Interspecies transmission of the transmissible spongiform encephalopathies (TSEs), or prion diseases, can result in the adaptation and selection of TSE strains with an expanded host range and increased virulence such as in the case of bovine spongiform encephalopathy and variant Creutzfeldt-Jakob disease. To investigate TSE strain adaptation, we serially passaged a biological clone of transmissible mink encephalopathy (TME) into Syrian golden hamsters and examined the selection of distinct strain phenotypes and conformations of the disease-specific isoform of the prion protein (PrP^Sc). Parasites of the fathead minnow, Pimephales promelas, were examined in fish collected from Elk Creek (40.88534°N, 96.83366°W) and West Oak Creek (40.90821°N, 96.81432°W), Lancaster County, Nebraska. These two streams are part of the Salt Valley watershed and flow together approximately 2 km downstream from the collection sites to form Oak Creek. This study examined the extent to which the two tributaries constitute a continuous habitat with respect to fish hosts. Eugregarines of the suborder Septatorina are apicomplexan parasites that are found mainly in arthropods. Some exceptions are species in the Metameridae that contains the only 5 septate gregarines recorded from annelids. The type genus is Metamera Duke, 1910 with two species, Metamera schubergi Duke, 1910, in European Glossiphonia complanata, and Metamera reynoldsi Jones, 1943, from North American G. complanata. Over the summers of 1995-1998, in Keith County, Nebraska, septate gregarines were found in the glossiphoniid leech Helobdella triserialis. The gregarines were determined to be a new species of Metamera, herein named Metamera sillasenorum. Measurements of size …

With the imminent publication of the Nebraska Breeding Bird Atlas and the turning of a new millennium, it is perhaps an appropriate time to survey the state of breeding birds in Nebraska. Wayne Mollhoff’s summary of the N.O.U.’s Nebraska Breeding Birds Aliasing Project (Mollhoff, 2000) provides important databases for the latter part of the past century, and the historic overview by James Ducey (1988) offers a useful basis for judging the breeding avifauna of Nebraska from about the beginning of the century. 1. We previously showed that actin is transported in an unassembled form with its associated proteins actin depolymerizing factor, cofilin, and profilin. Here we examine the specific activities of radioactively labeled tubulin and neurofilament proteins in subcellular fractions of the chicken sciatic nerve following injection of L-[35S]methionine into the lumbar spinal cord. 2. At intervals of 12 and 20 days after injection, nerves were cut into 1-cm segments and separated into Triton X-100-soluble and particulate fractions. Analysis of the fractions by high-resolution two-dimensional gel electrophoresis, immunoblotting, fluorography, and computer densitometry showed that tubulin was transported as a unimodal wave at …

Filarioid nematodes of the genus Litomosoides occur in the abdominal and (or) thoracic cavities of marsupials, rodents, and bats of the Nearctic and Neotropical regions. In this study, the phylogenetic relationships among these nematodes were estimated with a parsimony analysis of morphological characters derived from species descriptions. This nonweighted analysis produced 20 shortest trees. The monophyly of the genus was not supported in that Litomosoides thomomydis and Litomosoides westi failed to group with the other members of the genus. When these 2 taxa (parasites of pocket gophers) were excluded, monophyly of Litomosoides was supported by 2 synapomorphies (structure of the …

Nuclear ribosomal sequence data from the internal transcribed spacers (ITS-1 and ITS-2), 5.8S subunit, and regions of the 18S and 28S genes were used to investigate sequence diversity among geographic samples of Nematodirus battus, and to infer phylogenetic relationships among Nematodirus species. Phylogenetic analysis of these data yielded strong support for relationships among species, depicting Nematodirus helvetianus and Nematodirus spathiger as sister-taxa and a clade of these two species and Nematodirus filicollis. This tree is consistent with caprine bovids as ancestral hosts, with a subsequent host shift to Bovinae in N. helvetianus. Eleven of 14 N. battus …

Abstract Both body iron stores and dietary iron intake have been reported to increase risk of colorectal neoplasms. We assessed whether serum ferritin concentration was associated with recurrence of colorectal adenomas among 733 individuals with baseline determinations of ferritin as part of a multicenter clinical trial of antioxidant supplements for adenoma prevention. All study participants had at least one adenoma removed within 3 months before enrollment, and 269 of them developed one or more adenomas between follow-up colonoscopies conducted 1 and 4 years after enrollment. This research examined the relationships between attachment style and frequency o f suicidal ideation, urges to self-harm, and action to self-harm in Dialectical Behavioral Therapy patients. The Attachment Style Questionnaire served as the attachment measure. The Diary Cards, used in Dialectical Behavioral Therapy, served as the measure for suicide ideation, urges to self-harm and action to self-harm averages. Twenty women from the Omaha YWCA and Therapy Resource Associates participated in the study. Results o f the study suggest that there was no relationship between a high score on Avoidant and Anxious/Ambivalent attachment styles and frequency of suicide ideation, urges to …

