Higher Order Chromatin Degradation Induced By Hydrogen Peroxide In Glial Cells, Raymond Mouzannar Aug 2001

Higher Order Chromatin Degradation Induced By Hydrogen Peroxide In Glial Cells, Raymond Mouzannar

Graduate Theses, Dissertations, and Problem Reports (ETD)

This study addresses the genotoxicity of oxidative stress. We induced oxidative stress in cultured rat oligodendrocytes by exposure to hydrogen peroxide (H2O2). Intact genomic DNA was isolated by agarose embedding and its fragmentation was assessed by field inversion gel electrophoresis (FIGE). To detect single strand fragmentations, the DNA was digested with S1 nuclease before FIGE. We found that the exposure of oligodendrocytes to pathological concentrations of H2O2, induces a rapid digestion of nuclear chromatin in a pattern consistent with higher order chromatin degradation (HOCD). HOCD, the hallmark of programmed cell death (PCD), proceeds through single strand scissions that accumulate at …

A Novel Strategy Utilizing Co-Expression Of Murine Il-12 And Antisense Tgf-Pl Against H238 Tumor Formation, Craig A. Seheult

Loma Linda University Electronic Theses, Dissertations & Projects

Cytokines have a profound effect on immune modulation, thus playing a significant role in cancer gene therapy. A variety of human carcinomas secrete transforming growth factor-beta (TGF-β), a cytokine with potent immunosuppressive properties. Suppressing TGF-β could be a key to successfully combating malignancies, such as gliomas, mammary, and colon cancers, that over-express TGF-β. Conversely, interleukin-12 (IL-12) is a potent immunostimulatory cytokine capable of regulating T and NK cell-mediated cytotoxic responses (Trinchieri, 1994) during an anti-tumor response. Activation of the immune system with such immunostimulatory cytokines renders it inert to the suppressive effects of TGF-β. To better understand these processes our …

Effects Of Liposome-Encapsulated Tnf-Alpha And Radiation Against Colon Cancer, Dong Won Kim Aug 2001

Effects Of Liposome-Encapsulated Tnf-Alpha And Radiation Against Colon Cancer, Dong Won Kim

Loma Linda University Electronic Theses, Dissertations & Projects

Previous studies have shown that tumor necrosis factor-α (TNF-α) can augment the antitumor effects of radiation, inhibiting tumor progression to a greater extent than either modality alone. However, nonspecific toxicities associated with TNF-α have limited its use as an adjuvant. This limitation is potentially resolved with the encapsulation of TNF-α in sterically stabilized, PEGylated liposomes (STEALTH®). Thus, the overall hypothesis of the research was that the combination of radiation with STEALTH® liposome encapsulated TNF-α (SL-TNF-α) will have greater efficacy and fewer dose-limiting side effects in the LS174T human colorectal tumor xenograft model compared to the combination of free TNF-α and …

The Impact Of Logging On Aquatic Salamander Communities., Heather Noel Mcdonald Aug 2001

The Impact Of Logging On Aquatic Salamander Communities., Heather Noel Mcdonald

Electronic Theses and Dissertations

For this study, the abundance of the salamander fauna in two streams was compared to determine the impact of logging activities. After five months of study, the area around one stream was logged by the National Forest Service. Following logging, the abundance of salamanders in the stream was greatly reduced, while the population in the undisturbed stream remained relatively unchanged. Six abundant species were used as indicators; two of these species (Desmognathus quadramaculatus and Desmognathus ochrophaeus) were negatively affected by the logging, while four species (Demognathus fuscus, Desmognathus monticola, Eurycea wilderae, and Gyrinophilus porphyriticus …

Remembering The Forgotten Genocide: Armenia In The First World War., Dana Renee Smythe Aug 2001

Remembering The Forgotten Genocide: Armenia In The First World War., Dana Renee Smythe

Electronic Theses and Dissertations

The Ottoman Empire was in serious decline by the late nineteenth century. Years of misrule, war, and oppression of its various nationalities had virtually driven the Turks from Europe, leaving the weakened Empire on the verge of collapse. By the 1870s the Armenians were the most troubling group, having gained international sympathy at the Congress of Berlin. As a result, violence against the Armenians had escalated dramatically by the turn of the century. They felt, however, that their fortune had changed when the liberal Young Turks seized power from the Sultan in 1908. Unfortunately, the Young Turks had a much …

