To generate a plant-based multicomponent oral vaccine against cholera, rotavirus and enterotoxigenic E.coli (ETEC), we have constructed transgenic potato plants that synthesize the cholera toxin B and A2 subunits, the rotavirus enterotoxin NSP4 22 amino acid epitope and the ETEC fimbrial antigen CFA/L To use cholera toxin as a carrier molecule for delivery of multiple antigens to the gut-associated lymphoid tissues, the rotavirus NSP4 epitope was linked to the cholera toxin B subunit (CTB) and the ETEC fimbrial antigen CFA/I was linked to the cholera toxin A2 subunit. Bone is a dynamic tissue which, through the process of bone remodeling in the mature skeleton, renews itself during normal function and adapts to mechanical loads. It is, therefore, important to understand the effect of remodeling on the mechanical function of bone, as well as the effect of the inherent time lag in the remodeling process. In this study, we develop a constitutive model for bone remodeling which includes a number of relevant mechanical and biological processes and use this model to address differences in the remodeling behavior as a volume element of bone is placed in disuse or overload. …

Chronic human exposure to nonovertly toxic doses of arsenic is associated with an increased risk of cancer. Although its carcinogenic mechanism is still unknown, arsenic does not directly cause DNA damage or mutations and is therefore thought to act principally as a co-mutagen, co-carcinogen, and/or tumor promoter. Previous studies in our laboratory demonstrated that effects of low-dose arsenic (III) (arsenite) on expression of the hormone-regulated phosphoenolpyruvate carboxykinase (PEPCK) gene were strongly associated with the glucocorticoid receptor (GR)-mediated regulatory pathway. We therefore examined specifically the effects of arsenite on the biochemical function of GR in hormone-responsive H4IIE rat hepatoma cells. Completely …

In her article, "Gender, Literature, and Film in Contemporary East Central European Culture," Anikó Imre discusses gender, literature, and film in Hungary in the context of East Central European national cultures of the 1980s and 1990s. Anikó Imre analyzes the analogous gender structures that underlie both nation and literature in these transitional cultures. She challenges both social science studies of post-communist transitions and studies of East Central European literatures and cultures for their traditional neglect of gendered desire as a political factor. Black bear can inflict severe negative Impacts on timber stands in the northwestern United States. A supplemental feeding program to provide bears an alternative food source during spring is practiced in the stat e of Washington, and to a lesser extent in other states. We initiated concurrent studies to assess characteristics of bear that forage at feeding stations, the interactions of bears around feeders, and impacts of the program on bear territories. Numerous bears fed at stations, including females with and without cubs, yearlings. and males. Bear feeding bouts at stations were generally short, less than 15 minutes. Bears generally …

In her article, "Testimonial Poetry in East European Post-Totalitarian Literature," Albena Lutzkanova-Vassileva reexamines the belief that postmodern literature and deconstructive writing have parted literary and theoretical discourse from reality, thereby obstructing and annihilating our access to history. Lutzkanova-Vassileva exemplifies her prognosis in an inquiry into post-totalitarian and postmodern Bulgarian literature and its texts of poetry. Born in the turmoil of communism's debacle, the analysis is an attempt to illustrate that, contrary to denying reference, postmodernism solely rejects the reduction of reference to a world that is perceptible and cognitively masterable. Humans affect biodiversity at the genetic, species, community, and ecosystem levels. This impact on genetic diversity is critical, because genetic diversity is the raw material of evolutionary change, including adaptation and speciation. Two forces affecting genetic variation are genetic drift (which decreases genetic variation within but increases genetic differentiation among local populations) and gene flow (which increases variation within but decreases differentiation among local populations). Humans activities often augment drift and diminish gene flow for many species, which reduces genetic variation in local populations and prevents the spread of adaptive complexes outside their population of origin, thereby disrupting adaptive processes …

