A Mycobacterium Ulcerans Toxin, Mycolactone, Causes Apoptosis In Guinea Pig Ulcers And Tissue Culture Cells, Kathleen M. George, Lisa Pascopella, Diane M. Welty, P. L. C. Small
Public and Community Health Sciences Faculty Publications
Mycobacterium ulcerans is the causative agent of Buruli ulcer, a tropical ulcerative skin disease. One of the most intriguing aspects of this disease is the presence of extensive tissue damage in the absence of an acute inflammatory response. We recently purified and characterized a macrolide toxin, mycolactone, from M. ulcerans. Injection of this molecule into guinea pig skin reproduced cell death and lack of acute inflammatory response similar to that seen following the injection of viable bacteria. We also showed that mycolactone causes a cytopathic effect on mouse fibroblast L929 cells that is characterized by cytoskeletal rearrangements and growth arrest …

To investigate the role of certain neurohormones in agonistic behaviour, fights were staged between pairs of size-matched male shore crabs Carcinus maenas, and blood samples were taken immediately after the contests had been resolved. Samples were also taken from these crabs at rest (before and after fighting) and after walking on a treadmill. A control group of crabs also had samples taken on each experimental day. Concentrations of tyramine, dopamine, octopamine, serotonin (5-HT) and norepinephrine were determined in each blood sample using a gas chromatography/mass spectrometry (GC-MS) system. Norepinephrine was not detectable in any of the samples, but the standards …

Does Market Timing Contribute To The Cattle Cycle?, Stephen F. Hamilton, Terry L. Kastens

Recent evidence suggests that cyclical cattle inventories are driven by exogenous shocks. This article examines a second possible contributing factor to the cattle cycle: a market timing effect that arises from individual attempts to maintain countercyclical inventories. The model uncovers an important conceptual point: to the extent that cycles are driven by exogenous shocks, a representative producer should outperform one who maintains a constant inventory; whereas, for cycles induced by market timing, a representative producer should underperform one with a constant inventory. Simulated net returns over 1974–98 reveal that a constant-inventory manager significantly outperformed the representative U.S. producer, which indicates …

Selenium Poisoning Of Wildlife And Western Agriculture: Cause And Effect, N. E. Korte

This project examined the hypothesis that selenium contamination is not the principal cause of the decline of endemic fish species in the Upper Colorado Basin. Activities employed to test this hypothesis included a reconnaissance of locations altered by recent road construction, a reinterpretation of available literature regarding selenium toxicity, and the interpretation of unpublished data obtained from the Upper Colorado Basin Fish Recovery Program. The project demonstrates that most of the evidence implicating selenium is circumstantial. Specifically, this research demonstrates that neither the historical record nor the technical literature consistently supports the emphasis given selenium toxicity. For example, many locations …

A major challenge for the Western Australian community is to ensure the sustainability of these resources in the face of growing pressure, and to ensure that the industries based on these resources are able to develop in a controlled and sustainable fashion which will optimise long-term benefits to the community. Vestimentiferan tubeworms thriving in sulfidic deep-sea hydrothermal vents and cold seeps are constrained by their nutritional reliance on chemoautotrophic endosymbionts. In a recent phylogenetic study using 16S ribosomal DNA, we found that endosymbionts from vent and seep habitats form two distinct clades,vith little variation within each clade. In the present study, we used two different approaches to assess the genetic variation among biogeographically distinct vestimentiferan symbionts, DNA sequences were obtained for the noncoding, internal transcribed spacer (ITS) regions of the rRNA operons of symbionts associated with six different genera of vestimentiferan tubeworms. The objectives were 1) to investigate the effects of oocyte maturation in serum-free and amino acid-supplemented defined media on oocyte transcript levels, blastocyst cell number, and apoptosis; 2) to investigate the influence of oocyte maturation culture atmosphere on blastocyst development, total cell number, and apoptosis; and 3) to examine the influence of epidermal growth factor (EGF) during oocyte maturation on blastocyst cell number and apoptosis. The results demonstrate that blastocysts derived from in vitro maturation, fertilization, and embryo culture protocols undergo apoptosis but that apoptotic levels are not greatly influenced by the oocyte maturation environment.
