Multidrug-Resistant Strains Of Salmonella Enterica Serotype Typhi Are Genetically Homogenous And Coexist With Antibiotic-Sensitive Strains As Distinct, Independent Clones, Kwai Lin Thong, Z.A. Bhutta, T. Pang

Objective: The goal of this study was to report the molecular analysis of antibiotic-sensitive and multidrug-resistant (MDR) strains of Salmonella typhi, using pulsed-field gel electrophoresis (PFGE), with a particular emphasis on the coexistence of these strains in a typhoid-endemic region of Karachi, Pakistan. Methods: One hundred isolates of S. typhi in humans (50 MDR and 50 antibiotic-sensitive isolates) from sporadic cases of typhoid fever were analyzed by Vi-phage typing, antibiograms and PFGE. Results: The MDR S. typhi strains were resistant to ampicillin, chloramphenicol, and trimethoprim-sulfamethoxazole. The objective of this research was to demonstrate the application of ultrasound to field sites having problems with precipitation build up and corrosion. PTW s are passive reactive walls containing zero-valent iron metal for in-situ remediation of contaminated groundwater. However, loss of reactivity over time due to build up of corrosion and other precipitates on the iron surface is a major concern. Ultrasound energy has been established as an effective tool for revitalizing iron surface. This research applied ultrasound energy to a zero-valent iron wall constructed below the ground surface to remove precipitates and iron corrosion, increasing iron reactivity. Two …

A. tumefaciensis a plant pathogen and causative agent of Crown Gall Disease, an ailment that results in the development of tumors in plants. It is a unique organism in that it is the only known non-viral pathogen that transfers its DNA into the host cell. Consequently, the majority of the research conducted on A. tumefaciensinvestigates its mechanism of infection. However, A. tumefaciensis also rare to the bacterial world in that its genome is comprised of two chromosomes: one circular and one linear. Most other bacteria only have one circular chromosome. The goal of this study is to identify the causes of membrane failure observed during a 15-month operation of a low pressure reverse osmosis (RO) membrane pilot plant to treat a highly organic surface water from the Hillsborough River in Tampa, Florida, using various surface analytical techniques. Three different commercial RO membranes, made of cellulose acetate or polyamide, were used in this pilot study, and all of these membranes showed performance deterioration presumably due to membrane fouling and degradation at given experimental conditions. Schwann cells (SCs) differentiate into a myelinating cell when simultaneously adhering to an axon destined for myelination and basal lamina. We are interested in defining the signaling pathway activated by basal lamina. Using SC/sensory neuron (N) cocultures, we identified beta 1 integrin and F-actin as components of a pathway leading to myelin gene expression and myelination (Fernandez-Valle et al., 1994, 1997). Here, we show that focal adhesion kinase (FAK) and paxillin are constitutively expressed by SCs contacting axons in the absence of basal lamina. Tyrosine phosphorylation of FAK and paxillin increases as SCs form basal lamina and differentiate. FAK and …

We performed feasibility tests of photoelectron emission spectromicroscopy of wet samples in the water window (285-532 eV) soft x-ray spectral region. Water was successfully confined in an ultrahigh vacuum compatible compartment with x-ray transparent sides. This water cell was placed in the MEPHISTO spectromicroscope in a transmission geometry, and complete x-ray absorption spectra of the water window region were acquired. We also show micrographs of test samples, mounted outside of the compartment, and imaged through the water. This technique can be used to study liquid chemistry and, at least to the micron level, the microstructure of wet samples. Possibilities include …

Several Plasmodium falciparum genes encoding cdc2-related protein kinases have been identified, but the modalities of their regulation remains largely unexplored. Zn the present study, we investigated the regulation in vitro of PfPK5, a putative homologue of Cdk1 (cdc2) in P. falciparum. We show that (i) PfPK5 is efficiently activated by heterologous (human) cyclin H and p25, a cyclin-like molecule that specifically activates human Cdk5; (ii) the activated enzyme can be inhibited by chemical Cdk inhibitors; (iii) Pfmrk, a putative P. falciparum homologue of the Cdk-activating kinase, does neither activate nor phosphorylate PfPK5; and (iv) PfPK5 is able to autophosphorylate in …

Evidence Of Scattering Anisotropy Effects On Boundary Conditions Of The Diffusion Equation, G. Popescu, C. Mujat, A. Dogariu

We present experimental evidence that the diffusion process close to the boundary is fundamentally different from the bulk diffusion in the sense that it is affected by the scattering properties of the individual centers. Systematic measurements show a nontrivial behavior of the extrapolation length ratio on the scattering anisotropy of individual particles. The results are compared with other works and are supported by an independent Monte Carlo simulation. A simple analytical model that emphasizes the influence of the last scattering event successfully describes the results.

