Characterization Of Early Follicular Cdna Library Suggests Evidence For Genetic Polymorphisms In The Inbred Strain C108 Of Bombyx Mori, Marian R. Goldsmith, David R. Mills
Biological Sciences Faculty Publications
Recent work towards the completion of a saturated molecular genetic linkage map for the lepidopteran silkworm, Bombyx mori (n=28), has provided evidence for existing polymorphisms in the inbred strain C108. Two inbred parental strains, p50 and C108, were crossed to produce the F1 (P/C) hybrid offspring. The populations used in this project were comprised of a combination of 29 F2 (F1 x F1) and 31 reciprocal backcross (P/C x C/C, P/C x P/P) progeny. All restriction fragment length polymorphisms (RFLPs) for the initial analysis were hybridized with anonymous probes derived from a random early follicular cDNA (Rcf) library from Bombyx. …

Goldfish Brain Gap-43: Analysis By One And Two-Dimensional Gel Electrophoresis, Kendra Williams
McNair Scholars Journal
The concept of learning and memory is a neurological process believed to reside in the nervous system of living organisms. In particular; the growth-associated protein, GAP-43, has been shown to alter its state of phosphorylation via a protein kinase C mediated reaction following long-term potentiation, a paradigm of learning and memory. Most recent studies of GAP-43 in goldfish (Carassius auratus L.) hve focused on its presence in regenerating or developing optic nerves. The present study seeks to determine the existence of GAP-43 in the goldfish brain by Western blotting with commercially available antibodies and secondly, quantify phosphorylated isoforms of GAP-43 …

Apprenticeship And Conservation Incentives, Robin Alden, Jennifer F. Brewer
Geography
Apprentice programs offer a method to encourage responsible individual behavior by laying the foundation for successful collective property rights. Apprenticeship has three purposes: to restrict the rate of entry, to affect the quality of the participant, and to create the conditions for collective action for sustainability. Apprenticeship could be an important fishery management tool, particularly in decentralized, adaptive management regimes that require ongoing, multi-party negotiation for success. It is not vocational training; instead it serves a public purpose: to create the conditions for stewardship and participation in management. This perception of collective property right mimics customary practice in some successful …

The Internet is an exciting new territory for many young people. Nearly 24 million youth ages 10 through 17 were online regularly in 1999, and millions more are expected to join them shortly. They go there to Iearn, play, meet people, and explore the world. But stories from law-enforcement officials, parents, and young people themselves suggest that not every online adventure is a happy one. The Internet has a seamier side that young people seem to he encountering with great frequency.
This national survey confirms many of the stories. Large numbers of young people are encountering sexual solicitations they did …
Rendering Multispectral Data As Useful 'Super-Visual' Images, Matthew Gypson
Rendering Multispectral Data As Useful 'Super-Visual' Images, Matthew Gypson
Theses
Due to the nature of mulitspectral images, it is difficult to present all available information in a single image. This study addresses the need for data compression, to enable faster and easier evaluation of a subject containing important details in a wide spectral range. The specific aim of this research is to construct algorithms to in effect compress sampled wide-band electromagnetic data into the visible range. This will present maximum image details within a final color image. The developed compression scheme aims to achieve the high efficiency while keeping the best quality of image subjects. Ancient documents (from the Dead …

In contrast to the single metallothionein (MT)-1 gene of the mouse, the human MT-1 gene family is composed of seven active genes and six pseudogenes. In this study, the expression of mRNA representing the seven active human MT-1 genes was determined in cultured human proximal tubule (HPT) cells under basal conditions and after exposure to the metals Cd2+, Zn2+, Cu2+, Hg2+, Ag2+, and Pb2+. Basal expression of MT-1X and MT-1E mRNA in HPT cells was similar to expression of the housekeeping gene glyceraldehyde 3-phosphate dehydrogenase. In contrast, mRNAs representing the basal expression of MT-1A and MT-1F were a minor transcript …

Acute Silica Toxicity: Attenuation By Amiodarone-Induced Pulmonary Phospholipidosis., J M. Antonini, C M. Mccloud, M J. Reasor
Faculty & Staff Scholarship
Exposure to the toxic mineral dust silica has been shown to produce an acute inflammatory response in the lungs of both humans and laboratory animals. Coating silica with phospholipids reduces its toxicity when studied with in vitro systems. The drug amiodarone increases phospholipid within the cells, airways, and alveoli of the lungs. This increase in phospholipid is due to amiodarone's ability to inhibit phospholipase activity within alveolar macrophages (AMs) and whole lung. The purpose of this study was to determine whether the amiodarone-induced increase in pulmonary phospholipid would protect the lungs from acute damage caused by the intratracheal instillation of …

Evaluating The Potential Of Using 5-Azacytidine As An Epimutagen, Mary Ann Fieldes, L.M. Amyot
Biology Faculty Publications
A number of early flowering lines were induced when 5-azacytidine was applied to germinating flax (Linum usitatissimum L.) seed. The genetics of these lines indicate that the induced changes are epigenetic and probably result from demethylation of the genomic DNA at loci that affect flowering age. Although the growth and development of three stable early flowering lines are altered and the percentage of filled seed was reduced in all three lines compared with controls, measures of seed productivity demonstrated that harvest index was unaffected in two of the lines. In the third, harvest index was lower than normal and …

Isolation Of Current Components And Partial Reaction Cycles In The Glial Glutamate Transporter Eaat2, Thomas S. Otis, Michael Kavanaugh
Biomedical and Pharmaceutical Sciences Faculty Publications
The kinetic properties of the excitatory amino acid transporter EAAT2 were studied using rapid applications of l-glutamate to outside-out patches excised from transfected human embryonic kidney 293 cells. In the presence of the highly permeant anion SCN−, pulses of glutamate rapidly activated transient anion channel currents mediated by the transporter. In the presence of the impermeant anion gluconate, glutamate pulses activated smaller currents predicted to result from stoichiometric flux of cotransported ions. Both anion and stoichiometric currents displayed similar kinetics, suggesting that anion channel gating and stoichiometric charge movements are linked to early transitions in the transport cycle. …

Arginine 447 Plays A Pivotal Role In Substrate Interactions In A Neuronal Glutamate Transporter, Annie Bendahan, Ayelet Armon, Navid Madani, Michael Kavanaugh, Baruch I. Kanner
Biomedical and Pharmaceutical Sciences Faculty Publications
Glutamate transporters from the central nervous system play a crucial role in the clearance of the transmitter from the synaptic cleft. Glutamate is cotransported with sodium ions, and the electrogenic translocation cycle is completed by countertransport of potassium. Mutants that cannot interact with potassium are only capable of catalyzing electroneutral exchange. Here we identify a residue involved in controlling substrate recognition in the neuronal transporter EAAC-1 that transports acidic amino acids as well as cysteine. When arginine 447, a residue conserved in all glutamate transporters, is replaced by cysteine, transport of glutamate or aspartate is abolished, but sodium-dependent cysteine transport …
