Analysis Of Mitotic Microtubule-Associated Proteins Using Mass Spectrometry Identifies Astrin, A Spindle-Associated Protein, Gary J. Mack, Duane A. Compton

We purified microtubules from a mammalian mitotic extract and obtained an amino acid sequence from each microtubule-associated protein by using mass spectrometry. Most of these proteins are known spindle-associated components with essential functional roles in spindle organization. We generated antibodies against a protein identified in this collection and refer to it as astrin because of its association with astral microtubule arrays assembled in vitro. Astrin is approximately 134 kDa, and except for a large predicted coiled-coil domain in its C-terminal region it lacks any known functional motifs. Speech delivered by WKU president Gary Ransdell at the National Governor's Association Center for Best Practices. In response to Governor Patton's challenges to create Programs of Distinction and enhance the state's capacity for economic development, we, at Western, have created a series of applied science centers. These centers are directing faculty talent toward creating a relevant curriculum in the sciences, toward solving environmental and material problems which limit economic development, and toward creating practice-based experiences for students. A by-product of these centers is a strengthened core curriculum which brings the applied sciences into focus for all WKU students.

Synthesis, Characterization And Electrochemical Studies Of A New Class Of Ligand For Modeling The Bioredox Chemistry Of Copper: A Novel Sensor For Organic Contaminants In Water., Richard Jon Jaeger

In part I of this dissertation, a new set of sulfur containing pendant arm copper(II/I) macrocycles were synthesized to determine the effects of steric constraint on induced structural rearrangement. These new ligands have an N3S3 coordination sphere with nitrogens in a macrocyclic ring and pendant arms containing thiaether groups with varying terminal aliphatic groups. The complexes of these ligands with Cu(II) were characterized using Ultraviolet-visible (UV-Vis) spectroscopy, electronic spin resonance (ESR) spectroscopy and x-ray diffraction analysis. UV-Vis and ESR spectroscopy provided insight into different solution geometries of the copper complexes with respect to solvent and temperature. In general, as the …