Integrated Modulation Of Sympathetic Tone In The Microcirculation By Oxygen, Adenosine, And Nitric Oxide, Bryan Auston Sauls Aug 2001

Integrated Modulation Of Sympathetic Tone In The Microcirculation By Oxygen, Adenosine, And Nitric Oxide, Bryan Auston Sauls

Graduate Theses, Dissertations, and Problem Reports (ETD)

Despite a profound reduction in hemodynamic shear stress, a sustained release of endothelium-derived nitric oxide (NO) limits arteriolar constriction during periods of increased sympathetic nerve activity in the rat small intestine. In this project, we sought to test the hypothesis that adenosine, formed in response to a flow-dependent fall in local PO2, serves as the stimulus to preserve endothelial NO release under these conditions. Sympathetic nerve stimulation induced frequency-dependent arteriolar constrictions and a flow-dependent fall in local PO2. The arteriolar responses to nerve stimulation were enhanced after inhibition of NO synthase with NG-monomethyl-L-arginine (L-NMMA). Under a high (20%) O2 superfusate, …

A model with linear algebraic approximation to solids viscosity of the form, mus = 5.34epsilons, (epsilon s is the solids volume fraction) with an appropriate boundary condition at the wall obtained by approximate momentum balance solution at the wall to account for the solids recirculation is tested against experimental results. The work done was to predict the flow patterns in the CFB risers from available experimental data, including data from …

Seasonal Plasticity Of A15 Dopaminergic Neurons In The Ewe, Van Lee Adams Aug 2001

Seasonal Plasticity Of A15 Dopaminergic Neurons In The Ewe, Van Lee Adams

Graduate Theses, Dissertations, and Problem Reports (ETD)

In the ewe, anestrus is caused by increased sensitivity to E2 negative feedback that is mediated by A15 dopaminergic neurons that are only responsive to E2 in anestrus. Experiment 1, tested the hypothesis that synaptic input on A15 neurons varies with season by examining synapsin-positive close contacts on A15 cells using immunofluorescence and confocal microscopy. The number of contacts on dendrites, but not somata, increased during anestrus. There were also corresponding changes in dendritic morphology including an increase in dendritic length, surface area, and number of bifurcations. Because thyroid hormones are necessary for the transition to anestrus, we next examined …

Development Of A Micro-Dilution Tunnel System For In-Use, On-Board Heavy-Duty Vehicle Particulate Matter Emissions Measurement, Emily Deanna Cirillo Aug 2001

Development Of A Micro-Dilution Tunnel System For In-Use, On-Board Heavy-Duty Vehicle Particulate Matter Emissions Measurement, Emily Deanna Cirillo

Graduate Theses, Dissertations, and Problem Reports (ETD)

A novel portable particulate matter measuring system was designed, developed and qualified. The heavy-duty engine Federal Test Procedure (FTP) requires the use of a full-flow tunnel to measure particulate emissions from diesel fueled engines. However, a full-flow tunnel based constant volume sampler (CVS) system is not only expensive to build and maintain, it also requires a large work space. The portable micro-dilution system that could be used for measuring on-board, in-use emissions from heavy-duty vehicles would cost significantly less. The micro-dilution tunnel operates on the same principle as a full-flow dilution tunnel. The added advantage of the proposed design is …

Functional Profiles Of Growth-Related Genes During Embryogenesis And Postnatal Development Of Chicken And Mouse Skeletal Muscle, Hakan Kocamis

Graduate Theses, Dissertations, and Problem Reports (ETD)

Reverse-transcription polymerase chain reaction (RT-PCR) was performed to measure myostatin, follistatin, activin-B gene expressions during chicken embryonic development. Strong myostatin expression was found in the early chicken embryos (E 0, E 1) and the developmental expression pattern of myostatin mRNA coincided with the periods of primary and secondary muscle fiber formation. Follistatin transcripts followed a linear expression pattern from E 0 to E 20, while activin-B had a quadratic pattern. The ontogeny of myostatin gene expression was nearly identical in satellite cells isolated from pectoralis major (PM) and biceps femoris (BF) muscles of chicken. Activin-B mRNA level in PM satellite …

