Analysis Of Mitotic Microtubule-Associated Proteins Using Mass Spectrometry Identifies Astrin, A Spindle-Associated Protein, Gary J. Mack, Duane A. Compton
Dartmouth Scholarship
We purified microtubules from a mammalian mitotic extract and obtained an amino acid sequence from each microtubule-associated protein by using mass spectrometry. Most of these proteins are known spindle-associated components with essential functional roles in spindle organization. We generated antibodies against a protein identified in this collection and refer to it as astrin because of its association with astral microtubule arrays assembled in vitro. Astrin is approximately 134 kDa, and except for a large predicted coiled-coil domain in its C-terminal region it lacks any known functional motifs. Maintaining rural community economic viability, schools, and retaining youth are concerns for many rural areas. The current population shift from rural to urban areas is compounding the issue. One means of keeping young people and adults in rural communities and encouraging people to move from urban to rural communities is to provide high quality, relevant, affordable educational programs on demand. The Mead Agricultural Sciences Magnet School, the first rural magnet school, was created to fill such a void. Now in its third year of operation, the rural agricultural sciences magnet school concept is being incorporated into three other communities.

Genetic Dissection Of Behavioral And Neurogenomic Responses To Acute Ethanol, Aaron Wolen

Individual differences in initial sensitivity to ethanol are strongly related to the heritable risk of alcoholism in humans. To elucidate key molecular networks that modulate ethanol sensitivity we performed a systems genetics analysis of ethanol-responsive gene expression in brain regions of the mesocorticolimbic reward circuit (prefrontal cortex, nucleus accumbens and ventral midbrain) across the BXD RI panel, a highly diverse family of isogenic mouse strains before and after treatment with ethanol. Acute ethanol altered the expression of ~2,750 genes in one or more regions and 400 transcripts were jointly modulated in all three. Ethanol-responsive gene networks were extracted with a …

The comparative analysis of homologous characters is a staple of evolutionary developmental biology and often involves extrapolating from experimental data in model organisms to infer developmental events in non-model organisms. In order to determine the general importance of data obtained in model organisms, it is critical to know how often and to what degree similar phenotypes expressed in different taxa are formed by divergent developmental processes. Both comparative studies of distantly related species and genetic analysis of closely related species indicate that many characters known to be homologous between taxa have diverged in their morphogenetic or gene regulatory underpinnings. Detection Of Salinity By The Lobster, Homarus Americanus, Winsor H. Watson Iii, Christopher G. Dufort, Steven H. Jury, James M. Newcomb

Changes in the heart rates of lobsters (Homarus americanus) were used as an indicator that the animals were capable of sensing a reduction in the salinity of the ambient seawater. The typical response to a gradual (1 to 2 ppt/min) reduction in salinity consisted of a rapid increase in heart rate at a mean threshold of 26.6 ± 0.7 ppt, followed by a reduction in heart rate when the salinity reached 22.1 ± 0.5 ppt. Animals with lesioned cardioregulatory nerves did not exhibit a cardiac response to changes in salinity. A cardiac response was elicited from lobsters exposed to isotonic …

Estrogen and progestin receptors (ER, PgR) play a critical role in the regulation of neuroendocrine functions in females. The neuroanatomical distribution of the recently cloned, ERβ, overlaps with both ERα and PgR. To determine whether ERβ is found within ERα- or PgR-containing neurons in female rat, we used dual label immunocytochemistry. ERβ-immunoreactivity (ERβ-ir) was primarily detected in the nuclei of cells in the periventricular preoptic area (PvPO), the bed nucleus of the stria terminalis (BNSTpr), the paraventricular nucleus, the supraoptic nucleus, and the medial amygdala (MEApd). The Farm Service Agency guaranteed loan program is an important source of credit for family-sized farming operations in Arkansas and the other states of the U.S. This program provides loan guarantees to borrowers who are otherwise unable to obtain credit from traditional lenders at reasonable rates and terms. This study identifies those factors related to the program’s loss claim rate performance over the years fiscal 1989 through 1998 using state-level data from forty states. For both the operating loan and farm ownership loan programs, farm operator financial variables, interest rates, and commercial bank characteristics are found to be statistically significant …
