Bacterial Growth State Distinguished By Single-Cell Protein Profiling: Does Chlorination Kill Coliforms In Municipal Effluent?, David Rockabrand, Teresa Austin, Robyn Kaiser, Paul H. Blum
School of Biological Sciences: Faculty Publications
Municipal effluent is the largest reservoir of human enteric bacteria. Its public health significance, however,
depends upon the physiological status of the wastewater bacterial community. A novel immunofluorescence
assay was developed and used to examine the bacterial growth state during wastewater disinfection. Quantitative
levels of three highly conserved cytosolic proteins (DnaK, Dps, and Fis) were determined by using
enterobacterium-specific antibody fluorochrome-coupled probes. Enterobacterial Fis homologs were abundant
in growing cells and nearly undetectable in stationary-phase cells. In contrast, enterobacterial Dps homologs
were abundant in stationary-phase cells but virtually undetectable in growing cells. The range of variation in
the abundance of …

Chlorella Virus Pbcv-1 Encodes A Functional Homospermidine Synthase, Annette Kaiser, Michael Vollmert, Dorothea Tholl, Michael V. Graves, James Gurnon, Weimei Xing, Amber D. Lisec, Kenneth Nickerson, James L. Van Etten
Kenneth Nickerson Papers
Sequence analysis of the 330-kb genome of chlorella virus Paramecium bursaria chlorella virus 1 (PBCV-1) revealed an open reading frame, A237R, that encodes a protein with 34% amino acid identity to homospermidine synthase from Rhodopseudomonas viridis. Expression of the a237r gene product in Escherichia coli established that the recombinant enzyme catalyzes the NAD+-dependent formation of homospermidine from two molecules of putrescine. The a237r gene is expressed late in PBCV-1 infection. Both uninfected and PBCV-1-infected chlorella, as well as PBCV-1 virions, contain homospermidine, along with the more common polyamines putrescine, spermidine, and cadaverine. The total number of polyamine …

Absence Of Tuberculosis In Free-Ranging Deer In Nebraska, David J. Steffen, David W. Oates, Mauritz C. Sterner, Vickie L. Cooper
Harold W. Manter Laboratory of Parasitology: Faculty Publications
Lymph nodes from 271 white-tailed deer (Odocoileus virginianus) and mule deer (Odocoileus hemionus) in Nebraska (USA) were examined microscopically for tuberculoid lesions. Lymph nodes lesions in at least one node were found in 12 deer. Lesions were examined with Zeihl-Neelson acid fast stains and by polymerase chain reactions using M. bovis specific probes. No evidence of tuberculosis was found. The small granulomatous lesions were likely caused by other bacteria.

Experimental Infections Of Muskoxen (Ovibos Moschatus) And Domestic Sheep With Umingmakstrongylus Pallikuukensis (Nematoda: Protostrongylidae): Parasite Development, Population Structure, And Pathology, Susan J. Kutz, Eric P. Hoberg, Lydden Polley
Harold W. Manter Laboratory of Parasitology: Faculty Publications
Three captive muskoxen (Ovibos moschatus) were successfully infected with third-stage larvae of Umingmakstrongylus pallikuukensis digested or emerged from the slugs Deroceras reticulatum and D. laeve, for the first time completing the life cycle of this parasite under experimental conditions. The course of parasite development and patency was followed for 26 months post infection (p.i.) using fecal examinations and radiography. The prepatent periods in two of the muskoxen were 91 and 95 days and the patent period in one extended for 23 months. Larval production peaked 13–14 months p.i. On postmortem of two of the muskoxen at months …
Of Hungry Wolves And Horizontal Conflicts: Rethinking The Justifications For Bank Holding Company Liability, Eric J. Gouvin
Of Hungry Wolves And Horizontal Conflicts: Rethinking The Justifications For Bank Holding Company Liability, Eric J. Gouvin
Faculty Scholarship
To what extent should bank holding companies bear the costs of bank failure? Current banking law provides a number of ways to impose liability on bank holding companies for bank failure. Those devices, however, have developed haphazardly and sometimes rest on inconsistent theoretical foundations. This Article critiques the regulatory justifications that have been offered for holding company liability and offers an alternative justification for imposing liability on holding companies based on the idea that directors of bank subsidiaries suffer from an especially difficult form of horizontal conflict--the situation where the board of directors owes several different duties and chooses to …

