Phase structures and transformation mechanisms of nonracemic chiral biological and synthetic polymers are fundamentally important topics in understanding their macroscopic responses in different environments. It has been known for many years that helical structures and morphologies can exist in low-ordered chiral liquid crystalline (LC) phases. However, when the chiral liquid crystals form highly ordered smectic liquid crystal phases, the helical morphology is suppressed due to the crystallization process. A double-twisted morphology has been observed in many liquid crystalline biopolymers such as dinoflaggellate chromosomes (in Prorocentrum micans) in an in vivo arrangement. Jim Laue was a major figure in conflict resolution and he is sorely missed. His absence is particularly regretted because his was a strong voice in discussions over the ethical dimension of conflict resolution, arguing eloquently and passionately in favor of a self-consciously ethical theory and practice of conflict resolution. Sometime colleagues of Laue, our work over the last decade has argued equally passionately (if not as eloquently) in behalf of a self-consciously cultural theory and practice of conflict resolution, (see Avruch and Black 1987, 1991, 1993; Black and Avruch 1989, 1993).

Preparation And Characterization Of A Site-Directed Mutant Of Escherichia Coli L-Threonine Dehydrogenase, Qian Zhou

Threonine dehydrogenase from E.coli is a member of the medium-chain, Zn2+-containing alcohol dehydrogenase family. A site-directed mutant of theronine dehydrogenase was prepared at position 88 by replacing the native glutamate with cysteine using polymerase chain reaction. The mutated enzyme, designated TDH E88C, was expressed and purified to homogeneity in a three-step chromatographic procedure. The activity of the mutant after each step of purification was determined by a specific assay. Both the total activity and the specific activity of TDH E99C were significantly decreased compared to the wild-type TDH. The mutant enzyme was not sufficiently stable to obtain reliable Vmax or …

The rapidity of change has left scant opportunity for investigation of the consequences of biotechnology adoption on long-term ecosystem or economic system functioning. Economic theory suggests that, if the "Biotechnology Revolution" is left to market forces alone, there will be neglected public goods. Theory and limited empirical evidence suggests that there are significant incentives for private firms to discount and neglect negative environmental impacts and to develop products that meet only the needs of those able and willing to pay. Negative distributional impacts on rural societies and economies will not normally enter the private calculus nor will the long-term problems …

Association of a targeting ligand such as transferrin, or an endosome disrupting peptide such as GALA, with cationic liposome-DNA complexes ('lipoplexes') results in a significant enhancement of transfection of several cell types. Although these strategies can overcome some of the barriers to gene delivery by lipoplexes, the mechanisms by which they actually enhance transfection is not known. In studies designed to establish the targeting specificity of transferrin, we found that apo-transferrin enhances transfection to the same extent as transferrin, indicating that internalization of the lipoplexes is mostly independent of transferrin receptors. Lymphoproliferative diseases that occur in immunocompromised patients are frequently associated with herpesviruses. These patients often fare poorly after treatment with conventional chemotherapy. We reported previously that patients with AIDS-related Burkitt’s lymphoma (BL) responded to parenteral azidothymidine (AZT) and IFN-α. We found that EBV-positive lymphoma cells derived from these patients cultured with AZT express CD95 and undergo apoptosis. AZT-mediated apoptosis was caspase dependent and occurred despite Fas receptor blockade. In contrast, EBV-negative lymphomas were resistant to AZT-induced apoptosis, as were EBV-positive lymphomas that expressed high levels of bcl-2. The dependence on laser intensity of recently discovered plateau structures in laser-assisted x-ray–atom scattering, both with and without a static electric field present, is analyzed. Using the “three-step” model and the strong-field approximation we demonstrate a connection between laser-assisted, x-ray–atom scattering and high-order harmonic generation: For high laser-field intensities without a static field present, both processes have plateaus whose energies extend to the cutoff value 3.17U_p, where U_p is the ponderomotive potential energy. For x-ray–atom scattering in the presence of a static electric field we show that at high laser-field intensities two plateaus appear: One is …
