Since little is known of how the primitive protozoan parasite, Giardia lamblia, senses and responds to its changing environment, we characterized a giardial protein kinase A (gPKA) catalytic subunit with unusual subcellular localization. Sequence analysis of the 1080-base pair open reading frame shows 48% amino acid identity with the cyclic AMP-dependent kinase from Euglena gracilis, Northern analysis indicated a 1.28-kilobase pair transcript at relatively constant concentrations during growth and encystation, gPKA is autophosphorylated, although amino acid residues corresponding to Thr-197 and Ser-338 of human protein kinase A (PKA) that are important for autophosphorylation are absent, Kinetic analysis of the recombinant …

Statistics show the American population is aging at an explosive rate. The correlation between fitness and the aging process is once again becoming a ripe area for study. Previous studies show that fitness can improve quality of life and health for the elderly. A new defined category of seniors known as the young-old has emerged. This category is comprised of seniors that range in age from 55- 65. This study focuses on the relationship between routine exercise and quality of life issues for the young-old. For decades, geographical variations in cancer rates have been observed but the precise determinants of such geographic differences in breast cancer development are unclear. Various statistical models have been proposed. Applications of these models, however, require that the data be assembled from a variety of sources, converted into the statistical models’ parameters and delivered effectively to researchers and policy makers. A web-enabled and GIS-based system can be developed to provide the needed functionality. Strategic Asset Allocation For Individual Investors: The Impact Of The Present Value Of Social Security Benefits, Steve Fraser, William Jennings, David King

This paper demonstrates the dramatic effect of social security wealth on individuals’ asset allocation. We first discuss why social security wealth should be included in portfolio asset-mix decisions. We then draw parallels between social security benefits and inflation-indexed treasury bonds to help quantify the present value of social security benefits. Finally, we show the portfolio impact of including social security wealth under several asset-mix decision rules. Excluding social security wealth from the asset mix decision results in sub-optimal portfolios. Including social security wealth provides an incentive for including more stock in the asset mix.

The goal of these experiments is to determine the relative effects of various agents (Ptpt dinucleotide, lutein, and vitamin C) on the activity of long terminal repeat (LTR) in HIV 1 infected cells (HIV LTR) in both control and UV exposed cells. It is known that the LTR is important in transcription and therefore viral replication. A HeLa cell line transfected with a plasmid that has the HTV-LTR and an E. coli chloranphenicol acetyl transferase (CAT) reporter gene was used. This research assessed the relationships of dietary, sex, and biochemical factors to macular pigment optical density (MPOD) and macular pigment (MP) distribution in 108 adults. Dietary assessment tools were evaluated for their ability to predict MP status. Macular pigment was assessed at four foveal sites and one parafoveal site. An average composite macular pigment value (CMPV) was calculated based on the MPOD results. Group mean MPOD results from most to least central retinal locations, were 0.43 (SE +/- 0.017), 0.36 (SE +/- 0.013), 0.28 (SE +/- 0.012), and 0.13 (SE +/- 0.008), respectively. The opportunistic, deposit-feeding polychaete Capitella sp. I is the overwhelming numerical dominant in disturbed and enriched sediments and rarely co-occurs in appreciable numbers with other abundant mud-dwelling macrofauna. Rapid colonization and population increase in organicrich sediments is typically followed by subsequent sharp decline. The mechanistic basis for these characteristics was explored in flume-flow experiments that tested whether settling Capitella sp. I larvae avoid sediments inhabited by macrofaunal adults or sediments reworked by them. The first set of experiments consisted of four treatments: conspecific adults or no adults in reworked or non-reworked sediment. Capitella sp. I settlement was significantly altered (depressed) …

The purpose of this study was to advance understanding of the self-change process in recovery from anorexia nervosa (AN) and bulimia nervosa (BN) through: (a) development of a staging instrument for recovery from BN; (b) development of TTM-based measures, including decisional balance, self-efficacy, and processes of change; (c) invariance testing of these measures across AN and BN subsamples; and (d) characteristic description of individuals at each stage of change. The efficacy of the lipid soluble carotenoid astaxanthin (biosynthesised as a secondary metabolite by the microalga Haematococcus pluvialis) as a source of pigment to enhance the aesthetic appeal of the flesh of farmed salmonid species has been investigated, using rainbow trout (Oncorhynchus mykiss) as a model. Accretion of astaxanthin within rainbow trout epaxial white muscle was primarily shown to be dependent on dietary inclusion and the type of diet used as a pigment carrier. Different dietary inclusion rates demonstrated a dose response. Cold pressed low oil and commercially extruded high oil diets containing similar astaxanthin concentration were tested, fish receiving …

Studies were undertaken to clone and characterise a two-component regulatory system from a clinical isolate (204) of the human and animal pathogen Burkholderia pseudomallei. A number of genomic libraries were constructed in E. coli host-vector systems and screened for the presence of a two-component system using oligonucleotide probes based on nucleotide sequence homology. Fragments of genomic DNA were cloned and sequenced and found to possess two open reading frames (ORFs) that overlap with a single nucleotide and are believed to encode a novel two-component regulatory system. The biological uptake and transformation of inorganic nitrogen species is described for contrasting marine environments which include a sea loch, riverine plume, polar marginal ice-zone, continental shelf break, oligotrophic ocean and a mesoscale eddy. Uptake of nitrate and ammonium by phytoplankton has been determined using the stable isotope as a tracer and continuous flow stable isotope-ratio mass spectrometry. The analysis of dissolved and atmospheric nitrous oxide was made using electron capture detector gas chromatography in a novel configuration which also allowed for the simultaneous analysis of methane from a single sample. Consumer preference for commercially reared fish products that resemble their wild counterparts has resulted in the supplementation of pigments called carotenoids into aquafeeds to promote a pink-red colour in the flesh of salmonid fish. To date synthetic forms of these pigments have been commonly utilised to achieve this desired colouration, with the carotenoid astaxanthin being the regular choice for the feed manufacturer. A programme of research was carried out to attempt the molecular characterisation of the human and animal pathogen, Burkholderia pseudomallei, the causative agent of melioidosis and the newly described avirulent species, B.thailandensis for comparative purposes. Melioidosis is still little understood, and so the clinical approach to the prevention and control of melioidosis must ultimately rest upon the basic understanding of the causative organism, particularly the pathogenic properties of B.pseudomallei. The long-term effects of sub-lethal dietary Cu-exposure were investigated in relation to consequential energy shifts and resultant locomotory adaptation in 0. mykiss. This study represents the first to attempt to quantify the physiological cost of dietary heavy-metal exposure, the timing and extent of daily behavioural adaptation and resultant significance, in terms of ecologically important behaviours to the fish. Two long-term (2 & 3 months) dietary exposures of 730 mg Cu kgˉ¹ mg d.w. feed showed exposed fish to have a 3-fold increase in liver [Cu] and 10-fold increase in intestinal [Cu] compared to control fish. Carotenoid utilization in rainbow trout (Oncorhynchus mykiss Walbaum) and Atlantic salmon (Salmo salar L.) has been investigated with respect to tissue distribution of carotenoids and the role of the liver on the bioavailability of the lipid soluble carotenoids, astaxanthin and canthaxanthin. Species-specific and tissue-specific accumulations were noted for astaxanthin and canthaxanthin in the rainbow trout and Atlantic salmon, possibly indicating fundamental differences in their utilization in these species. The liver and the kidney were revealed to be the major tissues involved in carotenoid metabolism in both rainbow trout and Atlantic salmon.