Texture synthesis is the ability to create ensembles of images of similar structures from sample textures that have been photographed. The method we employ for texture synthesis is based on histogram matching of images at multiple scales and orientations. This paper reports two fast and in one case simple algorithms for histogram matching We show that the sort-matching and the optimal cumulative distribution function (CDF)-matching (OCM) algorithms provide high computational speed compared to that provided by the conventional approach. The sort-matching algorithm also provides exact histogram matching. Results of texture synthesis using either method show no subjective perceptual differences. The …

We reexamined geographic factors explaining the number of plant species on islands in the Galapagos Archipelago. We hypothesized that plant species richness (S) was related to the number of source pools and that plant species dispersal preferentially followed direct, oceanic pathways. To test different dispersal pathways from multiple source pools, the total number of islands within a given dispersal radius (i) was posed as the sum of the number of line-of-sight islands (C-i) and of the number of islands without line-of-sight connection (B-i). The mechanisms of transcriptional regulation are well conserved from yeast to human cells. Most genes are regulated at the transcriptional level, particularly, at the transcription initiation step. In Saccharomyces cerevisiae at least one transcriptional activator, ADR1, and one transcription complex, CCR4-NOT, participate in the activation of the ADH2 gene. The core 1 x 106 dalton CCR4-NOT complex consists of CCR4, CAF1, the five NOT proteins (NOT1-NOT5), and three other uncharacterized proteins. Deletion of NOT3, NOT4, or NOT5 did not affect the association of CCR4 with CAF1 in the gel filtration analysis. In contrast, NOT2 inactivation caused partial destabilization of the …

This exploratory study examined relations between frequency of Internet use among adults 65 and older and perceptions of social support and personal control. Additionally, standard multiple regression procedures were employed to assess the usefulness of frequency of Internet use, perceived social support, and perceived control as predictors of mental and physical health. Results were that frequency of Internet use was uncorrelated with perceptions of social support and personal control and was not predictive of mental or physical health. Perceived social support and perceived control emerged as predictors of mental and physical health. Recommendations for future research that refines measurement of …

Foliar nitrogen concentration represents a direct and primary link between carbon and nitrogen cycling in terrestrial ecosystems. Although foliar N is used by many ecosystem models to predict leaf-level photosynthetic rates, it has rarely been examined as a direct scalar to stand-level carbon gain. Significant improvements in remote sensing detector technology in the list decade now allow for improved landscape-level estimation of the biochemical attributes of forest ecosystems.

In this study, I examined the effects of ultraviolet radiation, highway contamination and a synergism of both factors on the spotted salamander, Ambystoma maculatum embryos in field and laboratory experiments. There are numerous reports of amphibian population declines from around the world. There are several possible factors causing the decline, including, habitat loss, environmental pollution, disease, and ultraviolet B radiation (UVB, 290--320 nm). Results of studies conducted to date seem to indicate that the impacts of these factors vary greatly among species and across geography. The exchange of carbon dioxide (CO2) between terrestrial ecosystems and the atmosphere plays a central role in the ecology of the biosphere and the climate system. Towards quantification of ecosystem-atmosphere CO 2 exchange, a generalized model of plant-soil-atmosphere CO2 exchange (OPTICAL) was described and evaluated using eddy covariance measurements of net ecosystem exchange of CO2 (NEE) in arctic, boreal, temperate, and tropical landscapes. The model requires no calibration and is based on theories of plant resource optimization and plant-soil nutrient feedbacks. The model predicts canopy photosynthetic capacity (Pcmax), canopy photosynthesis (P c), plant respiration (Rp), and soil heterotrophic respiration (RH). …