Capillary Electrophoresis/Tandem Mass Spectrometry With Array Detection, L.J. Deterding, K.B. Tomer, J.M.Y. Wellemans, Ronald Cerny, M.L. Gross
Ronald Cerny Publications
Mixtures of standard compounds of biological interest were analyzed by capillary electrophoresis in conjunction with tandem four sector mass spectrometry (MS/MS) using an array detector. Capillary electrophoresis offers extremely high separation efficiencies while array detection allows for the simultaneous acquisition of a large fraction of a tandem mass spectrum and high utilization of the sample ions produced. Consequently, improvements in sensitivity and structural information were observed for the combined techniques in comparison to similar measurements with a four-sector and a single-point detector. Coaxial continuous-flow fast atom bombardment was used to create (M + H) precursor ions of the separated analyses …
Affinity Chromatography: A Review Of Clinical Applications, David S. Hage
Affinity Chromatography: A Review Of Clinical Applications, David S. Hage
David Hage Publications
Affinity chromatography is a type of liquid chromatography that makes use of biological-like interactions for the separation and specific analysis of sample components. This review describes the basic principles of affinity chromatography and examines its use in the testing of clinical samples, with an emphasis on HPLCbased methods. Some traditional applications of this approach include the use of boronate, lectin, protein A or protein G, and immunoaffinity supports for the direct quantification of solutes. Newer techniques that use antibody-based columns for on- or off-line sample extraction are examined in detail, as are methods that use affinity chromatography in combination with …
Octamer-Based Genome Scanning Distinguishes A Unique Subpopulation Of Escherichia Coli O157:H7 Strains In Cattle, Jaehyoung Kim, Andrew K. Benson
Octamer-Based Genome Scanning Distinguishes A Unique Subpopulation Of Escherichia Coli O157:H7 Strains In Cattle, Jaehyoung Kim, Andrew K. Benson
Department of Food Science and Technology: Faculty Publications
Multilocus-genotyping methods have shown that Escherichia coli O157:H7 is a geographically disseminated clone. However, highresolution methods such as pulse-field gel electrophoresis demonstrate significant genomic diversity among different isolates. To assess the genetic relationship of human and bovine isolates of E. coli O157:H7 in detail, we have developed an octamer-based genome-scanning methodology, which compares the distance between over-represented, strand-biased octamers that occur in the genome. Comparison of octamer-based genome-scanning products derived from >1 megabase of the genome demonstrated the existence of two distinct lineages of E. coli O157:H7 that are disseminated within the United States. Human and bovine isolates are nonrandomly …

Capsicum-Laden Soils Decrease Contact Time By Northern Pocket Gophers, Ray T. Sterner, Kelly Hollenbeck, Stephen Shumake
United States Department of Agriculture Wildlife Services: Staff Publications
Fossorial rodents damage lawns/water impoundments/ crops. We conducted a two-choice, parametric-type study to determine the effects of capsicum-oleoresin/soil mixtures (0.00, 0.75, 1.50, and 2.25%) upon soil-contact, soil-digging, and pelage-grooming behaviors in northern pocket gophers (Thomomys talpoides). In 3 alternate-day (1-h/day) exposures to ≥1.50% capsicum-oleoresin soil mixtures, gophers decreased mean soil contact time by 46% relative to placebo-exposed animals. Grooming time yielded a concentration x trial interaction that showed intense grooming by capsicum-exposed animals during trial 1, with "convergence" of times to near those of the "placebos" (0.00% capsicum oleoresin) by trial 3. The significant decrease in grooming activity …
Effective Of Preventive Coyote Hunting On Sheep Losses To Coyote Predation, Kimberly Wagner, Michael Conover
Effective Of Preventive Coyote Hunting On Sheep Losses To Coyote Predation, Kimberly Wagner, Michael Conover
United States Department of Agriculture Wildlife Services: Staff Publications
Aerial hunting is commonly used by agriculture agencies in the Intermountain West to reduce coyote (Canis latrans) predation on domestic sheep. We assessed the effect of aerial hunting of coyotes on sheep losses to coyotes, and the need for corrective predation management (hours of work, device nights) on the same pastures when sheep arrived for the subsequent summer grazing season (3-6 months after aerial hunting). Comparisons were made between paired pastures with (treated) and without (untreated) winter aerial hunting from helicopters. Average (x ± SE) pasture size was 45.2 ± 14.1 km2 (n = 21) for treated …

