Usher syndrome type 1C (USH1C) occurs in a small population of Acadian descendants from southwestern Louisiana. Linkage and linkage disequilibrium analyses localize USH1C to chromosome 11p between markers D11S1397 and D11S1888, an interval of less than 680 kb. Here, we refine the USH1C linkage to a region less than 400 kb, between genetic markers D11S1397 and D11S1890. Using 17 genetic markers from this interval, we have isolated a contiguous set of 60 bacterial artificial chromosomes (BACs) that span the USH1C critical region. Exon trapping of BAC clones from this region resulted in the recovery of an exon of the nuclear …

Mammalian fertilization involves interactions of sperm surface receptors with the ligands of the zona pellucida, an extracellular matrix surrounding the ovulated oocytes. In humans, the zona pellucida is composed of three major glycoproteins. One of them, ZP3, participates in the primary sperm binding and in the subsequent triggering of the spermatozoa's acrosome reaction. Studies on the role of this specific protein in the human fertilization process are hampered by the limited amount of available biologically functional proteins.

The present study was designed to test the efficacy of using Electroencephalogram (EEG) and Event-Related Potentials (ERPs) for making task allocations decisions. Thirty-six participants were randomly assigned to an experimental, yoked, or control group condition. Under the experimental condition, a compensatory tracking task was switched between manual and automatic task modes based upon the participant's EEG. ERPs were also gathered to an auditory, oddball task. Participants in the yoked condition performed the same tasks under the exact sequence of task allocations that participants in the experimental group experienced. Clinical and theoretical evidence indicate that electric fields have biological effects ranging from recoverable disturbance to mortality induction depending on the field parameters and time of exposure. In this thesis, the effect of electrical field in pulsed form on aquatic nuisance species is investigated. Pulse parameters in terms of amplitude, width, shape, and repetition rate that stimulate the species to desirable levels are to be defined. Applying the electrical pulses with appropriate parameters to stun aquatic nuisance species entering marine cooling systems will prevent it from attaching to pipe walls. I have investigated the possibility of using an injection modelocked Ti-sapphire laser as a illumination source for GaAs photocathodes. A commercial Ti-sapphire laser was modified to accept a seed pulse from a gain-switched diode laser. Modelocked operation was obtained through gain modulation within the Ti-sapphire crystal as a result of injection seeding with a modelocked pulse train from a gain-switched diode laser. The Ti-sapphire laser essentially becomes a pulse amplifier for the diode laser. Both hypoglycemia (HG) and ethanol (ETH) intoxication can cause serious impairment in physiological and psychological functioning. Their combination could cause more severe cognitive impairment than either condition alone. This experiment investigated the effects of hypoglycemia, ethanol intoxication, and their combination on rat radial arm maze performance. Eleven male rats were trained to a stable, low error rate performance in achieving food rewards in an eight arm radial arm-maze. Rats were then given either insulin (2 units/kg) or saline in combination with ETH doses of 250, 500, 1000, or 1500 mg/kg 20 minutes prior to the maze run. Treatment order was …

Reverse transcriptase (RT) is an RNA-directed DNA polymerase isolated from human immunodeficiency virus (HIV) and other retroviruses. It has been the primary target for anti-HIV research since the discovery of its vital role in the retroviral life-cycle. The conventional method for carrying out kinetic characterization and inhibition studies of reverse transcriptase involves the use of radioactive materials. Although this method is effective, it is both time consuming, due to a necessary repeated washing step, and hazardous to the worker and environment. The N-terminal amino acid residues on polypeptides and proteins were determined in wastewaters prior to chlorination. The terminal amino groups'ere first derivatized with the well known derivatizing agent dansyl chloride, and then the resulting dansyl amino acid hydrolyzed from the peptide chain by a propionic acid/ hydrochloric acid mixture. The resulting dansyl amino acids were then separated and detected using reversed phase high performance liquid chromatography with fluorescence detection. The majority of N-terminal residues detected were the more polar amino acids. Concentrations ranged from 1 x 10^-10 to 3 x 10^-7 moles/liter. It is suggested that the shorter …

One enzyme which catalyzes the last step of the formation of the hormone retinoic acid from vitamin A (retinol) is retinal dehydrogenase type II (RalDH2). RalDH2, expressed in the Escherichia coil BL21(DE3) strain, was purified and crystallized using ammonium sulfate as a precipitant. These crystals belong to the space group P212121 (a = 108, b = 150, c = 168 Å, α = β = γ = 90°).

A replication banding procedure using fluorouracil (FU) and bromodeoxyuridine (BrdU) was developed for use with cultured leukocytes of channel catfish (Ictalurus punctatus). Analyzable banding patterns were produced on chromosomes stained with a fluorochrome plus Giemsa (FPG) method. The consistency of the chromosomal bands was evaluated by computer-assisted image analysis. Banding patterns of representative chromosomes including those bearing nucleolus organizer regions (NOR) were reproducible. A standard RBG-banded karyotype (R bands by BrdU and Giemsa) was established with idiograms of each chromosome. We also developed procedures to study sister- chromatid exchange (SCE) and sister-chromatid differentiation (SCD) in cultured leukocytes, without addition of …

Macroinvertebrate communities were examined on 4 winter dates over a 4-yr period in 2 high-altitude Rocky Mountain streams to document overwintering assemblages potentially experiencing spring acid pulses. Taxa richness values were comparable to other published lists for alpine and montane stream systems despite the fact that most literature reflected summer collections. Mean benthic density ranged from 1406 to 19,734 organisms/m², and drift rates ranged from 0 to 1740 organisms/100 m³. Benthic collections showed higher taxa richness than drift collections while the Ephemeroptera and Plecoptera occurred in greater proportions in drift than in benthos. The Nemouridae (Plecoptera), …

The tension reduction hypothesis postulates that consuming alcohol leads to a stress dampening effect. This stress reduction is negatively reinforcing, therefore, individuals learn that consuming alcohol will reduce unpleasant, tension type sensations and cognitions. Several studies have examined the validity of the tension reduction hypothesis as an explanatory model for the development o f alcohol problems, but no clear consensus has been found. As a result, researchers have begun to examine individual difference variables that may interact with the effects of alcohol on anxiety.
