Isolation And Characterization Of A Caenorhabditis Elegans Src Loss-Of-Function Allele Using Reverse Genetics, Jennifer Dignan Hogan Jan 1999

Isolation And Characterization Of A Caenorhabditis Elegans Src Loss-Of-Function Allele Using Reverse Genetics, Jennifer Dignan Hogan

Doctoral Dissertations

The vertebrate proto-oncogene Src is a protein-tyrosine kinase that has been implicated as a component of receptor-mediated signal transduction pathways important for cell growth and differentiation. Consistent with this notion, overexpression or activation of Src by mutation induces neoplastic transformation in cell culture, leads to tumorigenesis in laboratory animals, and has been observed in a number of human tumors. Despite years of intensive investigation, neither its role in oncogenesis nor its normal, biological role is understood.

To diminish the issue of redundancy that has complicated analysis of Src function in vertebrates and Drosophila, I have chosen to study Src function …

Finding A Voice: Poetry And Performance With First Graders, Lisa Lenz Bianchi Jan 1999

Finding A Voice: Poetry And Performance With First Graders, Lisa Lenz Bianchi

Doctoral Dissertations

This is a qualitative research study of a ten week immersion unit in the reading, writing, and performance of poetry conducted in a first grade classroom in Closter, New Jersey during the winter of 1995. The three girls selected as case studies show the ways in which remarkably different children expand their repertoires of ways with words as speakers, readers, and writers. Danielle, a performative speaker, learns to make her tacit knowledge about performance part of her explicit frames of reference. This shift enables her to serve as a coach for peers who are less adept at crafting performative texts. …

Behavioral And Physiological Responses Of The Lobster, Homarus Americanus, To Temperature: A New Synthesis, Steven Harold Jury Jan 1999

Behavioral And Physiological Responses Of The Lobster, Homarus Americanus, To Temperature: A New Synthesis, Steven Harold Jury

Doctoral Dissertations

Temperature has a pervasive influence on lobster behavior, physiology and ecology and affects their subsequent distribution in thermally variable habitats such as estuaries and coastal areas. A multidisciplinary approach, including field and laboratory studies, was used to show: (1) that lobsters sense temperature with warm and cool thresholds as small as 0.1--0.2°C; (2) the relationship between temperature and activity is not linear, but instead switches between a high activity level in warmer months (10--20°C) and a lower level in colder months (<10°C) with transition periods in the spring and fall; (3) Parallel studies in the lab and field show that daily levels of activity are not greatly influenced by small temperature variations (i.e. tidally induced changes of 14°C), but activity levels are significantly higher in the field (249 +/- 55.1 m/d) than in the laboratory (88 +/- 12.0 m/d); (4) lobsters prefer a narrow range of temperatures over others available in a thermal gradient and avoid temperatures >23.5 +/- 0.4°C, suggesting that they behaviorally thermoregulate. While this preferred temperature shifts seasonally, the final preferred temperature …

The Role Of Cytokines In The Pathogenesis Of Experimental Legionella Pneumophila Infections, Corinna Mary Krinos Jan 1999

The Role Of Cytokines In The Pathogenesis Of Experimental Legionella Pneumophila Infections, Corinna Mary Krinos

Doctoral Dissertations

Legionnaires' disease is an acute lobar pneumonia caused, primarily by the facultative intracellular pathogen Legionella pneumophila. This organism when inhaled by humans descends into the lower respiratory tract and parasitizes alveolar macrophages. L. pneumophila adhered to U-937 cells, A549 cells and peritoneal macrophages from A/J mice in an opsonin-independent fashion. Following attachment, the organism penetrated the cell membrane, replicated within these cells eventually inducing lysis. To better define the adhesion of L. pneumophila to host cells, an E. coli clone (LP 116), expressing the 25 kDa major outer membrane protein (MOMP) of L. pneumophila was used in binding studies. This …