Visual Iconic Object-Oriented Programming To Advance Computer Science Education And Novice Programming, Nancy Silva Martinez Aug 2001

Visual Iconic Object-Oriented Programming To Advance Computer Science Education And Novice Programming, Nancy Silva Martinez

Theses and Dissertations - UTB/UTPA

Learning how to program a computer is difficult for most people. Computer programming is a cognitively challenging, time consuming, labor intensive, and frustrating endeavor. Years of formal study and training are required to learn a programming language's world of algorithms and data structures. Instructions are coded in advance before the computer demonstrates the desired behavior. Seeing all the programming steps and instruction code is complicated. There exists a tremendous gap between the representations the human brain uses when thinking about a problem and the representations used in programming a computer. Often people are much better at dealing with specific, concrete …

The Hereford Mappa Mundi, a thirteenth-century world map, includes mention of fifty-four strange races. Many of the races can be found in three earlier sources: Pliny's Naturalis historia, Solinus's Collectanea rerum memorabilium, and Isidore's Etymologiae. By comparison to these three sources, the works used by the author of the map will be made clear.

This study provides an edition of all the inscriptions relating to these races, and compares them to excerpts relating to the races from the three above sources, as well as St. Augustine's De civitate Dei and Pomponius Mela's De chorographia. Translations …

The Hormesis Challenge For Environmental Health Regulators, Michael S. Baram Aug 2001

The Hormesis Challenge For Environmental Health Regulators, Michael S. Baram

Faculty Scholarship

The hormesis hypothesis provides that human, animal or plant exposure to a low external dose of a hazardous chemical substance or physical agent may have beneficial health outcomes'. For example, human exposure to a precisely defined, low external dose of a carcinogenic pollutant in the workplace or ambient environment may lead to an internal dose or concentration, which stimulates a dormant defense mechanism that inhibits the growth of tumors.

Development And Characterization Of Monoclonal Antibodies Directed Against Yaba-Like Disease Virus, Scott Haller Aug 2001

Development And Characterization Of Monoclonal Antibodies Directed Against Yaba-Like Disease Virus, Scott Haller

Masters Theses

The hybridoma technique has been used for the production of monoclonal antibodies (mAb) directed against yaba-like disease virus (YLDV). Spleen cells, obtained from female BALB/c mice immunized with YLDV, were fused with P3x63.Ag8.653 myeloma cells and hybrids were selected. Hybridoma clones were screened for production of anti-YLDV antibodies by Indirect Immunofluorescence (IF) microscopy. Positive hybridoma clones were recloned via limiting dilution and characterized. A single clone (H12) has been selected that specifically reacted with YLDV infected cells and not uninfected cells. Immunodiffusion technique determined H12 monoclonal antibody as an IgG1 isoptype immunoglobulin. H12 antibodies recognized a late YLDV protein, and …

Uptake and translocation of cationic nutrients play essential roles in physiological processes including plant growth, nutrition, signal transduction, and development. Approximately 5% of the Arabidopsis genome appears to encode membrane transport proteins. These proteins are classified in 46 unique families containing approximately 880 members. In addition, several hundred putative transporters have not yet been assigned to families. In this paper, we have analyzed the phylogenetic relationships of over 150 cation transport proteins. This analysis has focused on cation transporter gene families for which initial characterizations have been achieved for individual members, including potassium transporters and channels, sodium transporters, calcium antiporters, …

Sart, A Repressor Of Α-Hemolysin In Staphylococcus Aureus, Katherine A. Schmidt, Adhar C. Manna, Steven Gill, Ambrose L. Cheung

Dartmouth Scholarship

In searching the Staphylococcus aureus genome, we found several homologs to SarA. One of these genes, sarT, codes for a basic protein with 118 residues and a predicted molecular size of 16,096 Da. Northern blot analysis revealed that the expression of sarT was repressed by sarA and agr. An insertion sarT mutant generated in S. aureus RN6390 and 8325-4 backgrounds revealed minimal effect on the expression of sarR and sarA. The RNAIII level was notably increased in the sarT mutant, particularly in postexponential-phase cells, while the augmentative effect on RNAII was less. SarT repressed the expression of alpha-hemolysin, as determined …
