Abstract

Background

PCR amplification of target molecules involves sequence specific primers that flank the region to be amplified. While this technique is generally routine, its applicability may not be sufficient to generate a desired target molecule from two separate regions involving intron /exon boundaries. For these situations, the generation of full-length complementary DNAs from two partial genomic clones becomes necessary for the family of low abundance genes.

Results

The first approach we used for the isolation of full-length cDNA from two known genomic clones of Hox genes was based on fusion PCR. Here we describe a simple and efficient method …

Afghan After A Fashion: The Fusion Of Politics With Religion And Women's Textile Craft Economies, M. Catherine Daly Jan 2000

Afghan After A Fashion: The Fusion Of Politics With Religion And Women's Textile Craft Economies, M. Catherine Daly

Textile Society of America: Symposium Proceedings

Afghan men and women have been restricted in their use of traditional and contemporary items of textiles during several "jihad" periods of Afghan history, first during the Mujahedeen period of the late 1970's and since the mid-1990's by the Taleban Islamic Movement of Afghanistan (Daly, 1998b).2 "Use" during either period reflects varying viewpoints that govern not only the wearing of textiles but also involvement in the textile craft economies. In either circumstance textile craft economies have economically supported both men and women (Daly, 1999b). Most "outsiders" presume that the strict measures imposed by first the Mujahedeen then followed by …

Discussion Of Recent Approaches To Expanding Textile Studies, Patricia A. Cunningham Jan 2000

Discussion Of Recent Approaches To Expanding Textile Studies, Patricia A. Cunningham

Textile Society of America: Symposium Proceedings

We are all committed to extending the knowledge of textiles. These papers offer outstanding examples of how museum curators, an academic, and a conservator have expanded audiences and brought new materials to telling the textile story. Major themes have emerged from their papers, and are worthy of further consideration.

First, it is quite apparent that professionals in museums need to work together. Harold Mailand has shown what the conservator can bring to the curator in the interpretation of textiles. I contend that textile department and costume departments should work together, and collaborate with other departments in their institutions. I am …

Reinventing A Cultural Self: Textile Design And Native American Youth, Gloria Gonzalez-Kruger, Wendy Weiss, Mary Lee Johns

Textile Society of America: Symposium Proceedings

This paper presents a description of a textile based program for Native American youth and an investigation that resulted from a community-university collaborative effort. This paper can help inform practice, academic, and policy related professionals who are interested in developing, implementing, or evaluating youth programming and culturally-based programs. The purpose of the project was threefold: 1) to involve youth from the Indian Center, Inc. of Lincoln in the design and creation of an art project, which was installed at the Daywatch facility in Lincoln, Nebraska, 2) to examine how the cultural knowledge, perceptions, beliefs, and identity of youth from the …

Resurgent Malaria At The Millennium Control Strategies In Crisis, J. Kevin Baird Jan 2000

Resurgent Malaria At The Millennium Control Strategies In Crisis, J. Kevin Baird

United States Public Health Resources

Completion of the Panama Canal in 1914 marked the beginning of an era of vector control that achieved conspicuous success against malaria. In 1955 the World Health Organization (WHO) adopted the controversial Global Eradication Campaign emphasising DDT (dichlorodiphenyltrichloroethane) spraying in homes. The incidence of malaria fell sharply where the programme was implemented, but the strategy was not applied in holoendemic Africa. This, along with the failure to achieve eradication in larger tropical regions, contributed to disillusionment with the policy. The World Health Assembly abandoned the eradication strategy in 1969. Aresurgence of malaria began at about that time and today reaches …

Taylor, Doan Hanh Nhan, Nguyen Dieu Thuong, Tran Thi Uyen, D. J. Fryauff, E. Gomez-Saladin, K. C. Kain, Le Dinh Cong, J. Kevin Baird

United States Public Health Resources

Chloroquine-resistant Plasmodium vivax malaria is emerging in Oceania, Asia, and Latin America. We assessed the drug sensitivity of P. vivax to chloroquine or halofantrine in two villages in southern, central Vietnam. This area has chloroquine-resistant Plasmodium falciparum but no documented chloroquine-resistant P. vivax. Standard dose chloroquine (25 mg/kg, over 48 hours) or halofantrine (8 mg/kg, 3 doses) was administered to 29 and 25 patients, respectively. End points were parasite sensitivity or resistance determined at 28 days. Of the evaluable patients, 23/23 100% (95% confidence interval [CI] 85.1–100) chloroquine and 21/24 (87.5%) (95% CI 67.6–97.3) halofantrine-treated patients were sensitive. Three halofantrine …

‘Gene Shaving’ As A Method For Identifying Distinct Sets Of Genes With Similar Expression Patterns, Trevor Hastie, Robert Tibshirani, Michael B. Eisen, Ash Alizadeh, Ronald Levy, Louis Staudt, Wing C. Chan, David Botstein, Patrick Brown

United States Public Health Resources

Background: Large gene expression studies, such as those conducted using DNA arrays, often provide millions of different pieces of data. To address the problem of analyzing such data, we describe a statistical method, which we have called ‘gene shaving’. The method identifies subsets of genes with coherent expression patterns and large variation across conditions. Gene shaving differs from hierarchical clustering and other widely used methods for analyzing gene expression studies in that genes may belong to more than one cluster, and the clustering may be supervised by an outcome measure. The technique can be ‘unsupervised’, that is, the genes and …

