Tumor necrosis factor α (TNFα) is a pleiotropic cytokine that is predominantly produced by activated macrophages. The effects of TNFα are as diverse as the cells with which it interacts, e.g., stimulating fibroblast growth, exerting cytotoxic/cytostatic; activity against various human and murine cell lines, promoting inflammation through upregulation of endothelial adhesion molecules and IL-8 production. Yet TNFα is best known, and in fact was originally described, for its role in the bacterial-induced hemorrhagic necrosis of tumors and exacerbation of septic shock in which aberrant TNFα production leads to vascular collapse, cachexia, multiple organ failure, and ultimately death in as many …

A uniquely human interspersed repetitive DNA sequence family, the L2Hs, are highly polymorphic in human genomes. Several features of interspersed repeated DNA may contribute to the instability observed. Certain motifs (direct repeats, palindromes, and inverted repeats) comprising L2Hs elements may adopt unusual secondary structures such as cruciforms or hairpins. These motifs have been associated with features of genome instability in recombination, insertions and deletions. The L2Hs elements also are AT-rich (76%) compared to the bulk of human DNA (52%). That their dynamic nature (i.e. polymorphisms) may arise from recombination, insertions and deletions has led to the hypothesis that the L2Hs …

Molecular analysis of biological specimens usually requires extraction of high-molecular-weight DNA free of foreign DNA contaminants. DNA was extracted from black flies at different life stages that had been preserved by 4 methods: larvae and adults in ethanol, larvae in Carnoy’s solution, adults on card-points, and adults hand-swatted and sun-dried. Using specific primers for the mitochondrial ND4 gene, a 257-bp amplicon was obtained from specimens preserved by ethanol, card-point mounting, and sun-drying. Successful amplification often required DNA dilutions ≥ 1:20 (<1–10 ng). DNA from specimens preserved in Carnoy’s solution (ethanol: acetic acid, 3:1) yielded degraded DNA, resulting in fewer successful amplifications. Parasitic nematodes and, to a lesser extent, gut contents resulted in extra products when amplified with randomly amplified polymorphic DNA (RAPD) primers. Sufficient DNA was extracted from the head of a larva for a successful polymerase chain reaction (PCR), eliminating the need to remove the contaminating gut and parasites.

The effects of conjoint behavioral consultation (CBC) and a structured homework program on math homework completion and accuracy in junior high school students (grades seven through nine) who were at risk for academic failure were investigated. A multiple baseline design across five participants was utilized. Follow-up data were collected approximately one month after CBC was completed, to assess maintenance over time. Four of the five students improved their completion rates during treatment. Accuracy rates increased during treatment, but to a lesser degree. At follow-up, three of the five students maintained or improved gains they made during treatment. One student who …

The last systematic review of the yellow-shouldered bats of the Neotropical genus Sturnira in the Lesser Antilles was in 1976 (Jones and Phillips, 1976). At that point in time, two species--Sturnira lilium and Sturnira thomasi--were known from these islands. Sturnira lilium was represented by five subspecies, beginning with Trinidad and moving northward, these subspecies were lilium on Trinidad (Goodwin and Greenhall, 1961), paulsoni on St. Vincent (de Ia Torre and Schwartz, 19661, luciae on St. Lucia (Jones and Phillip, 19761, zygomaticus on Martinique (Jones and Phillips, 1976), and angeli on Dominica (de la Tom, 1966).

Sequence analysis identified significant variation in the second exon of equine infectious anemia virus (EIAV) rev. Functional analysis indicated that limited amino acid variation in Rev significantly altered the export activity of the protein but did not affect Rev-dependent alternative splicing. EIAV Rev can mediate export through two independent cis -acting Rev-responsive elements (RREs), and differences among Rev variants were more pronounced when both RREs were present. Variation in Rev may be an important mechanism for regulation of virus replication in vivo and may contribute to changes in clinical disease.

Cladistic analysis based on comparative morphology was used to examine the subfamily-level relationships within the cestode order Proteocephalidea. A single most parsimonious tree (70 steps, CI = 0.571; RC = 0.295; HI = 0.471) is consistent with monophyly for the Proteocephalidea and showed a relatively high consistency at the family level with the diagnosis of two major subclades. Unambiguous support for a Proteocephalidae subclade, including the Corallobothriinae, Proteocephalinae, Gangesiinae and Sandonellinae, and a Monticelliidae subclade, including the Marsypocephalinae, Zygobothriinae, Monticelliinae, Rudolphiellinae, Ephedrocephalinae and Othinoscolecinae was evident. This research sought to identify factors that might be useful in helping to predict family strengths in families caring for a child with a disability. Based on the ABCX model designed for families in crisis, this research examined severity of child's disability (A); the family's existing resources of household income, education of the mother, and mother's marital status (B); the family's perception of family resources, family support, and parenting stress (C); and how these influence family strength (X) over time. This study examined the effects of different modes of acculturation upon self reported levels of gender role conflict, and explored the relationship among acculturative stress, the concepts of Machismo and Familism and gender role conflicts. Hispanic males (N = 164) from a university with an 90% Hispanic enrollment were surveyed. Low Bicultural subjects differed significantly upon the factors of restrictive emotionality and restrictive affectionate behavior between men than other modes of acculturation. Machismo was found to be significantly related to stress and conflicts with gender roles, while Familism was found to be related to two of the patterns of gender …

