymphoid leukosis and Marek's disease. The tissues were macroscopically and microscopically examined. Tissues belonging to some flocks showed typical lesions for lymphoid leukosis and Marek's disease. For enzymatic analysis and plasmid isolation, the gene was also expressed in xylanase and beta glucanase negative B.subtilis YB886 strain with pUB110 vector. In the survey study, a total of 19 canine mast cell tumors were examined with regard to sex, age, breed of dogs, tumor location, and pathological findings in the period of 1960-1999. The majority of tumors occurred in males (78%) and between the ages of 6 and 11 years (67%). The pointer was predisposed to development of tumors (35%). The hindquarters and hind legs were most commonly involved, followed by the extragenital inguinal area and trunk. The tumors clearly had a site preference for the posterior region of the body (79%). Carbon isotopic data of interstitial dissolved CO2 (ΣCO2), CO2 gas, and methane show that a variety of microbial diagenetic processes produce the observed isotopic trends. Anaerobic methane oxidation (AMO) is an important process near the sulfate-methane interface (SMI) that strongly influences the isotopic composition of ΣCO2 in the sulfate reduction and upper methanogenic zones, which in turn impacts methane isotopic composition. Dissolved CO2 and methane are maximally depleted in 3C near the SMI, where C values are as light as –31.8‰ and –101‰ PDB for ΣCO2 and methane, respectively. Survey Of European Foulbrood Disease In Bee Farms Located In Elazığ And The Surrounding Provinces, Hali̇l Şi̇mşek, Canan Özcan

A total of 250 specimens from 110 bee farms located in Elazığ and the surrounding provinces were taken in 1999 and 2000. The specimens were examined for European Foulbrood Disease. Under macroscopic examination clinical symptoms were noted in 12 specimens, and Paenibacillus alvei was isolated and identified from 8 of these specimens. As a result of the antibiotic sensitivity test, the isolate was found sensitive to enrofloxacin, streptomycin and erythromycin 75%, 75%, and 62,5% of the time respectively. In this study, the presence of European Foulbrood Disease was determined in 3.2% of cases in Elazığ and the surrounding provinces.

A total of 473 sera from hens of different age groups and different breeds and a total of 430 eggs were collected from 11 breeders and egg producers in the districts of Adapazarı, Afyon, Ankara and Konya. Lipopolysaccharide (LPS), heat-extract (HE) and flagella antigens were tested in the standardizing ELISA technique. Flagella antigen was found useful in the screening of field samples. Of the 48 (10.2%) rapid plate agglutination positive sera, 7 were negative in ELISA. Exposure of buffalo liver and kidney tissue arginases to a 150 w light source from a distance of 8-10 cm, in the presence of 0.05mM methylene blue, caused significant inhibition of arginase activity. It was determined that this inhibition occurred differently in liver and kidney tissues. Arginase activity was spectrophotometrically measured using the Thiosemicarbazide diacetyl-monoxime urea (TDMU) method. Genetic parentage in the socially monogamous and territorial Eastern Kingbird( Tyrannust tyrannus) was examined in a central New York population by multilocus DNA fingerprinting. Extra-pair young were identified in 60% (12 of 20) of nests. Of the 64 nestlings profiled, 42% were sired by extra-pair males, but no cases of conspecific brood parasitism were detected. These results are markedly different from a previous electrophoretic study of the same species in a Michigan population, which reported 39% of nestlings were unrelated to one (typically the mother, quasiparasitismo)r both (conspecificb roodp arasitism) of the putative parents. The paper presents an outline of the issues and a preliminary appraisal of the use of trade sanctions by the World Trade Organization (WTO) as a means of promoting compliance by parties. The WTO is unique among intergovernmental organizations (IGOs) in using trade sanctions to enforce independent adjudications. Many commentators have suggested using trade sanctions analogously in other IGOs, or alternatively broadening trade rules so that the sanctions can be used for other purposes, such as enforcing basic human rights.
