Farnesyl Pyrophosphate Analogs, Hans Peter Spielmann, Douglas A. Andres, Kareem A.H. Chehade
Molecular and Cellular Biochemistry Faculty Patents
The post-translational addition of a farnesyl moiety to the Ras oncoprotein is essential for its membrane localization and is required for both its biological activity and ability to induce malignant transformation. The present invention describes design and synthesis of a farnesylpyrophosphate (FPP) analog, 8-anilinogeranyl pyrophosphate (AGPP) that is transferred to Ras by farnesyltransferase (FTase), in which the ω-terminal isoprene unit of the farnesyl group has been replaced with an aniline functionality. AGPP potently inhibited FTase activity in vitro (IC50=0.6 μM) and is highly selective showing little inhibitory activity against either geranylgeranyl-protein transferase type I (GGTase I) (IC50 …

Dna Vector For Determining The Presence Of Out-Of-Reading-Frame Mutations, Gary W. Litman, Noel A. Hawke, Robert N. Haire, Scott J. Strong

A DNA expression vector for positively selecting in-frame or out-of-reading-frame mutations in DNA sequences to be tested comprising a promotor operatively linked to an expressible reporter gene through a linkage sequence is disclosed. The linkage sequence includes at least two restriction sites and an engineered frameshift mutation. In an embodiment the frameshift is established by complementary sequences SEQ ID Nos:1 and 2. The expressible reporter gene is expressed as a fusion product including a green fluorescent protein and the promoter can be lacZ and inducible in E. coli.

Tyrannasorus rex Ratcliffe and Ocampo (Coleoptera: Scarabaeoidea: Hybosoridae), a new genus and new species of fossil hybosorid, is described from the Dominican Republic. This fossil scarabaeoid is embedded in the amber resin of Hymenaea protera Poinar (Leguminosae), and it dates from the Miocene.

Eliciting The Low-Activity Aldehyde Dehydrogenase Asian Phenotype By An Antisense Mechanism Results In An Aversion To Ethanol., E Garver, Tu Gc, Q N Cao, M Aini, F Zhou, Y Israel

A mutation in the gene encoding for the liver mitochondrial aldehyde dehydrogenase (ALDH2-2), present in some Asian populations, lowers or abolishes the activity of this enzyme and results in elevations in blood acetaldehyde upon ethanol consumption, a phenotype that greatly protects against alcohol abuse and alcoholism. We have determined whether the administration of antisense phosphorothioate oligonucleotides (ASOs) can mimic the low-activity ALDH2-2 Asian phenotype. Rat hepatoma cells incubated for 24 h with an antisense oligonucleotide (ASO-9) showed reductions in ALDH2 mRNA levels of 85% and ALDH2 (half-life of 22 h) activity of 55% equivalent to a >90% inhibition in ALDH2 …

A Review Of The Neotropical Genus Neocorvicoana Ratcliffe And Micó, New Genus (Coleoptera: Scarabaeidae: Cetoniinae: Gymnetini), Brett C. Ratcliffe, Estefanía Micó

The southern Neotropical cetoniine genus Neocorvicoana Ratcliffe and Micó new genus (Coleoptera: Scarabaeidae: Cetoniinae: Gymnetini) is established for three species previously placed in Corvicoana Strand, 1934 (nomen nudum) and Gymnetis MacLeay, 1819: N. chalybea (Kirby), N. reticulata (Schürhoff), and N. tricolor (Schürhoff), new combinations. The type species of the new genus is N. reticulata (Kirby). Corvicoana rudolfi (Frölich) is transferred to the genus Gymnetis. Corvicoana suavis (Schürhoff) and C. castanea (Schürhoff) are junior synonyms of N. chalybea. We include a taxonomic key to the species, nomenclatural history, descriptions, illustrations, and commentary.

Opportunities for fall spraying have improved considerably with significant precipitation over a wide area. Prior to that, regrowth was slow and growth diminished. Early frost in some areas did not affect Canada thistle foliage and the outlook for warm, sunny weather (60 to 70F) is excellent for response to fall spraying. Best spraying opportunities are for: pasture, roadsides, and CRP that were mowed, grazed, or sprayed with 2,4-D in early season. It is also a good opportunity to have access to fencelines. Guidelines for each are suggested.

Stem Morphology And Anatomy In Amaranthus L. (Amaranthaceae)—Taxonomic Significance, Mihai Costea, Darleen A. Demason

The range of variation within the genus Amaranthus L. (Amaranthaceae) is described for a number of stem characters including: morphology, epidermis, primary stem vasculature and mechanism of secondary growth. The results provide new characters (phyllotaxy, complexity of leaf vascular supply and relative amount of secondary growth) that support (1) a new infrageneric classification (subgenus Amaranthus vs subgenus Albersia (Kunth)Gren. & Dodr.), and (2) the separation within the “hybridus” complex of cultivated amaranths (A. caudatus L., A. cruentus L., and A. hypochondriacus L.) from their presumed wild ancestors (A. hybridus L. subsp. quitensis (Kunth) Costea & Carretero, …