Among the most common critiques of globalization is that it increasingly constrains the ability of democratic communities to make unfettered choices about policies that affect the fundamental welfare of their citizens, including those of health and safety, the environment, and consumer protection. Traditionally, free trade rules were about constraining border measures such as tariffs and quantitative restrictions on imports. In 1993, the Environmental Protection Agency and the U.S. Army Corps of Engineers adopted a new regulation, known as the Tulloch Rule, based on a 1992 settlement between certain environmental groups and a developer who had engaged in dredging activities that fell outside of the Clean Water Act's section 404 protection. The Tulloch Rule in essence closed a loophole in the Clean Water Act that developers had been taking advantage of for years. The FDA Food Quality Protection Act of 1996 and amendments to the EPA Safe Drinking Water Act require the EPA Administrator, in consultation with the Secretary of Health and Human Services, to develop a screening program for endocrine disrupting effects. This Comment explores the science of endocrine disruptors and examines whether the current science supports a successful products liability claim. The various methods for proving liability, causation and harm are presented and analyzed against the current science of endocrine disruption. More than 130 different mutations in the gap junction integral plasma membrane protein connexin32 (Cx32) have been linked to the human peripheral neuropathy X-linked Charcot–Marie–Tooth disease (CMTX). How these various mutants are processed by the cell and the mechanism(s) by which they cause CMTX are unknown. To address these issues, we have studied the intracellular transport, assembly, and degradation of three CMTX-linked Cx32 mutants stably expressed in PC12 cells. Each mutant had a distinct fate: E208K Cx32 appeared to be retained in the endoplasmic reticulum (ER), whereas both the E186K and R142W mutants were transported to perinuclear compartments from which …

Telomerase is a ribonucleoprotein enzyme that is able to add telomeric repeats to the ends of eukaryotic DNA to stabilize chromosomes. Telomerase is active in germline cells, stem cells and tumor cells, but not in most human somatic cells. In this study we report the extended use of the Telomeric Repeat Amplification Protocol (TRAP) assay for the detection of telomerase activity in prostate needle biopsy specimens and freshly collected prostatic fluid. Clinical application of a newly developed PCR-ELISA telomerase assay method is also evaluated. Aims: To carry out an audit ascertaining the importance of condition of lymph node specimen, submission of clinical history including site of biopsy and imrnunohistochemcial studies on conclusiveness of diagnosis made.

Methodology: Computer records of the Aga Khan University Hospital, Histopathology Laboratory were used to analyze all cases of lymphoproliferative malignancies presented at the hospital from 1992 to 1998.

Results: Out of a total of 466 cases studied, in 283 (61%) the lymph nodes were fragmented. The site of biopsy was mentioned in 361 (77.5%) cases with the cervical region forming the most common site (56.5%). A clinical history was …

The purpose of this study was to examine how including alternative assessments in a reform-based mathematics course affects pre-service teachers’ mathematical beliefs. A single section of a mathematics course designed for elementary education majors that employed three different alternative assessments was the setting for the study.

A pre- and post-belief survey completed by students enrolled in the section under study represented the first level of data collection and analysis. The results of the pre-belief survey guided the selection of seven informants from the section and provided belief statements for the informants to verify during subsequent interviews. Nitrogen uptake and cycling was examined using a six‐week tracer addition of 15N‐labeled ammonium in early spring in Walker Branch, a first‐order deciduous forest stream in eastern Tennessee. Prior to the 15N addition, standing stocks of N were determined for the major biomass compartments. During and after the addition, 15N was measured in water and in dominant biomass compartments upstream and at several locations downstream. Residence time of ammonium in stream water (5–6 min) and ammonium uptake lengths (23–27 m) were short and relatively constant during the addition. These experiments were performed to test the hypotheses that myosin light chain 17 (MLC₁₇) a and b isoform expression varies between individual vascular smooth muscle (SM) cells and that their expression correlates with cell unloaded shortening velocity. Single SM cells isolated from rabbit aorta and carotid arteries were used to measure unloaded shortening velocity and subsequently were analyzed via RT-PCR for MLC₁₇ a and b mRNA ratio. The MLC_17b/a mRNA and protein ratios from adjacent tissue sections correlate very well (R² = 0.68), allowing use of the mRNA ratio to predict the protein ratio. …
