Mutational Analysis Of The Tra Locus Of The Broad-Host-Range Streptomyces Plasmid Pij101, Gregg S. Pettis, Stanley N. Cohen

The tra gene of Streptomyces lividans plasmid pIJ101 encodes a 621- amino-acid protein that can mediate both plasmid transfer and the interbacterial transfer of chromosomal genes (i.e., chromosome-mobilizing ability [Cma]) during mating. Here we report the results of in-frame insertional mutagenesis studies aimed at defining regions of Tra required for these functions. While hexameric linker insertions throughout the tra gene affected plasmid and chromosomal gene transfer, insertions in a 200-amino- acid region of the Tra protein that contains presumed nucleotide-binding motifs and that is widely conserved among a functionally diverse family of bacterial and plasmid proteins (K. Alkenones produced by the haptophyte algae are currently being used as indices of sea surface temperature in recent and past ocean environments, but limited information is available concerning the impact of biotic and abiotic processes on the integrity of these long chain lipids. This synthesis provides selected background information on major alteration processes that must be considered before such indices can be used with confidence. A number of processes in the water column and surface sediments have the potential to impact the structural integrity of alkenones and compromise their ability as temperature markers. Mass spectrometry (MS) promises to be an invaluable tool for functional genomics, by supporting low-cost, high-throughput experiments. However, large-scale MS faces the potential problem of mass degeneracy -- indistinguishable masses for multiple biopolymer fragments (e.g. from a limited proteolytic digest). This paper studies the tasks of planning and interpreting MS experiments that use selective isotopic labeling, thereby substantially reducing potential mass degeneracy. Our algorithms support an experimental-computational protocol called Structure-Activity Relation by Mass Spectrometry (SAR by MS), for elucidating the function of protein-DNA and protein-protein complexes. The nuclear factor-КB (NF-КB)/Rel family of inducible dimeric transcription factors regulates numerous genes associated with the immune, mitogenic, and apoptotic responses. Investigations were made into the role of NF-КB in breast carcinoma (MCF-7, T47D, MDA-MB-231) and glioblastoma (U87MG, U373MG, Ul 18MG) cell lines. NF-КB binding to DNA was quantified using both gel mobility shift assays and transient transfections of a vector containing a luciferase gene under the control of 4 tandem copies of the NF-КB binding sequence. An evaluation of the training and its effects is needed to answer the questions about the usefulness, and utility of the training program. Training is typically assessed using some combination of the criteria presented in Kirkpatrick's hierarchical model (Noe & Schmitt, 1986). 1 Regeneration in forest canopy gaps is thought to lead invariably to the rapid recruitment and growth of trees and the redevelopment of the canopy. Our observations, however, suggest that an alternate successional pathway is also likely, whereby gap‐phase regeneration is dominated by lianas and stalled in a low‐canopy state for many years. We investigated gap‐phase regeneration in an old‐growth tropical forest on Barro Colorado Island (BCI) in Panama to test the following two hypotheses: (i) many gaps follow a pathway in which they remain at a low canopy height and are dominated by lianas and (ii) the paucity of …

This thesis describes the isolation and characterization of a region containing several genes involved in metal tolerance in the common soil bacterium, Pseudomonas fluorescens ATCC 13525. The DNA region was identified in a cadmium and zinc sensitive mutant, 45-B10, which contains a lacZ-based reporter gene transposon Tn5-B20 inserted in the chromosome. The DNA surrounding the transposon was cloned from this mutant strain and DNA sequencing of this region revealed seven genes that share similarity with genes whose products are involved in the cation-proton antiporter efflux system of other Gram-negative bacteria. The Arabidopsis AINTEGUMENTA (ANT) gene has been shown previously to be involved in ovule development and in the initiation and growth of floral organs. Here, we show that ANT acts in additional processes during flower development, including repression of AGAMOUS (AG) in second whorl cells, promotion of petal epidermal cell identity, and gynoecium development. Analyses of ap2-1 ant-6 double mutants reveal that ANT acts redundantly with AP2 to repress AG in second whorl cells. The abaxial surface of ant petals contains features such as stomata and elongated, interdigitated cells that are not present on wild-type petals. …

The primary purpose of this study was to examine the effect of biculturalism on depression in a propsed sample of 52 Northern Plains Native American College students (32 female, 20 male). This study tested the applicability of the Orthogonal Theory of Biculturalism (Oetting and Beauvais, 1992). Subjects completed the Northern Plains Biculturalism scale (NPBI) and the Beck Depression Inventory-Second edition (BDI-II). A descriptive analysis, Pearson product moment correlational analysis, independent t-tests, and multiple regression analysis were performed. According to the response style theory of depression (Nolen-Hoeksema, 1987), the way in which individuals respond to depressed mood affects the severity and duration of that mood. Prior laboratory studies of response style have not included psychophysiological measures or investigated the relationship between ruminative response style and the conceptually-related constructs of worry and neuroticism. Dysphoric (n = 84) and nondysphoric participants (n = 86) were randomly assigned to either a rumination task (i.e., self-focused attention) or a distraction task (i.e., other-focused attention). Pseudomonas syringae is a member of an important group of Gram-negative bacterial pathogens of plants and animals that depend on a type III secretion system to inject virulence effector proteins into host cells. In P. syringae, hrpyhrc genes encode the Hrp (type III secretion) system, and avirulence (avr) and Hrpdependent outer protein (hop) genes encode effector proteins. Reaction Of Caffeic Acid With Amino Acids, Yonggang Wang

