Uncharged Trna Activates Gcn2 By Displacing The Protein Kinase Moiety From A Bipartite Trna-Binding Domain, Jinsheng Dong, Hongfang Qiu, Minerva Garcia-Barrio, James T. Anderson, Alan G. Hinnebusch

Protein kinase GCN2 regulates translation in amino acid–starved cells by phosphorylating eIF2. GCN2 contains a regulatory domain related to histidyl-tRNA synthetase (HisRS) postulated to bind multiple deacylated tRNAs as a general sensor of starvation. In accordance with this model, GCN2 bound several deacylated tRNAs with similar affinities, and aminoacylation of tRNAPhe weakened its interaction with GCN2. Unexpectedly, the C-terminal ribosome binding segment of GCN2 (C-term) was required in addition to the HisRS domain for strong tRNA binding. A combined HisRS+C-term segment bound to the isolated protein kinase (PK) domain in vitro, and tRNA impeded this interaction. An activating mutation …

This article discusses how data warehousing, data mining, and decision support systems can reduce the national cancer burden or the oral complications of cancer therapies, especially as related to oral and pharyngeal cancers. An information system is presented that will deliver the necessary information technology to clinical, administrative, and policy researchers and analysts in an effective and efficient manner. The system will deliver the technology and knowledge that users need to readily: (1) organize relevant claims data, (2) detect cancer patterns in general and special populations, (3) formulate models that explain the patterns, and (4) evaluate the efficacy of specified …

Methicillin resistant Staphylococcus aureus (MRSA) is a pathogen first emerging in Great Britian in 1961. Since that time, it has spread worldwide and caused epidemic infection rates in several countries. It is most commonly spread in the healthcare setting where it is labeled a "nosocomial" infection, frequently infecting patients undergoing invasive medical procedures and immune-compromised patients. Depending on the severity and location of infection in conjunction with the patient's health status, MRSA infection can be mild to fatal. The surgical setting is a key area of concern. Solid phase microextraction (SPME) is an effective method for concentrating organic compounds prior to analysis by a variety of detection methods, including Fourier transform infrared spectroscopy (FTIR) and gas chromatography (GC). The first part of this work focused on the selection of an infrared-compatible solid phase that would partition phenols out of aqueous solutions. The criteria we employed dictated that potential solid phases be polar yet insoluble in water, have open regions in their IR spectra between 400-1200 cm·', 3010-3095 cm·', or 3200-3650 cm·' and be less than 200 μm thick.

This paper discusses the role of public policy in the skills development system of the U.S. It further examines the implications of that policy for the skill development and career progression of black workers. The paper describes the current "system" for skills development in the United States as a two- tiered system: The "first-chance" or conventional system allows individuals to proceed through an extensive public elementary, secondary, and postsecondary educational sector that is supplemented by private educational institutions and is followed by employer-provided job training and work experience. The "second-chance" system is designed for individuals who do not successfully traverse …

Noncompliance with parent requests is a commonly reported behavior problem for clinic-based child referrals. Parent skills training has been empirically demonstrated as an effective treatment for helping parents increase positive child behaviors and decrease negative behaviors. Unfortunately, many therapists and agencies are unable to implement parent-training programs due to limited therapist training, limited economic resources, or long waiting lists.
Adjunctive, computer-based, treatment programs have become increasingly popular in both research and clinical settings for facilitating psychological assessment and intervention. Reuse of engineering thermoplastics retrieved from discarded computers, monitors and printers called as electronic shredder residue is described in the following thesis. Virgin, blend of virgin and recycled, and 100% recycled polymers such as acrylonitrile butadiene styrene (ABS) and polycarbonate (PC) are characterized for mechanical properties as part of this research. This study used an inexpensive and versatile environmental exposure system to test the hypothesis that hypoxia promoted free radical production in primary cultures of rat main pulmonary artery smooth muscle cells (PASMCs). Production of reactive species was detected by fluorescence microscopy with the probe 2', 7'-dichlorodihydrofluorescein, which is converted to the fluorescent dichlorofluorescein (DCF) in the presence of various oxidants. Flushing the airspace above the PASMC cultures with normoxic gas (20% O(2), 75% N(2), and 5% CO(2)) resulted in stable PO(2) values of approximately 150 Torr, whereas perfusion of the airspace with hypoxic gas (0% O(2), 95% N(2), and 5% …

Erythromycin has long been recognized for its ability to inhibit protein synthesis by interfering with mRNA translation on the bacterial ribosome. We have recently shown that erythromycin also inhibits the assembly of the 50S ribosomal subunit in growing bacterial cells. The nature of this assembly inhibition has been investigated using 3H-uridine pulse-chase labeling of control and erythromycin treated E. coli cells.
Subunit assembly was examined by sucrose gradient centrifugation of labeled cell lysates. Normal assembly kinetics of subunit assembly were observed in control cells at 37°C. Formation of the 30S subunit was completed by 7.5 minutes and assembly of …

This study examined adults’ perceptions of aggression in toys and toy commercials targeted toward young boys. The subjects, 262 undergraduate psychology students, completed the Buss/Perry Aggression Questionnaire and rated either a videotape of 10 boy-oriented toy commercials or slides of 10 non-advertised boy-toys. Parental status, exposure to an advertised versus non-advertised toy, and level of self-reported aggression were assessed to determine their relationship to perceptions of aggressiveness in a toy’s image and predicted play with the toy.
Univariate analyses of variance and paired sample t-tests were conducted. The results indicate that toys advertised in commercials are judged to portray more …

The 1999 PLA Conference, which was hosted jointly by the American Enterprise Institute and the U.S. Army War College's Strategic Studies Institute, convened September 10-12, 1999, at Carlisle Barracks, Pennsylvania. The goal of this conference was to comprehensively examine Chinese military modernization efforts. The meeting drew together leading experts on the PLA army, navy, air force, missile forces, and national defense industries and included PLA experts with opposing views on the pace and likely success of Chinese military modernization. Turnip crinkle virus (TCV) is a single-stranded positive-sense RNA virus of the Carmovirus genus. Two of its five open reading frames (ORFs), encoding proteins of 8 and 9 kDa, are required for cell-to-cell movement of the virus. These movement proteins (MPs) were fused to green fluorescent protein (GFP) to determine their cellular localization. In protoplasts, p9-GFP, like GFP itself, is found throughout the cytoplasm, as well as in cell nuclei. In contrast, p8-GFP was confined to the cell nucleus. Similar localization patterns were observed when specific small peptide epitopes were fused to p8 and p9 proteins instead of GFP. The …

OBJECTIVE: The association between cancer and thromboembolic disease has been known for over a century. Increased tissue factor expression by endothelial cells, monocytes or macrophages is implicated. Thus, monocyte tissue factor measurements may reflect disease presence or progression. METHODS: Using a 2 stage kinetic chromogenic assay, monocyte tissue factor levels were assessed in normal controls (n=60), patient controls (hernia or cholecystectomy, n=60) and in patients with benign and malignant disease of the bladder (n=73), prostate (n=81), breast (n=83) and colorectum (n=62). OBJECTIVE: To determine the age, sex and relative frequencies of various gastrointestinal malignancies in patients registered with the National Cancer Registry from the Western Region of Saudi Arabia from January 1994 till December 1997, and compare this data with previous hospital based studies about the pattern of these malignancies in Saudi Arabia.