The Effects Of Reversible Inactivation By Lidocaine Of Ventral Striatum And Intralaminar Nucleus Of The Thalamus In Match To Sample Tasks With And Without Delays Trained In The Lever Box, Mary Christine Porter Jan 1999

The Effects Of Reversible Inactivation By Lidocaine Of Ventral Striatum And Intralaminar Nucleus Of The Thalamus In Match To Sample Tasks With And Without Delays Trained In The Lever Box, Mary Christine Porter

Doctoral Dissertations

Three experiments systematically examined the effects of reversibly inactivating intralaminar nuclei and ventral striatum with infusions of lidocaine. The ventral striatum and the intralaminar nucleus were targeted based on deficits in accuracy and speed of responding in delayed conditional discrimination tasks associated with permanent lesions of these structures (Mair, Burk and Porter, 1998).

The present research showed that a 4.0 mul of 2% lidocaine caused impairments accuracy, but not in response speed, in a match to sample task trained in the lever box without delays when the internal cannula projects 0.5 mm from the guide cannula. One microliter of 2%, …

Motherwork, Artwork: The Mother/Artist In Fiction By Parton, Phelps, Chopin, Woolf, Drabble, And Walker, Nancy Hoyt Lecourt Jan 1999

Motherwork, Artwork: The Mother/Artist In Fiction By Parton, Phelps, Chopin, Woolf, Drabble, And Walker, Nancy Hoyt Lecourt

Doctoral Dissertations

This study asks the question, What happens to a practicing (fictional) mother who also tries to be a practicing artist? How do literary texts represent such people? How do they represent the relationship between material and artistic work? The primary works studied are Sarah Parton's Ruth Hall, (1855), Elizabeth Stuart Phelps' The Story of Avis (1877), Kate Chopin's The Awakening (1899), Virginia Woolf's To the Lighthouse (1927), and Margaret Drabble's The Millstone (1965). The conclusion focuses on Alice Walker's short story, "Everyday Use."

Mother-artists finds themselves on the "wrong" side of the nature/culture binary, where ideologies about "true womanhood" and …

The Mut-2 Mutator Of Caenorhabditis Elegans, Kathrine Queta Flint Boese Jan 1999

The Mut-2 Mutator Of Caenorhabditis Elegans, Kathrine Queta Flint Boese

Doctoral Dissertations

The mut-2 mutator plays multiple regulatory roles in the germ line of C. elegans. In addition to regulating germ line transposition of at least four distinct transposon families (Tc1, Tc3, Tc4 and Tc5) mut-2 is implicated in chromosome segregation. Animals that harbor the mut-2 mutator produce broods with a higher incidence of males phenotype (Him) as a result of an increase in X chromosome non-disjunction during meiosis.

Using the Him phenotype conferred by mut-2(r459), I mapped the gene to the dpy-14 sem-4 interval on LGI. However, efforts to identify a molecular clone of the gene were hampered because the available …

The Development Of Rna Probe And Rt-Pcr Assays For The Detection Of Enteroviruses In Sludge, Amy Elisabeth Moore Jan 1999

The Development Of Rna Probe And Rt-Pcr Assays For The Detection Of Enteroviruses In Sludge, Amy Elisabeth Moore

Doctoral Dissertations

Many wastewater treatment plants generate more sludge than can be disposed of by conventional means. The United States Environmental Protection Agency (USEPA) has encouraged communities to dispose of sludge by land application. Sludge may contain enteric viruses that are known to survive for long periods of time in sludge-amended soil and can travel great distances, potentially contaminating surface and ground water.

Standard cell culture methods for the detection of enteric viruses are costly and results are not obtained for 30 or more days. The development of methods that provide results more quickly and with lower cost are needed.