The wing-polymorphic cricket, Gryllus firmus, contains (1) a flight-capable morph (LWf) with long wings and functional flight muscles, (2) a flightless morph with reduced wings and underdeveloped flight muscles (SW), and (3) a flightless morph with histolyzed flight muscles but with fully developed wings (LWh). The LWf morph differed genetically from the SW morph and phenotypically from the LWh morph in the size of flight muscles, ovarian growth during the first week of adulthood, and the hemolymph titer of juvenile hormone (JH). The hormonal basis of variation in life-history traits is a poorly studied topic in life-history evolution. An important step in identifying the endocrine-genetic causes of life-history variation is documenting statistical and functional associations between hormone titers and genotypes/phenotypes that vary in life-history traits. To this end, we compared the blood ecdysteroid titer and the mass of the ovaries during the first week of adulthood among a flight-capable morph and two fl ightless morphs of the wing-polymorphic cricket Gryllus firmus. Using published experimental data on the thermal conductivity, mutual diffusivity, and heats of transport, the degree of coupling between heat and mass flows has been calculated for binary and ternary nonideal liquid mixtures. The binary mixtures consist of two types: the first is six systems of six-to-eight-carbon straight and branched chain alkanes in chloroform and in carbon tetrachloride; and the second is mixtures of carbon tetrachloride with benzene, toluene, 2-propanone, n-hexane, and 11-octane. The ternary mixture considered is toluene-chlorobenzene-bromobenzene. The published data are available at 35°C, 30°C, and 35°C and ambient pressure. The newest synthesis of evolutionary thought is emerging, and promises to return evolutionary biology to Darwin’s panoramic view of life. The key element is a long-standing dualism in evolutionary theory. This dualism has a long history within evolutionary biology, being manifested under guises such as: (1) the nature of the organism and the nature of the conditions, (2) internal and external, or intrinsic and extrinsic, factors, (3) production and exchanges, (4) boundary and initial conditions, (5) metabolism and replication, (6) energy and information, and (7) costs and benefits, and conflict and resolution. Carvacrol and thymol in combination at 6.7 mM each completely inhibited the production of short-chain volatile fatty acids and lactate from cattle waste in anoxic flasks over 23 days. Fecal coliforms were reduced from 4.6 x 106 to 2.0 x 103 cells per ml 2 days after treatment and were nondetectable within 4 days. Total anaerobic bacteria were reduced from 8.4 x 1010 to 1.5 x 107 cells per ml after 2 days and continued to be suppressed to that level after 14 days. The ability to accurately predict a polypeptide's molecular structure given its amino acid sequence is important to numerous scientific, medical, and engineering applications. Studies have been conducted in the application of Genetic Algorithms (GAs) to this problem with promising initial results. In this thesis report, we use the fast messy Genetic Algorithm (fmGA) to attempt to find the minimization of an empirical CHARMM energy model and generation of the associated conformation. Previous work has shown that the fmGA provided favorable results, at least when applied to the pentapeptide Met-Enkephalin. We extend these results to a larger Polyalinine peptide by utilizing …

Chlorinated ethenes are among the most prevalent groundwater contaminants at hazardous waste sites nationwide. In an attempt to manage the risks posed by these contaminants, while controlling costs, monitored natural attenuation (MNA) is being considered as a remediation strategy at many sites. MNA relies on naturally occurring physical, chemical, and biological processes in the subsurface to reduce the risk posed by the contamination. The implementation of MNA, however, requires a detailed understanding of these processes, and how they impact contamination at a particular site. Multicolor infrared imaging missile-warning systems require real-time detection techniques that can process the wide instantaneous field of regard of focal plane array sensors with a low false alarm rate. Current technology applies classical statistical methods to this problem and ignores neural network techniques. Thus the research reported here is novel in that it investigates the use of radial basis function (RBF) neural networks to detect sub-pixel missile signatures. Conventional technologies for the treatment of groundwater contaminated with chlorinated solvents have limitations that have motivated development of innovative technologies. One such technology currently under development involves using palladium-on-alumina (Pd/Al) as a catalyst to promote dechlorination. Pd/Al catalyst may be used in-well as part of a re-circulating horizontal flow treatment well (HFTW) system. An HFTW system involves two or more dual-screened wells, with in-well reactors, to capture and treat contaminated groundwater without the need to pump the water to the surface.