The reaction of caffeic acid with tryptophan was studied as a model for the reaction of caffeic acid with amino acids in the aquatic environment. Although the autoxidation reaction of caffeic acid in aqueous solution with subsequent dimerization had been previously studied, it was examined as well for comparison purposes. The present studies demonstrate that oxidized caffeic acid would react with tryptophan. The reaction, like the dimerization reaction, was catalyzed by the presence of iron(III) ions at low pH ( <4) and by hydroxide ions at high pH (10). The reaction of caffeic acid with itself yielded six products, four of which were identified by mass spectroscopy as dimers, with the other two being trimers. The reaction of caffeic acid with tryptophan yielded three product peaks. Two of the product peaks had similar retention times to the dimers from the self-reaction of caffeic acid; the third was a new product resulting from the reaction of caffeic acid with the tryptophan.

A mechanism for the reaction of caffeic acid with tryptophan has been proposed. This mechanism was based on the analysis …

This research examines the rhetoric of belle epoque English and French middle class social observers. Addressing the "social question," they engaged in a debate on how best to appease an impoverished, alienated, and increasingly militant working class. Historians cite this discourse, and the meager legislation it fostered, as a transition from the unbridled nineteenth century laissez-faire to the welfare democracy characteristic of the twentieth century.

Central to the "social question" was the issue of improving social relations without altering existing class hierarchies (which favored the middle classes). Many social commentators shunned the passage of legislative safety nets in favor of …

Advocates of a self-preserving soul must somehow integrate that soul with an organic body which is the product of an evolutionary history. Efforts to resolve this version of the sorites paradox (at what point in the gradual evolution of Homo sapiens was the step taken from the soul-less to the ensouled?) can result in conflict between the two theories. The position is taken here that, given the standard of Popperian verisimilitude, any such conflict signals a weakness in the soul-theory, and not in evolutionism.

Theoretically, a paradigm shift in the 1960s made community-based care and out-patient treatment an acceptable alternative to institutionalization of the chronically mentally ill. Utilizing the Levenson Multidimensional Locus of Control Scale for Psychiatric Patients, the author conducted a causal-comparative study to determine if a link exists between having bipolar disorder, a chronic mental illness, and an external locus of control, a link which might explain the inability of this population to maintain their optimal level of mental health utilizing the out-patient treatment model. High-molecular-weight polynuclear aromatic hydrocarbons (PAHs) are persistent in the environment although a wide variety of microorganisms can metabolize PAHs. In the past decades, laboratory and filed studies have shown that PAH degradation in soil is often limited by poor bioavailability and oxygen availability. Bound residue formation of PAHs with macromolecules of soil organic matter is an important fate mechanism. More recently, phytoremediation for PAH-contaminated soils is being explored. A thorough review of relevant publications, as well as intercept surveys, content analyses and email surveys, was conducted to test the hypotheses.

Fifty intercept surveys were conducted over two weekends to discover opinions about the issue. Respondents appearing to be age 25 or older were randomly selected. It is increasingly clear that the packaging of DNA in nucleosome arrays serves not only to constrain the genome within the nucleus, but also to encode information concerning the activity state of the gene. Packaging limits the accessibility of many regulatory DNA sequence elements and is functionally significant in the control of transcription, replication, repair and recombination. Aspartate transcarbamoylase (ATCase) catalyzes the first step in the pyrimidine biosynthetic pathway, the reaction between carbamoyl phosphate and L-aspartate to form N-carbamoyl-L-aspartate and phosphate. The structural analysis of the ATCase catalytic trimer from Methanococcus jannaschii, a unicellular thermophilic archaeabacterium, has been undertaken in order to gain insight into the structural features that are responsible for the thermostability of the enzyme. As a first step, the catalytic trimer was crystallized in space group R32, with unit-cell parameters a = b = 265.3, c = 195.5 Å and two trimers in the asymmetric unit. On December 22, Congress authorized the Flood Control Act of 1944, later named the Pick-Sloan Missouri Basin Program (PSMBP). The primary purpose of the PSMBP, which would require the construction of six main stem dams on the Missouri River, was to provide flood control, navigation, irrigation, and hydropower. The Mandan, Hidatsa, and Arikara nations, the Three Affiliated Tribes, maintained a reservation and way of life on Fort Berthold which lay in the direct path of the water that would form the reservoir behind one of the dams, the Garrison Dam.