How To Use Marketing Strategy And Communication For Rekindling A New Life Of The Old Street, Chao-Wang Wang Jan 1999

How To Use Marketing Strategy And Communication For Rekindling A New Life Of The Old Street, Chao-Wang Wang

Theses

History is the record of a community or culture in its past life. It symbolizes the continuation of our lives. Different areas nurtured different cultures and these cultures represent different peoples' ways of life and particular values. Not only does history provide evidence for us to trace and explore our ancestors' footprints, it also gives a blueprint for us to learn lessons and experiences so that we may be able to avoid quarrels and wars, and to create a peaceful world. Therefore history and culture has the functionality of carrying on the heritage to provide a way for the future …

The Relationship Between Working Status Of Mothers And Parental-Child Attachment, Sherri Denise Brown Jan 1999

The Relationship Between Working Status Of Mothers And Parental-Child Attachment, Sherri Denise Brown

Theses

The factors which influence the development of attachment between mothers and children were examined in this study. The researcher specifically looked at the differences in attachment with regard to the mother's work status during infancy. Thirty-five adolescent subjects completed the Inventory of Parent and Peer Attachment test (lPPA). The subjects were categorized into four groups based on what their mothers' work status was during their infancy. The information regarding work status was obtained through a demographic sheet completed by the parents of the subjects.

The test completed by the subjects consisted of twenty-five statements regarding a relationship with their mother. …

A Factual Review Of How Nursing Homes Are Accommodating The Elderly With Mental Illness, Martha L. Straughter Jan 1999

A Factual Review Of How Nursing Homes Are Accommodating The Elderly With Mental Illness, Martha L. Straughter

Theses

Thanks to improved drugs, diet and medical treatment, America's population is living longer than ever. While this is certainly a good occurrence, there are problems arising from it. Experts are seeing that many of these elderly people have a variety of mental illnesses. These illnesses have either been with the individual since youth, are the result of a deteriorating mind. State hospitals and other mental health facilities are unable to provide continuous, long-term care for the large number of elderly affected with these unfortunate illnesses.

This thesis will address this complex, ever-growing, issue in three parts by identifying and defining …

Ultrastructural And Morphometric Analysis Ofperipheral Nerve Regenerationwithin Silicone Tubes, Leyla Canpolat, Aysel Kükner, İbrahi̇m Canpolat, Enver Ozan

Turkish Journal of Medical Sciences

Semi-thin and thin sections were used to observe the morphogenesis of peripheral nerves regenerating in silicone tubes. The progress of peripheral nerve regeneration across a 10 mm gap within the silicone tubes was examined after 3, 6 and 9 months. The left sciatic nerves of rats were transected. The proximal and distal nerve stumps were inserted into a silicone tube, in which they were separeted by a 10 mm gap. A tissue cable formed inside the silicone tube with regenerated axons. In the 3rd month, the 10 mm gap was bridged by a structure composed of regenerating axons, blood vessels …

Turkish Journal of Medical Sciences

The effects of Rumex patientia and Urtica dioica on levels of blood glucose, plasma amino acids and other parameters, urine excreta, and liver and kidney histology were examined in diabetic rats induced by streptozotocin. Streptozotocin increased blood glucose and changed the levels of amino acids and other parameters, and caused degenerative changes in the liver and kidney. Rumex patientia had some protective effect on these parameters changed by streptozotocin, while Urtica diocia had no protective effects.

The Therapeutic Effect Of Nifedipine As A Ca+2entry Blocker On Experimental Transientischemic Retina, Murat Kaya, Cemal Gündoğdu

Turkish Journal of Medical Sciences

We evaluated the effect of nifedip-ine, an L-type voltage sensitive Ca+2 channel (VSCC) blocker, on the onset of neuronal damage induced by transient ischemia of guinea pig retinas in vivo. The animals were divided into two groups. In the first group 2 mg/kg Nifedipine was administered intraperi-toneally, while only 2 cc/kg NaCl 0.9% solution was given to the second group thirty minutes before the ischemic insult. Seven days after the temporary ligation of bilateral common carotid arteries for 45 minutes, the animals were dispatched and the eyes were enucleated for histopathologic analysis. It was observed that in the Nifedipine treated …
