Acetyl-CoA carboxylase catalyzes the first committed step in fatty acid synthesis in all plants, animals, and bacteria. The Escherichia coli form is a multimeric protein complex consisting of three distinct and separate components: biotin carboxylase, carboxyltransferase, and the biotin carboxyl carrier protein. The biotin carboxylase component catalyzes the ATP-dependent carboxylation of biotin using bicarbonate as the carboxylate source and has a distinct architecture that is characteristic of the ATP-grasp superfamily of enzymes. Included in this superfamily are D-Ala D-Ala ligase, glutathione synthetase, carbamyl phosphate synthetase, N⁵-carboxyaminoimidazole ribonucleotide synthetase, and glycinamide ribonucleotide transformylase, all of which have known three-dimensional …

CYP83B1 from Arabidopsis thaliana has been identified as the oxime-metabolizing enzyme in the biosynthetic pathway of glucosinolates. Biosynthetically active microsomes isolated from Sinapis alba converted p-hydroxyphenylacetaldoxime and cysteine into S-alkylated p-hydroxyphenylacetothiohydroximate, S-(p-hydroxyphenylacetohydroximoyl)-L-cysteine, the next proposed intermediate in the glucosinolate pathway. The production was shown to be dependent on a cytochrome P450 monooxygenase. We searched the genome of A. thaliana for homologues of CYP71E1 (P450ox), the only known oxime-metabolizing enzyme in the biosynthetic pathway of the evolutionarily related cyanogenic glucosides. The effects of Escherichia coli heat-stable enterotoxin (ST) and uroguanylin were examined on the proliferation of T84 and Caco2 human colon carcinoma cells that express guanylyl cyclase C (GC-C) and SW480 human colon carcinoma cells that do not express this receptor. ST or uroguanylin inhibited proliferation of T84 and Caco2 cells, but not SW480 cells, in a concentration-dependent fashion, assessed by quantifying cell number, cell protein, and [(3)H]thymidine incorporation into DNA. These agonists did not inhibit proliferation by induction of apoptosis, assessed by TUNEL (terminal deoxynucleotidyl transferase-mediated dNTP-biotin nick end labeling of DNA fragments) assay and DNA laddering, or necrosis, …

A dual beam low coherence interferometer with improved signal to noise ratio. The interferometer directs both a collimated beam about a focused beam to a target. A detector detects the multiscattering from the target with a splitter where a reference arm with matched mirrors allows for measuring both the intensity and magnitude values of the beam to be measured. The intensity and magnitude values indicate the imaging of the target in applications such as OCT(optical coherence tomography) having medical imaging applications, while improving the signal to noise ratios. Other applications include material characterization in ceramics, composites and other granular material. …

Recently we have identified a novel protein NIP45 (nuclear factor of activated T cells [NFAT]-interacting protein) which substantially augments interleukin (IL)-4 gene transcription. The provision of NIP45 together with NFAT and the T helper cell type 2 (Th2)-specific transcription factor c-Maf to cells normally refractory to IL-4 production, such as B cells or Th1 clones, results in substantial IL-4 secretion to levels that approximate those produced by primary Th2 cells. In studies designed to further our understanding of NIP45 activity, we have uncovered a novel facet of IL-4 gene regulation. We present evidence that members of the tumor necrosis factor …

The Foraging Ecology Of The Juvenile Gopher Tortoise, Gopheruspol Yphemus, Terri Anne Stilson

Food plant preferences of juvenile gopher tortoises, Gopherus polyphemus, were investigated in a west central Florida population. The juvenile gopher tortoises were free-ranging and were located in a sandhill habitat subject to yearly prescribed burns. Diet was determined by foraging observations, in which the number of bites taken per taxon, the number of plants sampled per taxon, and the amount of each taxon available along the foraging path were recorded. Preference was determined with Manly's alpha index of preference and Jacob's D electivity index. Selected plants in the habitat were analyzed for relative nitrogen content to investigate whether preferred …