Repellents To Reduce Cable Gnawing By Northern Pocket Gophers, Stephen A. Shumake, Ray T. Sterner, Stanley E. Gaddis
United States Department of Agriculture Wildlife Services: Staff Publications
There is a continuing need to develop improved repellent formulations to protect buried cable installations from damage by problem wildlife. We evaluated 2.0% mass/mass levels of capsaicin and denatonium benzoate in a polybutene carrier material (Indopol®) and an aboveground, rodent-deer plastic mesh barrier (Vexar®) for reducing gnawing by northern pocket gophers (Thomomys talpoides) on communications cable (RG-8U). When treatments were applied as surface coatings, neither capsaicin nor denatonium samples were lower (P > 0.05) in measnres of cable damage compared to control (Indopol® alone) or samples treated with Vexar® plastic mesh. When the test was repeated with a new …

Potential Associations Among Genetic Markers In The Serotonergic System And The Antisocial Alcoholism Subtype, E. M. Hill, S. F. Stoltenberg, M. Burmeister, M. Closser, R. A. Zucker
Department of Psychology: Faculty Publications
Alcoholism is transmitted in families. The complexity and heterogeneity of this disorder has made it difficult to identify specific genetic correlates. One design with the potential to do so is the family-based association study, in which the frequencies of genetic polymorphisms are compared between affected and nonaffected members. Reduced central serotonin neurotransmission is associated with features of an antisocial subtype of alcoholism, although a primary deficit has not been traced to a particular component. Genetic markers related to the serotonergic system have been identified, located, and cloned. If associations can be discovered, the development process for pharmacotherapy could be facilitated. …

Objective: To develop and test a quantitative, interviewer-administered food frequency questionnaire (FFQ) to ascertain nutrient intakes of individuals in northern India. Design: A 92-item FFQ was developed based on food use and market surveys of the study area. A validation study was conducted consisting of 24-h diet recalls (24HR) administered on 6 randomly selected days over 1 year. Two FFQs were administered, one each at the beginning and end of the 1-year period. FFQ and 24HR-derived nutrient scores were compared using correlation and regression analyses and by computing differences between nutrient intakes estimated by the two methods. Setting: Rural villages …
Brownfields Policies For Sustainable Cities, Joel B. Eisen
Brownfields Policies For Sustainable Cities, Joel B. Eisen
Law Faculty Publications
My analysis begins in Part I with a short summary of brownfields law and policy, and a description of our current understanding of "sustainable development." There is no hard and fast definition of the term, but it is increasingly more clear that a body of sustainable development "law" will take shape as the product of a host of decisions made now and in the near future. The optimal way to ensure that brownfields programs mesh with this body of law-whatever it turns out to be-is to incorporate basic norms of sustainable development about which there is widespread agreement. Those agreed-upon …

Blocked Pathways: Potential Legal Responses To Endocrine Disrupting Chemicals, Noah M. Sachs
Law Faculty Publications
Over fifty different chemicals may have endocrine disrupting effects, and these chemicals permeate our environment. We may be exposed to them through the products we use, the food we eat, the water we drink, and the air we breath. Although there is a clear need for further research into EDCs, the legal and regulatory communities should begin to consider the types of responses that may be appropriate if the scientific evidence grows stronger.
This article describes and assesses potential legal responses to EDCs, focusing on regulation and litigation, and concludes that existing legal tools will probably be inadequate to respond …