Small hand-held microscopes have been developed for self-observation of salivary ferning patterns to detect the fertile time of a woman’s menstrual cycle. The purpose of this study was to evaluate one such microscope, the Lady Free Biotester®, by comparing it with the self-observation of cervical-vaginal mucus and with the self-detection of luteinizing hormone (LH) in the urine. Twelve natural family planning teachers (average age 34.6 years) observed their cervical-vaginal mucus, tested their urine for LH, and observed salivary and cervical-vaginal mucus ferning patterns (with the Lady Free Biotester) for two menstrual cycles. This approach to summarizing available information has also been taken for other taxa, such as Atelopus (Lotters, 1996). However, the task of compiling data for Xenopus is enormous relative to any other amphibian group, because Xenopus laevis has become a model system for molecular and development research (Cannatella and de Sa, 1993). Transmissible Spongiform Encephalopathies (TSE's) are neurodegenerative disorders characterized by a long generation time, spongy degeneration in the cerebral gray matter, neuronal loss and proliferation and hypertrophy of glial cells. An abnormal form of the prion protein (PrP) plays a major part in TSE pathogenesis and has been hypothesized to be the only component of the infectious agent. Some animals exposed to scrapie, the TSE affecting sheep and goats, seem to be resistant to development of the disease. Few reviews have been written related to the benefits of supplementing vitamin C and E in diabetic individuals. Excessive protein glycosylation is seen in persons with diabetes, and vitamin E and C have been shown to be beneficial in decreasing the glycosylation of proteins. In addition, vitamins E and Care potent antioxidants and actively reduce oxidation, triglycerides, and free fatty acid levels in diabetics. Diabetics may benefit from supplementing vitamins E and C.

Artificial neural network (ANN) is considered to be a universal function approximator, and genetic algorithm (GA) is considered to be a robust optimization technique. As such, ANN regression analysis and ANN-GA optimization techniques can be used to perform inverse groundwater modeling for parameter estimation. In this manuscript the applicability of these two techniques in solving an inverse problem related to a light-hydrocarbon-contaminated site is assessed. The purpose of this experiment was to determine if the estrogen receptor gene (ESR) could be used as a genetic marker for litter size in sheep. The estrogen receptor gene was chosen because it has been successfully used as a genetic marker for litter size in swine. In this study, DNA dilutions from two experimental flocks, totaling approximately 200 animals, were used. Amplification of exon 1/intron 1 of the ESR gene was performed using PCR. Asbestos is a general term for a group of fibrous silicates. Diseases such as asbestosis, bronchogenic carcinoma, and mesothelioma have been linked to asbestos exposure (1,2). The 1 two classes of asbestos, serpentine and amphibole, are distinguished by differences in structure and chemical composition (3). The amphibole crocidolite contains 27% iron while the serpentine chrysotile contains only 2-3% iron. The higher iron content of crocidolite is proposed to be a contributing factor for a much higher incidence of cancer with crocidolite exposure compared with chrysotile exposure ( 4). I visited Primary Children's Medical Center on a fresh snow morning near the beginning of last winter. Erythronium grandiflorum, a montane herb, was sampled to address the degree of genetic differentiation allocated to populations by isolation by distance vs. the amount of genetic differentiation produced by different flowering times, or isolation by phenology. Isozyme analysis was used to obtain genetic data from five populations of E. grandiflorum, three comprising the elevational scale which addressed isolation by phenology, and three comprising the spatial scale which explored isolation by distance. Based on the small sample size used in this study, the elevational scale shows a slightly higher degree of genetic differentiation than the spatial scale. Future studies …

Iron plays a critical role in biological systems. Its variable redox properties make it a tremendously versatile element. It is involved in numerous cellular processes including; oxygen transport, nucleotide synthesis, nitrogen fixation, electron transport and a host of metabolic reactions. Without it, nearly all life would cease to function. However, under certain circumstances iron can be extremely detrimental to biological systems and has been linked to numerous pathologies, including cancer, diabetes, Parkinson's disease, Alzheimer's and atherosclerosis.
