Complexes Between Nascent Polypeptides And Their Molecular Chaperones In The Cytosol Of Mammalian Cells, Daryl K. Eggers, W. J. Welch, W. J. Hansen

Folding of newly synthesized proteins in vivo is believed to be facilitated by the cooperative interaction of a defined group of proteins known as molecular chaperones. We investigated the direct interaction of chaperones with nascent polypeptides in the cytosol of mammalian cells by multiple methods. A new approach using a polyclonal antibody to puromycin allowed us to tag and capture a population of truncated nascent polypeptides with no bias as to the identity of the bound chaperones. In addition, antibodies that recognize the cytosolic chaperones hsp70, CCT (TRiC), hsp40, p48 (Hip), and hsp90 were compared on the basis of their …

Rna Base-Amino Acid Interaction Strengths Derived From Structures And Sequences, Brooke Lustig, S Arora, R L. Jernigan

We investigate RNA base-amino acid interactions by counting their contacts in structures and their implicit contacts in various functional sequences where the structures can be assumed to be preserved. These frequencies are cast into equations to extract relative interaction energetics. Previously we used this approach in considering the major groove interactions of DNA, and here we apply it to the more diverse interactions observed in RNA. Structures considered are the three different tRNA synthetase complexes, the U1A spliceosomal protein with an RNA hairpin and the BIV TARTat complex. The mitochondrion imports and processes the vast majority of the proteins which comprise its structural elements and metabolic pathways. Studies in the last decade have been successful at defining the mechanism and protein machinery responsible for recognizing, importing, and processing cytosolic precursor proteins into the mature mitochondrial proteins. Little is known, however, about physiological factors which might influence or compromise mitochondrial protein processing. The objective of this work was to develop a novel approach to study mitochondrial import and assembly of precursor proteins as a process within the context of the cellular environment. Skeletal muscle lipogenesis in rodents may equal that of liver but its importance in the obese animals is not known. We evaluated the rate of fatty acid and triglyceride synthesis and storage content in red skeletal muscles, liver, inguinal fat, and retroperitoneal fat of the LA/N-fak (“corpulent”) and Zucker fa obese rat strains. Obese-lean pairs of both rat strains, weaned at 32 days, fed a nonlipogenic diet (54% starch for 21 days) were either dosed with 2 µCi 3H-H2 O per gram of body weight and tested after 60 minutes for whole body 3H-incorporation into fatty acids and …

Conidial Germination And Infection By Diplocarpon Rosae On Susceptible And Resistant Rose Species, Robert J. Wiggers, J. G. West, Josephine Taylor

Conidial germination and infection by Dipplocarpon rosae, the causal organism of rose blackspot, were examined on two resistant species of roses, Rosa roxburghii and R. wichuraiana and two susceptible hybrid tea roses (R. hybrid cv. Chicago Peace and Garden Party). Fungal conidia germinated and gave rise to subcuticular mycelium that formed haustoria within epidermal cells of all four roses tested. On the resistant rose species, epidermal cells associated with the infection site became necrotic, indicating that a hypersensitive response is involved in conferring their resistance D. rosae.

Sodium Nitrite Alone Protects The Brain O _I_ Microsomal Ca -Atpase Against Potassium Cyanide-Induced Neurotoxicity In Rats, Odutayo O. Odunuge, G.. A. Adenuga

The effect of a short-term oral administration of potassium cyanide (KCN) (200 ppm in diet) with or without sodium nitrite (NaNO2) pretreatment on rat brain microsomal Ca2* ATPase was investigated. The specific activity value of the enzyme significantly decreased (p<0.05) by 50% compared with control and by 63% for KCN-treated rats compared with KCN-treated rats pretreated with NaNO;,. There was no significant difference at the h = 0.05 level between the values obtained for the control and KCN-treated rats pretreated with NaNO,. These results show both that feeding lowers brain microsomal Ca2f-ATPase activity and that NaNO, has a protective role (antidote function) in that respect.

The design of Virtual Reality has only recently come into discussion, as well as the implications for this sort of design. I hope to determine how a user can work most efficiently and accurately in a Virtual World. By studying this, I hope to help in the standardization of Virtual Reality design.

About 5.2 million acres, or 25 percent, of northern Utah is forested. Fifty-two percent of this forest area is capable of producing commercial wood products and is classified as timberland. Forty-eight percent is classified as woodland, primarily pinyon-juniper. The predominant forest types on the timberland are aspen, Douglas-fir, lodgepole pine, and spruce-fir. The National Forest System manages 70 percent of the timberland; 23 percent is under private ownership, and 7 percent is under other public ownership (local, State, and other Federal). Thirteen percent of the timberland is withdrawn from commercial timber production and is in a reserved status. Most reserved …

Selective predation on the seeds of woody plants. J. Torrey Bot. Soc. 124: 67-70). 1997.-Seed predation may be an important factor influencing the structure of successional plant communities. We used a cafeteria-style experiment, placed in an old field and an early successional forest, to determine predator preferences for seeds of nine species of woody plants. Intensity of seed predation was equivalent in both sites. Seed predators preferred Acer saccharum, flex vertic illata, and Viburnum dentatum, but this was not related to seed mass. Predation intensity was more variable in the old field than in the forest, possibly related to the …

Validation of Animal Experimentation Collection
Since the early 1970s, scientists have used preference tests (tests that require animals to choose between two or more different options or environments) as a means of answering questions about animal welfare. Preference tests have been used to establish animals' preferences for common housing options such as ambient temperature, illumination and preferred types of bedding and flooring; to improve the effectiveness of devices such as loading ramps and nest boxes; and to clarify how strongly animals avoid various aspects of confinement and methods of restraint.

Previous research showed that pigs reared in substrate-impoverished conditions performed a smaller proportion of their total behavioural repertoire in their home pens (showed lower behavioural diversity), than pigs reared in substrate-enriched conditions. This study examined whether these differences were the result of fundamental changes in behavioural organisation. A T-maze task was used to test the hypothesis that substrate-impoverished pigs are prone to develop fixed, unvarying behaviour which may underlie their reduced behavioural diversity. They were predicted to be poorer at reversing previous response patterns in the maze, and less able to alter their behaviour in response to a novel (distracting) …

With respect to the ethical debate about the treatment of animals in biomedical and behavioral research, Harry F. Harlow represents a paradox. On the one hand, his work on monkey cognition and social development fostered a view of the animals as having rich subjective lives filled with intention and emotion. On the other, he has been criticized for the conduct of research that seemed to ignore the ethical implications of his own discoveries. The basis of this contradiction is discussed and propositions for current research practice are presented.

From September 1995 to April 1996 we studied interactions among dogs, people, and the environment in Boulder, Colorado. Data on behavioral disturbances by off-leash dogs who were accompanied by a person were collected with respect to dog-dog and dog-human interactions, dog-wildlife encounters, dogs trampling vegetation, and dogs entering and disturbing bodies of water. A questionnaire also was administered. Behavioral data showed that off-leash dogs generally did not travel far off trail, that when they did it was for short periods of time, and that they rarely were observed to chase other dogs, disturb people, chase wildlife, destroy vegetation, or enter …

Scientific research on 'animal welfare' began because of ethical concerns over the quality of life of animals, and the public looks to animal welfare research for guidance regarding these concerns. The conception of animal welfare used by scientists must relate closely to these ethical concerns if the orientation of the research and the interpretation of the findings is to address them successfully.
At least three overlapping ethical concerns are commonly expressed regarding the quality of life of animals: (1) that animals should lead natural lives through the development and use of their natural adaptations and capabilities, (2) that animals should …