Managed Care And Mental Health: Clinical Perspectives And Legal Realities, Jesse Goldner
All Faculty Scholarship
Managed care is beginning to dominate the delivery of mental health services. The Article reviews limitations on managed care's ability to deal adequately with mental illness. It discusses empirical and other research examining the use of primary care providers as gatekeepers and it explores utilization review mechanisms, focusing particularly on providers' responses to UR. The impact on quality, access and continuity of care on discrete populations is analyzed. The article then surveys a variety of legal issues in the regulation of managed care, particularly as they apply to the provision of mental health services. These include ERISA, parity and liability …
The Effects Of Moulting On Muscle Fibre Characteristics Of The Yabby, Cherax Albidus, Tina Marie Lamey
The Effects Of Moulting On Muscle Fibre Characteristics Of The Yabby, Cherax Albidus, Tina Marie Lamey
Theses : Honours
In order to grow, crustaceans must shed their exoskeleton in a process known as "moulting'. Although this process is short and intermittent, it remains dominant over the life of a crustacean. Physiological changes in the period between moults (intermoult) are comparatively quiescent as opposed to the periods directly before and after the actual moult, known as premoult, and postmoult respectively (West, 1997). Moulting is associated with distinct physiological changes including the breakdown of muscle (Musgrove and Geddes, 1985). This muscle breakdown, known as atrophy is the diminution in size of the actual muscle mass and is very specific, occurring to …
Investigation Of The Contractile And Regulatory Proteins In Malignant Hyperthermia And Normal Skeletal Muscle, Louise C. Norbury
Investigation Of The Contractile And Regulatory Proteins In Malignant Hyperthermia And Normal Skeletal Muscle, Louise C. Norbury
Theses : Honours
Malignant hyperthermia/hyperpyrexia (MH) is a serious human skeletal muscle disorder that manifests itself during general anaesthesia. Currently, the most reliable method of diagnosing this disease is the in-vitro contracture test (IVCT). This procedure is both highly invasive for the patient and expensive to perform. The skinned muscle fibre technique is less invasive, and could possibly be used as an alternative diagnostic test for MH. In this study, skeletal muscle samples were obtained from B adults (age 23-54 years) undergoing elective surgery at Royal Perth Hospital. Six patients were diagnosed as normal (controls) and two equivocal for MH (MHE). Using the …
Evaluating Oral Histories For Restoration Ecology, Margaret Robertson
Evaluating Oral Histories For Restoration Ecology, Margaret Robertson
Theses : Honours
Significant ecological restoration is required in the agricultural regions of south-western Australia. Environmental history, including local knowledge based on long-term observation of the biophysical environment, can help guide this process. Scientists already use local anecdotal information because other information sources are absent or too recent, yet they are often skeptical of its veracity. This study focused on whether environmental oral histories can be evaluated for factual accuracy and their capacity to be useful in the restoration process. Some of the complexities associated with linking environmental histories with restoration ecology, and the role played by oral histories in establishing the link …

Digital Imaging has had an enormous impact on industrial applications such as the Internet and video-phone systems. However, demand for industrial applications is growing enormously. In particular, internet application users are, growing at a near exponential rate. The sharp increase in applications using digital images has caused much emphasis on the fields of image coding, storage, processing and communications. New techniques are continuously developed with the main aim of increasing efficiency. Image coding is in particular a field of great commercial interest. A digital image requires a large amount of data to be created. This large amount of data causes …

Short-Term Memory For Nonsense Strings In Children With Reading Disabilities, Linda K. Lane
Theses : Honours
A large body of evidence exists that demonstrates strong correlations between reading ability, phonological awareness and memory. The current study was designed to compare the performance of 24 10-year-old students with reading disabilities and a group of 24 8-year-old average readers, who were matched according to reading age. These students were given a decoding task requiring the recall of nonsense strings. This task was designed to measure working memory for phonological elements. An ANOVA yielded a significant main effect for group in favour of the older students, and a main effect for total errors and vowel and space location. Participants …
Is There A Need For Speed? : Risky Driving Behaviour And Young Male Drivers : A Review Of The Literature, Catherine A. Ferguson
Is There A Need For Speed? : Risky Driving Behaviour And Young Male Drivers : A Review Of The Literature, Catherine A. Ferguson
Theses : Honours
Young male drivers have been identified as a high risk for Motor Vehicle Accident. This paper reviews three components for accident risk: the driving environment, society and individual factors associated with accident histories. Possible predictors for identifying young men at risk for accident involvement are driving behaviour, driving attitude and sensation seeking. Protective factors to reduce the incidence of risky driving in youth might include legal restrictions, increased education and training. The paper recommends that future research tests the above predictors for a relationship with accident involvement in young men.