Helicobacter pylori infection is associated with a variety of clinical outcomes including gastric cancer and duodenal ulcer disease1. The reasons for this variation are not clear, but the gastric physiological response is influenced by the severity and anatomical distribution of gastritis induced by H. pylori. Thus, individuals with gastritis predominantly localized to the antrum retain normal (or even high) acid secretion2, whereas individuals with extensive corpus gastritis develop hypochlorhydria and gastric atrophy3, which are presumptive precursors of gastric cancer4. Here we report that interleukin-1 gene cluster polymorphisms suspected of enhancing production of interleukin-1-beta …

U.S. Obstetric Procedures, 1998, Sally C. Curtin, T. J. Mathews

United States Public Health Resources

According to the latest birth data from the National Center for Health Statistics, the use of electronic fetal monitoring, ultrasound, stimulation of labor, and induction of labor continued to rise in 1998 to unprecedented levels (1). At the same time, the total cesarean rate, which had fallen each year from 1989 to 1996, rose for the second consecutive year while the primary cesarean rate (rate per 100 births to women with no previous cesarean) rose for the first time during the 1989–1998 period. After increasing by 50 percent between 1989 and 1996, the rate of vaginal birth after previous cesarean …

An expert panel was convened to evaluate the U.S. Environmental Protection Agency’s “Proposed Guidelines for Carcinogen Risk Assessment” through their application to data sets for chloroform (CHCl3) and dichloroacetic acid (DCA). The panel also commented on perceived strengths and limitations encountered in applying the guidelines to these specific compounds. This latter aspect of the panel’s activities is the focus of this perspective. The panel was very enthusiastic about the evolution of these proposed guidelines, which represent a major step forward from earlier EPA guidance on cancer-risk assessment. These new guidelines provide the latitude to consider diverse scientific data …

Hyperphosphorylation Of The Asialoglycoprotein Receptor In Isolated Rat Hepatocytes Following Ethanol Administration, Benita L. Mcvicker, Dean J. Tuma, Carol A. Casey Jan 2000

Hyperphosphorylation Of The Asialoglycoprotein Receptor In Isolated Rat Hepatocytes Following Ethanol Administration, Benita L. Mcvicker, Dean J. Tuma, Carol A. Casey

United States Public Health Resources

Ethanol administration leads to altered function and impaired receptor-mediated endocytosis of the hepatocyte asialoglycoprotein receptor (ASGP-R). The purpose of the present study was to examine the effects of ethanol on the phosphorylation of the ASGP-R to determine whether this post-translational modification could contribute mechanistically to the observed ethanol-induced alterations. The methodological approach of this work involved the measurement of the phosphorylation state of the receptor obtained from isolated rat hepatocytes, using a combination of experimental designs from the biosynthetic incorporation of phosphate to the determination of steady-state phosphotyrosine levels. We report here that both short-term (1- to 2-week) and chronic …

Schutten, Lynn A. Cooper, Catherine B. Smith, Kanta Subbarao, Gary A. Anderson, Suzanne Carman, Christopher W. Olsen

United States Public Health Resources

Since 1998, H3N2 viruses have caused epizootics of respiratory disease in pigs throughout the major swine production regions of the U.S. These outbreaks are remarkable because swine influenza in North America had previously been caused almost exclusively by H1N1 viruses. We sequenced the full-length protein coding regions of all eight RNA segments from four H3N2 viruses that we isolated from pigs in the Midwestern U.S. between March 1998 and March 1999, as well as from H3N2 viruses recovered from a piglet in Canada in January 1997 and from a pig in Colorado in 1977. Phylogenetic analyses demonstrated that the 1977 …

Tivaevae: Local Aesthetics And Cook Islands Quilts, Suzanne P. Macaulay Jan 2000

Tivaevae: Local Aesthetics And Cook Islands Quilts, Suzanne P. Macaulay

Textile Society of America: Symposium Proceedings

What better image for a Cook Islands tivaevae than the vision of this colorful messenger embellishing the fabric's design in a magical moment of trust and oblivion? The sleeping bird brings tidings from Havaiki., the source of myth and memory, and is lured to rest in a field of imaginary flowers a long way from its poetic homeland.

The applique or patchwork tivaevae most representative of this genre appear to have been vigorously brushed by the mythical bird's feathers and left shimmering in an array of color. Cook Islands women who have created these patterns speak a language of …

Social Workers: Their Role And Relationship With The People They Serve, Sheila Moriarty Jan 2000

Social Workers: Their Role And Relationship With The People They Serve, Sheila Moriarty

Theses and Graduate Projects

The purpose of this study was to analyze the perceptions that social service workers have of their role and relationship with the people they serve. This study explored these questions of role and relationship through stories told by the social service workers about their work. The literature review reveals that there has been little research looking at the role and relationship with people using the stories of social service workers. Postmodernism and its challenge to the social service worker role as expert is the conceptual framework for this thesis. The research method is hermeneutics. It recognizes the legitimacy of lived …

Leadership In The Future: Preparation For Future Generations Of Leaders, Sherry A. Myers Jan 2000

Leadership In The Future: Preparation For Future Generations Of Leaders, Sherry A. Myers

Theses and Graduate Projects

What does the future of leadership hold? As books, lectures, seminars, and college courses are created, guiding the public through ideas on how to develop leadership skills, we remain concerned about what the leaders of tomorrow will need to know. Will the leadership skills of today apply to the workplace of tomorrow? Will our world politicians be those with the most education or the one with charisma? How do you lead numerous people spread throughout the world?

Beginning with a discussion of two leadership theories existing today that will be viable in the future, we will look at how this …
