Development Of A Blocking Enzyme-Linked Immunosorbent Assay For Detection Of Serum Antibodies To O157 Antigen Of Escherichia Coli, William Laegreid, Mark Hoffman, James Keen, Robert Elder, Jimmy Kwang

The O157 antigen of Escherichia coli shares structural elements with lipopolysaccharide (LPS) antigens of other bacterial species, notably Brucella abortus and Yersinia enterocolitica O9, a fact that confounds the interpretation of assays for anti-O157 antibodies. To address this problem, a blocking enzyme-linked immunosorbent assay (bELISA) was designed with E. coli O157:H7 LPS as the antigen and a monoclonal antibody specific for E. coli O157, designated 13B3, as the competing antibody. The bELISA had equivalent sensitivity to, and significantly higher specificity than, the indirect ELISA (iELISA), detecting anti-O157 antibodies in sera from cattle experimentally inoculated with O157:H7. Differential Expression Of The Arabidopsis Nia1 And Nia2 Genes: Cytokinin-Induced Nitrate Reductase Activity Is Correlated With Increased Nia1 Transcription And Mrna Levels, Xiaodan Yu, Sujatha Sukumaran, László Márton

Nitrate reductase (NR) activity increased up to 14-fold in response to treatment of Arabidopsis thaliana seedlings with the cytokinin benzyladenine. NR induction was observed in seedlings germinated directly on cytokinin-containing medium, seedlings transferred to cytokinin medium, and seedlings grown in soil in which cytokinin was applied directly to the leaves. About the same level of induction was seen in both wild-type and Nia2-deletion mutants, indicating that increased NR activity is related to the expression of the minor NR gene, Nia1. From Star Charts To Stoneflies: Detecting Relationships In Continuous Bivariate Data, James E. Garvey, Elizabeth A. Marschall, Russell A. Wright

Within many ecological systems, relationships between controlling factors and associated response variables are complex. In many cases, the response should vary little when the controlling factor exerts strong effects. Conversely, when the effect of the controlling factor is weak or absent, the response may vary greatly with effects of other factors. Correlation or regression analyses often may not be appropriate for testing these relationships, because variance of the response changes with values of the controlling factor. We suggest using a technique from the astronomy literature, a two-dimensional Kolmogorov- Smirnov (2DKS) test, to detect relationships in bivariate data with these patterns …

A Cladistic Analysis Of NdhF Sequences From Representative Species Of Saintpaulia And Streptocarpus Subgenera And Streptocarpella (Gesneriaceae), James F. Smith, M. E. Kresge, M. Möller, Q. C. B. Cronk

Biology Faculty Publications and Presentations

Two African genera of the Gesneriaceae, Saintpaulia and Streptocarpus, are similar in many respects. Both genera have blue to purple flowers, pollen of similar shape and exine sculpting, embryos with one-celled uninucleate chalazal haustoria, shared vegetative structures among some species, and are among the few genera in the Gesneriaceae which include species with chromosome count of n=15. Similarity of these features has indicated that the two genera are likely to be closely related. This study examines the sequences of the chloroplast gene ndhF among several representative Saintpaulia and representatives of Streptocarpus subgenera Streptocarpella and Streptocarpus. The …

Three spatial data sets consisting of high spatial resolution (1 m) remote sensing images acquired in 12 spectral bands, an on-the-go yield map, and a Digital Elevation Model were co-registered and evaluated for spatial variability studies in a Geographic Information Systems environment. Separate on-the-go yield maps were developed for 3, 5, and 12 statistically significant mean yield classes. For each yield class, the corresponding mean spectral and elevation data were extracted. The relationship between mean spectral and yield data was strongly linear (r = 0.99). Also, a strong linear relationship between mean yield and elevation data (r = 0.92) was …
