Abstract

Background

PCR amplification of target molecules involves sequence specific primers that flank the region to be amplified. While this technique is generally routine, its applicability may not be sufficient to generate a desired target molecule from two separate regions involving intron /exon boundaries. For these situations, the generation of full-length complementary DNAs from two partial genomic clones becomes necessary for the family of low abundance genes.

Results

The first approach we used for the isolation of full-length cDNA from two known genomic clones of Hox genes was based on fusion PCR. Here we describe a simple and efficient method …

Background: Large gene expression studies, such as those conducted using DNA arrays, often provide millions of different pieces of data. To address the problem of analyzing such data, we describe a statistical method, which we have called ‘gene shaving’. The method identifies subsets of genes with coherent expression patterns and large variation across conditions. Gene shaving differs from hierarchical clustering and other widely used methods for analyzing gene expression studies in that genes may belong to more than one cluster, and the clustering may be supervised by an outcome measure. Helicobacter pylori infection is associated with a variety of clinical outcomes including gastric cancer and duodenal ulcer disease¹. The reasons for this variation are not clear, but the gastric physiological response is influenced by the severity and anatomical distribution of gastritis induced by H. pylori. Thus, individuals with gastritis predominantly localized to the antrum retain normal (or even high) acid secretion², whereas individuals with extensive corpus gastritis develop hypochlorhydria and gastric atrophy³, which are presumptive precursors of gastric cancer4. An expert panel was convened to evaluate the U.S. Environmental Protection Agency’s “Proposed Guidelines for Carcinogen Risk Assessment” through their application to data sets for chloroform (CHCl₃) and dichloroacetic acid (DCA). The panel also commented on perceived strengths and limitations encountered in applying the guidelines to these specific compounds. This latter aspect of the panel’s activities is the focus of this perspective. The panel was very enthusiastic about the evolution of these proposed guidelines, which represent a major step forward from earlier EPA guidance on cancer-risk assessment. These new guidelines provide the latitude to consider diverse scientific data …

Ethanol administration leads to altered function and impaired receptor-mediated endocytosis of the hepatocyte asialoglycoprotein receptor (ASGP-R). The purpose of the present study was to examine the effects of ethanol on the phosphorylation of the ASGP-R to determine whether this post-translational modification could contribute mechanistically to the observed ethanol-induced alterations. The methodological approach of this work involved the measurement of the phosphorylation state of the receptor obtained from isolated rat hepatocytes, using a combination of experimental designs from the biosynthetic incorporation of phosphate to the determination of steady-state phosphotyrosine levels. Since 1998, H3N2 viruses have caused epizootics of respiratory disease in pigs throughout the major swine production regions of the U.S. These outbreaks are remarkable because swine influenza in North America had previously been caused almost exclusively by H1N1 viruses. We sequenced the full-length protein coding regions of all eight RNA segments from four H3N2 viruses that we isolated from pigs in the Midwestern U.S. between March 1998 and March 1999, as well as from H3N2 viruses recovered from a piglet in Canada in January 1997 and from a pig in Colorado in 1977. The purpose of this study was to explore the need for a community grief support program for children, Nineteen professionals who work directly with children participated in this study. Data were collected utilizing an interview with a closed- and open-ended item questionnaire. Among the topics discussed were knowledge of existing grief support services, interventions to facilitate a child's adjustment to death and the need for a grief support program for children Findings indicated all of those who participated in the study felt a grief support program for children was needed. Participants provided useful information for the development of future programs. …

The purpose of this project was to develop a workshop that will help to secure culturally competent social services for African immigrant and refugee families. The framework of the workshop focused on two areas. The first is to help the workers to become culturally competent by carrying out ethnic sensitive inventory to assess their competency. Skills developed included what to do during pre-contact. during problem identification and specification, goal formation, problem solving and termination. Adolescents And Young Adults In Transition: From Homelessness To Permanent Housing, Tona L. Willand

This study explores the intrapersonal and environmental factors associated with change in the transition from homelessness to permanent housing from the perspective of young women who have experienced it. Three young women were interviewed face-to-face in semi-structured interviews, using a non-probability convenience sample. Content analysis was used to identify intrapersonal and environmental factors that affected the process of change. Research participants identified goal-directed behavior, persistence and hope as intrapersonal factors in effecting change. Supportive relationships and practical assistance were identified as supportive factors in the transition period. The findings are discussed in relation to research from the literature, and the …

This is a quantitative secondary data analysis which researched the effects of parental involvement on youth in foster care within a private non-profit treatment foster care agency. The study was intended to educate the agency and their foster parents on the significance of parental involvement with youth in foster eare to successfully achieve each youth's initial placement goal of reunification. According to current laws, reunification is considered the preferred permanency plan for foster youth. This case study poses the following question; does Greenleaf's servant- leadership theory provide a sufficiently comprehensive framework for effective leadership?

This study offers a review of literature that explores five key tenets of servant-leadership. These tenets include the exploration of the interconnections between leader, follower and institution, the idea that individuals are active agents in the creation of their life, the transformational nature of servant- leadership, the importance of a common purpose or vision and the use of persuasion as the primary means of influence.

Exposing The "Pretty Woman" Myth: A Qualitative Examination Of The Lives Of Female Streetwalking Prostitutes, Rochelle L. Dalla

Intensive interviews were conducted with 43 women involved in streetwalking prostitution. Data were analyzed according to Phenomenological Descriptive Analysis, results of which are presented in two parts. Detailed accounts of the lives of a subgroup of 5 participants are described first, followed by a broader discussion of results including the entire sample of 43. Themes common across the larger group are presented in three segments, including (a) early development, (b) life in "the game,” and (c) leaving the streets. Implications for advocacy and further research are presented.

Can intense preferences be accommodated in voting schemes without shifting power to wealthier citizens and organized interests? This article first situates the question within the larger issue of the inalienability of some legal rights, and then focuses on collective action problems among voters. These problems offer a way to explain our present rules and intuitions regarding vote buying and related matters in areas ranging from corporate law to associations and to campaign finance reform. But in large-scale general elections, collective action problems are likely to doom strategies for extracting information about intense preferences, and they may even produce perverse results. …
